Genetically identical cells often show significant variation in gene expression profile and behaviour even in the same physiological condition. Notably, embryonic cells destined to the same tissue maintain a uniform transcriptional regulatory state and form a homogeneous cell group. One mechanism to keep the homogeneity within embryonic tissues is the so-called community effect in animal development. The community effect is an interaction among a group of many nearby precursor cells, and is necessary for them to maintain tissue-specific gene expression and differentiate in a coordinated manner. During Xenopus myogenesis, the muscle precursor cells must be in group contact throughout gastrulation in order to develop into terminally differentiated muscle. The molecular basis of this community interaction has not to date been elucidated. We have developed an assay for testing potential community factors, in which isolated muscle precursor cells are treated with a candidate protein and cultured in dispersion. We have tested a number of candidate factors and we find that only eFGF protein is able to mediate a community effect, stimulating stable muscle-specific gene expression in demonstrably single muscle precursor cells. Owing to their single genome, haploid cells are powerful to uncover unknown genes by performing genetic screening in mammals. However, no haploid cell line from an extraembryonic lineage has been achieved yet, which limits the application of haploid cells in placental genetic screening. Here, we show that overexpression of Cdx2 can convert haploid embryonic stem cells to trophoblast stem cells (TSCs). p53 deletion reduces diploidization during the conversion and guarantees the generation of haploid-induced TSCs (haiTSCs). haiTSCs not only share the same molecular characterization with trophoderm-derived TSCs but also possess multipotency to placental lineages in various procedures. Our results indicate that TSC progenitors are distributed widely in the ExE, and then the ChE, prior to fusion with the EPC, on the basis of the non-local distribution of TSC colonies and lines in vitro (Table 1 and Table 2), and mitoses in vivo (Fig. 6A and Table 5). These findings are supported by the pattern of ExE cell division that revealed that labelled sister cells were frequently not immediately adjacent to each other (Fig. 2 and Table 3), indicating that considerable cell mixing and dispersal occurs during growth of the ExE/ChE.. When the placenta develops abnormally or malfunctions, babies are born small and/or premature. In mouse, X-chromosome inactivation (XCI) can either be imprinted or random. The mammalian placenta consists of different trophoblast cell types that assist in the variety of functions required for the maintenance of pregnancy. In rodents, labyrinthine trophoblasts of the placenta are especially important, because they are capable of differentiating into fused labyrinthine cells, which form the feto-maternal exchange surface. Even though the molecular signals triggering labyrinthine trophoblast differentiation are poorly understood, transforming growth factor-β (TGF-β) has been shown to be present in the placental environment and alter trophoblast development. In this study, we investigated the effects of TGF-β on the differentiation of the labyrinthine trophoblast stem cell lines SM10 and HRP-1. The trophoblast cell lineage is an interesting model system because it is composed of a limited number of cell types that are spatially patterned. Trophoblast stem (TS) cells reside within a layer called the chorion and either remain as stem cells or differentiate into spongiotrophoblast (SpT), trophoblast giant (TG) cells or syncytiotrophoblast cells (SynT) of the labyrinth. Eif2s1(tm1RjK) mice, in which Ser51 of eukaryotic initiation factor 2 subunit alpha (eIF2α) is mutated, display a 30w increase in basal translation. In Eif2s1(tm1RjK) placentas, we observed increased ER stress and anomalous accumulation of glycoproteins in the endocrine junctional zone (Jz), but not in labyrinthine zone where physiological exchange occurs. Placental and fetal weights were reduced by 15% (97mg to 82mg) and 20% (1009mg to 798mg) respectively. Mouse embryonic fibroblasts (MEFs) were derived from Eif2s1(tm1RjK) mutants. These MEFs exhibited ER stress, grew 50% slower and showed reduced Akt-mTOR signalling compared to wild-type cells.
