Abstract: The mode of inheritance of resistance to cowpea aphid-borne mosaic virus (CAMV) and southern bean mosaic virus (SBMV) was determined in the following cowpea (Vigna unguiculata s.sp. unguiculata (L) Walp) varieties: A44/2, TVu 222, TVu 612 all resistant to CAMV and TVu 1948, which is resistant to both CAMV and SBMV. Crosses were made between each of the resistant varieties and C20-55, a cowpea variety susceptible to CAMV and SBMV. The parents, FI, F2, B1 and B2 progenies of each cross were raised in the greenhouse and inoculated with the appropriate virus. The results showed that two recessive genes control resistance to CAMV in A44/2, a single dominant gene controls resistance to the same virus in TVu 222, while a single recessive gene was responsible for resistance in each of the varieties TVu 612 and TVu 1948. The role of plant defensins against weevils is based on inhibitor of alpha-amylase activity in weevil gut, therefore, weevils can not digest starch and then die. The low content of Vigna radiata plant defensin 1 (VrPDF1) in mung bean seeds is promted researches to increase the VrPDF1content, in which leads to enhance alpha-amylase inhibition and improve bruchid resistance in mung bean. This article presents the results of overexpression of VrPDF1 gene in transgenic mung bean plants. VrPDF1 gene was successfully transferred into DX22 mung bean cultivar and expressed in T1 generation transgenic mung bean seeds. The extract containing recombinant VrPDF1 protein inhibited alpha-amylase of weevil larvae in its larval stage. Leaf tissue was collected from both disease free and artificially disease challenged plants at 7th, 15th and 30th DAI and syntesized cDNA. The expression level of selected 10 RGA genes of french bean was measured in both disease free and artificially inoculated leaf tissues separately at 7th, 15th and 30th DAI. 10 RGA genes were tested under MYMV infested condition in french bean. All 10 selected RGA genes were expressed in leaf tissues at different days after inoculation with MYMV. COHFBRGA-2, 6, 8 and 10 genes of french bean were up-regulated in leaf of susceptible genotypes at disease inoculation condition at all the intervals. COHFBRGA-3, 4, 5 and 7 was down- regulated among all the intervals of disease development compared to control. Whereas, COHFBRGA-9 expressed only at 30 DAI.

Phytosterols have been widely studied for their cholesterol-lowering effect. Conjugated phytosterol forms have been found more active than free moieties. There are no reports about the sterol profile of black bean seed coats neither its effects on cholesterol metabolism. The aim of this research was to identify and quantify phytosterols from black bean seed coats and to determine their effects on cholesterol micellar solubility and on mRNA and key protein levels involved in lipid/cholesterol metabolism and cholesterol transport in primary rat hepatocytes. Free phytosterols, acylated steryl glycosides, and steryl glycosides were extracted from black bean seed coats. They were identified through HPLC-MS-TOF and quantified through HPLC equipped with UV-visible and evaporative light-scattering detectors. Free and conjugated phytosterols from the coats significantly increased the inhibitory effect of cholesterol micelle formation compared with stigmasterol, which was used as control (P < 0.05). Colletotrichum lindemuthianum is one of the main pathogens affecting common bean (Phaseolus vulgaris) in tropical regions. In the present work we describe three mutants of C. lindemuthianum obtained by insertional mutagenesis using the REMI technique (Restriction Enzyme-Mediated Integration). The mutants were selected based on their lower virulence/pathogenicity and the partial characterization of the mutated genes is presented. In addition, the mitochondrial genome of this phytopathogen was partially characterized. The results show that the gene pacC1 plays a role in vegetative growth and pathogenicity. The use of the restriction enzyme NarI in the transformation of C. lindemuthianum increased approximately 10-fold the transformation efficiency using plasmid pAN7.1. The presence of native Rhizobia which are efficient in symbiotic nitrogen fixation in legumes introduced in the fields of Rio Grande do Sul, such as the forages plants of the genus Lotus, is one of the factors determining the success or the failure of the adaptation of these new plant species. This work aimed to evaluate the symbiotic compatibility of Rhizobia in association with Desmodium incanum, a native plant from the fields of Rio Grande do Sul, as well as with L. corniculatus, L. uliginosus, L. glaber and L. subbiflorus species. For this, 35 isolates and 17 lineages of Rhizobia were analyzed. The isolates were characterized phenotypically based on the colony morphology and on the indole acetic acid and siderophore production. The Rhizobia were studied for symbiotic compatibility by cross-inoculation with the plant species studied. The use and management of angico (Anadenanthera colubrina (Vell.) Brenan) by a rural community in northeastern Brazil was examined. By employing different techniques of data collection and population structure analysis, it was determined that this species had multiple uses within the local community (especially as timber and for other wood products), and that local management of this species is based on simple maintenance and harvesting of individuals in agroforest homegardens. The study of the population structure of this tree species indicated that management and conservation strategies must include the participation of the local community.

Assessment of genetic diversity is an essential component in germplasm characterization and utilization. Molecular markers serve as a valuable tool to assess the genetic variation and germplasm identification, which play a key role for faba (Vicia faba L.) bean breeding. In this study, we analyzed the genetic diversity of faba bean accessions based on simple sequence repeats (SSRs) and morphological traits. Forty-six faba bean accessions, originating from different countries and from the ICARDA breeding program, were evaluated by using 15 morphological and agronomic traits and 17 simple sequence repeat (SSR) loci. Significant differences among accessions for the 15 morphological descriptors were observed. Abstract: As a follow-up to recent outbreaks of food poisoning (nausea, vomiting, diarrhea) associated with raw or undercooked red kidney beans, a study was conducted on the lethality and heat destruction of the implicated lectin causative agent present in the beans. Agglutinating activity towards trypsinized rabbit erythrocytes was used as an in vitro assay for lectin content. Only the 3 varieties of P. vulgaris (red and white kidney beans, rose coco bean) contained large amounts of lectins. Soaking overnight removed 20-70% of the lectin. Red and white kidney beans caused 100% mortality to various sets of rats when fed at 10% or more of the diet for up to 14 days. Complete destruction of the hemagglutinin content of the beans (due to lectin) was generally attained by heating for up to 20 minutes at 100 degrees Centigrade. A study was designed to examine the relationships between protein, condensed tannin and cell wall carbohydrate content and composition and the nutritional quality of seven tropical legumes (Desmodium ovalifolium, Flemingia macrophylla, Leucaena leucocephala, L pallida, L macrophylla, Calliandra calothyrsus and Clitoria fairchildiana). Among the legume species studied, D ovalifolium showed the lowest concentration of nitrogen, while L leucocephala showed the highest. A 1.9 kb DNA region of Rhizobium leguminosarum biovar viciae strain VF39 capable of promoting microaerobic and symbiotic induction of the Rhizobium meliloti fixN gene was identified by heterologous complementation. Sequence analysis of this DNA region revealed the presence of two complete open reading frames, orf240 and orf114. The deduced amino acid sequence of orf240 showed significant homology to Escherichia coli Fnr and R. meliloti FixK. The properties of oil/water emulsions stabilized with 1% w/v common bean (Phaseolus vulgaris L.) or scarlet runner bean (P. coccineus L.) proteins, extracted by isoelectric precipitation or ultrafiltration, at pH 7.0 and 5.5 were studied. The stability of emulsions, evaluated on the basis of droplet size, creaming, viscosity and protein adsorption measurements, is increased by the addition of xanthan (0.1 and 0.25% w/v). This is probably due to the increase in the continuous phase viscosity and the creation of a network, which prevents the oil droplets from coalescing. Also, the ability and stability of 1 or 2% w/v foams was studied. Xanthan (0.25% w/v) does not enhance foam formation, but promotes foam stability, possibly owing to the increased viscosity of the aqueous phase, making it more difficult for air to enter the system and create a satisfactory foam volume. Cyclophosphamide is an anti-neoplastic chemotherapy drug which, when administered to animals during the gestational period, provokes visceral, skeletal and external malformations. Copaiba oil obtained from Copaifera L. genus is traditionally used in popular medicine for its anti-inflammatory and antimicrobial activities. However, the effect of copaiba oil onteratogenesis remains unknown. This study aimed to investigate the possible protector effects of copaiba oil on the model of teratogenesis induced by cyclophosphamide in mice. Pregnant female Swiss mice were divided into 8 groups (n = 15). Three groups received copaiba oil, via gavage, in the following doses: 0.3 mL·Kg-1, 0.6 mL·Kg-1 and 0.9 mL·Kg-1 (b.w.), associated to phosphate-buffered saline (PBS), intraperitoneal (i.p.). The present research was aimed at determining the ploidy level of 20 accessions of winged bean (Psophoscarpus tetragonolobus) using known diploid standard as control and also subjecting two accessions to colchicine mutagenesis. For flow cytometric analysis, approximately 50mm2 of young fresh intact leaves of 8 weeks post-seedling emergence from the 20 accessions were used. Crude isolation of nuclei was done by chopping the leaf in 1ml of ice cold buffer from DNA kit (cysteine UV Partec GmbH Germany) using a razor blade.
