Puffer fish have evolved two remarkable abilities. First and most visually striking is the ability of the puffer fish to inflate itself to over three times its natural size. The second outstanding ability of the puffer fish is to produce a deadly poison, tetrodotoxin.
The zebrafish genomic sequence database was analysed for the presence of genes encoding members of the Toll-like receptors (TLR) and interleukin receptors (IL-R) and associated adaptor proteins containing a TIR domain. The resulting predictions show the presence of one or more counterparts for the human TLR1, TLR2, TLR3, TLR4, TLR5, TLR7, TLR8, TLR9, IL-1R and IL-18R genes and one copy of the adaptor genes MyD88, MAL, TRIF and SARM. In contrast to data for the pufferfish Fugu rubripes, zebrafish has two genes that are highly similar to human TLR4. In addition, one fish-specific TLR group can be distinguished that is closely related to the Drosophila melanogaster Toll-9 gene. Let us look at the puffer fish. The puffer fish is known scientifically as the Tetraodontidae. Interestingly enough, it is considered to be the second-most poisonous vertebrae in the world. Also, the puffer fish are covered with spines, which are only visible when the fish is puffed up. While the puffer fish is highly maneuverable, it is very slow. This makes it seeming easy prey. Their excellent eyesight and great maneuverability generally keep them safe. Their back up defense mechanism, used of they are successfully pursued, is to fill their extremely elastic stomachs with water (or air when outside the water) until they are much larger and almost spherical in shape. Even if they are not visible when the puffer is not inflated, all puffers have pointed spines, so a hungry predator may suddenly find itself facing an unpalatable pointy ball rather than a slow, tasty fish. In Wade Davis excellent article The Ethnobiology of the Haitian Zombi (abstract), he lists the composition of zombie poisons from five separate locations around Haiti. Although there were many differences, based on what was available at the different geographical locations, there were several key ingredients. One of the most important was the puffer fish (Diodon hystrix, Diodon holacanthus, and others). Puffer fish in the region have very high levels of tetrodotoxin (TTX), which is, of course, the same toxin found in Japanese fugu fish. Tetrodotoxins are extremely toxic molecules and act by blocking Na ion channels, presumably through the positively charged guanidido group, which competes for the Na binding site in the channel. Without Na exchange, nerve impulses do not propagate which leads to paralysis, as well as cardiac and respiratory failure. The LD50 of TTX is estimated to be about 5 ug/kg in humans (less than1 mg of TTX), so what if a person ingested something a little less than that? ...

Some of you may recall I had to put my 18 year old cat, Savvy, to sleep the month after the boys were born. Since then, my other 18 year old cat, Bunny, has rapidly declined. The last two weeks have been the most notable and I dont want to wait as long as we did with Savvy. After she was gone, we realized that wed waited too long and just how bad her quality of life had been for quite some time. This cat is very special to me. He was my favorite (I know, you are not supposed to have one…) and I know he is suffering. I was truly hoping that both of them would pass before the babies were born so they would never be pushed out of the spotlight. But that didnt happen. It makes me so sad to know that Bunny has spent the last months of his life not getting the attention he was used to for 18 years.
