Silymarin flavonolignans are well-known agencies that possess antioxidative typically, anti-inflammatory, and hepatoprotective features. predictive, additional in vitro and in vivo research would be essential to confirm the antiviral impact. 2.2. Influenza A Pathogen Influenza A pathogen (IAV) is an extremely contagious pathogen and a respected reason behind mortality and morbidity internationally. Influenza pandemics and epidemics pose a significant threat to both individual and pet populations. Although effective vaccines can be found against IAV, these vaccines should be frequently updated because of the ability from the pathogen to induce regular antigenic drift and periodic antigenic shifts to its envelope glycoproteins. Furthermore, just few IAV antiviral therapeutics have already been clinically accepted and currently used including neuraminidase inhibitors [19] as well as the newer inhibitor of cap-dependent endonuclease [20]. As a result, continuous id of novel healing strategies to ...
Flavonolignans are natural phenols composed of a part flavonoid and a part lignan. Flavonolignans identified in Silybum marianum (milk thistle) silymarin complex include silibinin, silychristin, silydianin, dehydrosilybin, deoxysilycistin, deoxysilydianin, silandrin, silybinome, silyhermin and neosilyhermin and can be produced in vitro. Silibinin is found in the roots of S. marianum while silyamandin can be found in the fruit. Hydnocarpin can be found naturally in Onopordon corymbosum and can be synthetised. Scutellaprostin A, B, C, D, E and F can be isolated from Scutellaria prostrata and can also be synthesized. Hydnowightin can be isolated from Hydnocarpus wightiana seeds. Three flavonolignans derived from the flavone tricin have been isolated from the herb Avena sativa. Palstatin has been isolated from the Amazon tree Hymeneae palustris. Salcolin A and salcolin B can be found in Salsola collina. Rhodiolin, the product of the oxidative coupling of coniferyl alcohol with the 7,8-dihydroxy ...
Silymarin, an extract of crushed achenes of the milk thistle plant Silybum marianum is a multi-constituent mixture, 70-80% of which consists of a complex assortment containing the flavonolignans silybin A and B, isosilybin A and B, silydianin, and silychristin, and the flavonoid taxifolin. To date, numerous pharmacological actions of the silymarin extract have been documented in the biomedical literature, including hepatoprotective, anti-inflammatory, anti-tumor, and anti-fibrotic activities. The present study describes a novel liquid chromatographic-tandem mass spectrometric method for simultaneous analysis of silychristin, silydianin, silybin A and silybin B, isosilybin A and isosilybin B, and taxifolin in human plasma employing liquid-liquid extraction ...
There is increasing evidence that nutraceutical agents are important for cancer prevention and intervention (1, 11, 26) because dietary and nondietary agents and lifestyle determine cancer incidence and may also affect the growth, progression, or aggressiveness of cancer cells (26). Silybin is a flavonolignan and the major active constituent of silymarin, which is a complex mixture of flavonolignans and polyphenols that are extracted from milk thistle seeds. The German Commission E has recommended its use for dyspeptic complaints and liver conditions including toxin-induced liver damage and hepatic cirrhosis. It has also been recommended as a supportive therapy for chronic inflammatory liver conditions (27). Neither milk thistle extract nor silybin is currently approved for any medical use in the United States; however, it is sold as a dietary supplement and is one of the most frequently sold herbal products in the United States (27). The anticancer efficacy of silybin is clearly evident in the ...
Silibinin (INN), also known as silybin (both from Silybum, the generic name of the plant from which it is extracted), is the major active constituent of silymarin, a standardized extract of the milk thistle seeds, containing a mixture of flavonolignans consisting of silibinin, isosilibinin, silicristin, silidianin, and others. Silibinin itself is a mixture of two diastereomers, silybin A and silybin B, in approximately equimolar ratio. The mixture exhibits a number of pharmacological effects, particularly in the liver, and there is some clinical evidence for the use of silibinin as a supportive element in alcoholic and child grade A liver cirrhosis. Poor water solubility and bioavailability of silymarin led to the development of enhanced formulations. Silipide (trade name Siliphos), a complex of silymarin and phosphatidylcholine (lecithin), is about 10 times more bioavailable than silymarin. An earlier study had concluded Siliphos to have 4.6 fold higher bioavailability.[unreliable medical ...

Non-stop carbocationic polycyclizations of isoprenoids have been called the most complex chemical reactions occurring in nature. We describe a strategy for the initiation of tail-to-head polycyclization that relies on the sequestration of the counteranion away from the carbocation, which allows full propagation of the cationic charge. If the anion is mobile, Coulombic forces hold this species in close proximity to the carbocation and cause preemptive termination through elimination. Anion sequestration is crucial for effecting the biomimetic synthesis of complex and unstable terpenes, including the highly strained funebrenes. This study illustrates the deleterious role of the counterion in tail-to-head carbocationic polycyclization reactions, which to the best of our knowledge has not been rigorously explored. These observations are also expected to find use in the design and control of cationic polycyclization along biosynthetic pathways that have previously been inaccessible in bulk solvent ...

Milk thistle (Silybum Marianum L.) is a natural source of complex flavonolignans called silymarin. As an antioxidant silymarin has highly antyhepatotoxic properties. Not only does it enhance formation and regeneration of the liver cells, but also effectively protects all the newly formed cells. Milk thistle promotes bile secretion, as well as is antitoxic and anti-inflammatory. Its administration is particularly recommended during any liver-straining pharmacotherapy, as well as upon its completion, throughout the convalescence period.. Did you know? Milk thistle may also be used as a dietary supplement in the treatment of diabetes and cholelithiasis (gallstones). It is indicated in digestive disorders, upon inadequate gastric pH, lack of appetite, flatulence, hypotension, and motion sickness. It also significantly accelerates the skin regeneration.. ...
Women at high risk of developing breast cancer are prescribed selective estrogen response modulators, including raloxifene, as chemoprevention. Patients often seek complementary and alternative treatment modalities, including herbal products, to supplement prescribed medications. Milk thistle preparations, including silibinin and silymarin, are top-selling herbal products that may be consumed by women taking raloxifene, which undergoes extensive first-pass glucuronidation in the intestine. Key constituents in milk thistle, flavonolignans, were shown previously to be potent inhibitors of intestinal UDP-glucuronosyl transferases (UGTs), with IC50s ≤ 10 μM. Taken together, milk thistle preparations may perpetrate unwanted interactions with raloxifene. The objective of this work was to evaluate the inhibitory effects of individual milk thistle constituents on the intestinal glucuronidation of raloxifene using human intestinal microsomes (HIMs) and human embryonic kidney cell lysates ...
A compound from milk thistle kept liver cancer cells from spreading and reduced their numbers in a new laboratory study. Researchers from the University of California at Irvine tested various doses of silibinin, the major constituent of silymarin (the mixture of flavonolignans extracted from blessed milk thistle), in its pure form. Doctors found that a dose of 180 micromoles per liter (mm/L) of silibinin kept liver cancer cells from spreading. At a dose of 240 mm/L, silibinin accelerated the natural death rate of cancer cells by a factor of nine times.. In a major finding, scientists noted that silibinin appeared to allow cells enough time to detect and repair DNA damage before passing the bad trait onto new cells. Silibinin also decreased the abnormal blood vessels that feed tumors. Study author, Dr. Ke-Qi Hu, noted that silibinin has novel anti-cancer mechanisms and provides researchers with new options for preventing cancer. Milk thistle is a herb from which its fruits are used. It contains flavonolignans, silymarin and especially silibinin isomers, silychristin and silydianin, flavonoids, steroids and estrogen-like compounds. Attributed properties: Liver Protector. Indications based on the Commission E of the German Ministry of Health: cholelithiasis, liver disorders. Devoid of toxicity. It is excreted into breast milk in little or no amount. There is no evidence of its effectiveness as a galactogogue. Two studies have been performed (one with cows and the other with women) trying to show a galactogogue effect, but both are limited by a very flawed methodology. Best galactogogue is a frequent and on-demand breastfeeding together witn an appropriate technique. Precaution is recommended before taking herbal infusions: 1. Ensure a reliable source: poisoning occurred by confusion with another plant that resulted to be toxic, poisoning from heavy metals and food poisoning by contamination with bacteria or fungi. 2
Silymarin and green tea are the two most widely used botanical products used by patients with chronic hepatitis C virus (HCV) infection. A major limitation of prior clinical investigations that may account for lack of efficacy is their use of inadequate customary oral dose regimens. Ongoing Phase II studies utilizing higher than customary doses of silymarin are based on potential disease modifying effects resulting from the potent antioxidant properties of silymarin flavonolignans. Since patients often use more than one herbal product to self-treat their disease, the use of higher than customary doses of silymarin may change silymarins potential for herbal-herbal interactions. Pharmacokinetic interactions between silymarin and green tea catechins may change their safety/tolerability profiles, and may increase their antioxidant effects through additive or synergistic pharmacodynamic interactions. The objective of this double blind, active placebo controlled, randomized clinical trial is to ...

The present study evaluated the dry matter mass production in different Passiflora alata Dryander organs and phenological stages, as well as their correlation with phytochemical quality in relation to the flavonoids vitexin, isoquercetin, quercetin and rutin. Two experiments were carried out: Experiment 1, effect of different organs (flower, leaves, fruits and branches), and Experiment 2, effect of phenological stages (pre-flowering, flowering, fruiting and post-fruiting). Both experimental designs were in randomized blocks with five and six replicates, respectively. The highest flavonoid concentration and production were detected in leaves. On the other hand, branches and fruits did not have significant rutin and isoquercetin content. The largest leaf dry matter mass and flavonoid production was observed in the post-fruiting stage, that is, at 300 days after transplanting (DAT). Flower formation was a strong deleterious competitor to leaf and branch dry matter mass production, as well as to ...

I believe very strongly that Rod Rickards greatest lesson was to encourage me to always be inquisitive and to look at an unexpected outcome in chemistry not as a failure but as an opportunity to understand the science more deeply and even help address new problems. I have been guided by this lesson throughout my academic career. ANU Alumni Joanne Jamie completed her PhD at the RSC in 1992. Focusing on organic chemistry, Joanne says her studies encouraged her to think about the potential positive outcomes that could be found in chemical research.. I did my PhD with Rod Rickards, Joanne explains. It was on biomimetic synthesis of important anticancer and antimicrobial molecules. So it was fundamentally organic synthesis, but its approach provided an understanding of biological processes and applications.. My PhD was really my starting point of wanting to always have a research goal that had some medicinal or other value in its output. Thats actually something Ive always wanted in my ...

Many aldehydes are important components of natural flavours. They are used in food, cosmetic, and biomedical industries in large amounts. Plant cells or microorganisms carry out their production by biotransformation, which is one of the biotechnological methods that allow them to be defined as natural. Cell cultures of Silybum marianum and Peganum harmala have been studied with a view to investigat their abilities to produce flavonolignans and b-carboline alkaloids respectively. However, we have isolated S. marianum and P. harmala culture strain, which are able to metabolise several aromatic aldehydes. Ten culture strains derived from S. marianum and P. harmala were examined for their ability to biotransform exogenous aromatic aldehyde compounds, including benzaldehyde, 2-methoxybenzaldehyde, 4-methoxybenzaldehyde, cinnamaldehyde and 3-methoxy, 4-hydroxy benzaldehyde. Callus cultures of Silybum marianum and Peganum harmala were established from seedlings, and healthy suspensions were grown using
Elicitation with middle-viscous chitosan (30 mg/50 mL) significantly stimulated silymarin synthesis in Silybum marianum hairy root cultures. The root cultures established by infection with Agrobacterium rhizogenes AR15834 showed a potential for production of silymarin. Elicitation with medium molecular weight of chitosan (0, 5, 10, 20, and 30 mg/50 mL) was used in order to improve silymarin production. Total silymarin increased about 5.26-fold after 96 h of treatment with 30 mg/50 mL chitosan. Dry weight of the hairy roots reached the highest point (0.530 and 0.535 g) after 96 h in presence of 20 and 30 mg/50 mL chitosan, respectively. Five different flavonolignans were isolated; taxifolin, silychristin, silydianin, silybin and isosilybin) 0.133, 0.200, 0.120, 0.041 and 0.056 mg/g dry weight, respectively). 30 days old hairy roots were treated by 30 mg/50 mL chitosan in different times (12, 24, 48, 72, 96 and 120 h). The amount of silymarin accumulation significantly increased (0.705 mg/gDW) in hairy

A study published recently in Phytotherapy Research (DOI: 10.1002/ptr.1988) demonstrated that the extract from the seeds of Milk Thistle, Silybum marianum (silymarin) may help in reducing blood sugar among diabetes patients.. The data came from a randomized double-blind clinical trial involving 51 people who had had type II diabetes for at least 2 years. One group of 25 patients received 200 mg of silymarin three times a day for 4 months, while the remaining 26 received a placebo treatment. All of the patients continued to use conventional oral hypoglycaemic treatment during the trial.. At the end of the trial, the treatment group showed a significantly reduced fasting blood glucose levels and a reduction in glycosylated haemoglobin.. Silymarin extracts mechanism of action, however, remains unknown. It may be attributed to the antioxidants called flavolignans. To verify these data, large multi-center studies will have to be conducted, according to the researchers.. ...
An extract (silymarin) from the plant Silybum marianum (milk thistle) was shown to increase circulating concentrations of prolactin in cycling rats. A dose-response study was undertaken to determine if silymarin does have hyperprolactinemic properties in cycling swine. Forty-four weaned sows were allotted to four groups receiving 0, 1, 2 or 4 g d1 of silymarin over a period of 8 d. Blood samples were obtained on days 1 (first day of treatment starting 24 to 48 h after the onset of the standing estrus), 2, 3, and 9. Prolactin, progesterone, estradiol-17p and leptin concentrations were determined. Silymarin did not increase prolactin concentrations at any of the doses studied, nor did it affect concentrations of the other hormones (P , 0.1).. ...
Possibly native near the coast of southeast England, it has been widely introduced outside its natural range, for example into North America, Iran, Australia and New Zealand where it is considered an invasive weed. Cultivated fields for the production of raw material for the pharmaceutical industry exist on a larger scale in Austria (Waldviertel region), Germany, Hungary, Poland, China and Argentina. In Europe it is sown yearly in March-April. The harvest in two steps (cutting and threshing) takes place in August, about 2-3 weeks after the flowering.[citation needed]. Milk thistle is a member of the sunflower family native to a narrow area of the Mediterranean. It has since been naturalized throughout Europe and can commonly be found in Oregon and California where it is considered a common garden weed.
