In a qualitative test a sample is first reacted with a mixture of acetic acid and hydrogen iodide in a test tube. The ensuing reaction results in the cleavage of the ether or the ester into an alkyl iodide and respectively an alcohol or an carboxylic acid. By heating this mixture, the gases are allowed to come into contact with a piece of paper higher up the test tube saturated with mercury(II) nitrate. Any alkyl iodide present will give a reaction with the mercury compound to mercury(II) iodide which has a red or yellow color. A spectrophotometric technique is described for the continuous recording of sulfobromophthalein uptake by isolated hepatocytes. The technique is based on the principle that sulfobromophthalein behaves as a pH-indicator and may be followed photometrically when moving from the medium at pH 7.8 into the interior of the cell. Data show that upon addition of cells to a sulfobromophthalein solution, an absorbance change can be recorded. The kinetics of the process is biphasic and the initial rate is linearly related to the amount of cells added. By this technique it was confirmed that the substrate dependence of the initial velocity of transport is a compound function including a saturable portion with an apparent Km in the mu molar region. Our study demonstrates that the antihyperlipidemic clofibrate derivatives, which are structurally different from the known activator, sulfobromophthalein, exert stimulatory effects specifically on AKR 1C4 of human liver 3αHSD isoforms. For the activation by sulfobromophthalein, there has been no direct information about the structurally specific interaction between the molecule and the enzyme, except for its sulfonyl group(s) (Matsuura et al., 1996,1997).
