Isoprostanes are stable products of arachidonic acid peroxidation and are regarded as the most reliable markers of oxidative stress in vivo. Here we describe the LC-MS/MS procedure enabling simultaneous determination of four regioisomers (8-iso prostaglandin F2α, 8-iso-15(R)-prostaglandin F2α, 11β-prostaglandin F2α, 15(R)-prostaglandin F2α) in plasma samples collected from mice. The four plasma isoprostanes are determined by LC-ESI-MS/MS with deuterated 8-iso-PGF2α-d4 as an internal standard (I.S.). For plasma samples spiked with the isoprostanes at a level of 200 pg/mL each, the method imprecision has been below 7.1% and mean inaccuracy equaled 8.7%. The role of dietary fat unsaturation and the supplementation of coenzyme Q have been evaluated in relation to bone health. Male Wistar rats were maintained for 6 or 24 months on two diets varying in the fat source, namely virgin olive oil, rich in monounsaturated fatty acids, or sunflower oil, rich in n-6 polyunsaturated fatty acids. Both dietary fats were supplemented or not with coenzyme Q10 (CoQ10). Bone mineral density (BMD) was evaluated in the femur. Serum levels of osteocalcin, osteopontin, receptor activator of nuclear factor κB ligand (RANKL), osteoprotegerin (OPG), adrenocorticotropin (ACTH) and parathyroid hormone (PTH), as well as urinary F2-isoprostanes were measured. Aged animals fed on virgin olive oil showed higher BMD than those fed on sunflower oil. In addition, CoQ10 prevented the age-related decline in BMD in animals fed on sunflower oil. With higher burden of malaria in Nigeria and other Sub-Saharan African countries coupled with fragile healthcare system and delivery, these may pose a threat in the diagnosis and management of COVID-19 patients co-infected with malaria. Free radicals have been implicated in the progression and pathogenesis of malaria and COVID-19 through Fentons reaction and cytokine storm respectively. METHODS: the current research comprises of seventy-four (74) participants; 20 apparently healthy controls and 54 COVID-19 patients (20 among which were co-infected with malaria). Some diseases and environmental exposures, including those that are risk factors for atherosclerosis, are associated with increased oxidant stress. The objective of this cross-sectional, observational study was to determine whether oxidant stress is increased during human immunodeficiency virus type 1 (HIV-1) infection or its therapy. To quantify oxidant stress, plasma F2 isoprostane (F2-IsoP) concentrations were determined by gas chromatography/mass spectroscopy. A total of 120 subjects were enrolled during routine primary care visits. The median CD4+ T cell count was 341 cells/mm3, the median HIV-1 RNA level was 3.4 log10 copies/mL, and 74% of patients were receiving antiretroviral therapy. Plasma F2-IsoP concentrations were 12-149 pg/mL (median, 31 pg/mL). In univariate analysis, higher F2-IsoP concentrations were associated with lower log10 plasma HIV-1 RNA levels (P = .009) and with efavirenz use (P = .02). This study deals with the important phenomenon of oxidative stress induced by the chronic inflammatory state in rheumatic patients of various categories and stages of disease. It investigates the local (knee joint) and systemic inflammatory response index and oxidative injury by measuring a major PGF2α metabolite, 15-keto-dihydro-PGF2α, and F2-isoprostane in the synovial fluid and blood, respectively. This study reports the results of a multi-laboratory validation study by COST Action B35 to assess inter-laboratory and intra-laboratory variation in the measurement of lipid peroxidation. Human plasma samples were exposed to UVA irradiation at different doses (0, 15 J, 20 J), encoded and shipped to 15 laboratories, where analyses of malondialdehyde, 4-hydroxynonenal and isoprostanes were conducted. The results demonstrate a low within-day-variation and a good correlation of results observed on two different days. However, high coefficients of variation were observed between the laboratories. Malondialdehyde determined by HPLC was found to be the most sensitive and reproducible lipid peroxidation product in plasma upon UVA treatment. Mjölkens förmåga att koagulera är avgörande för tillverkning av ost. Vi har tidigare visat en hög förekomst av icke-koagulerande mjölk från SRB och identifierat mjölkproteiner som potentiella markörer för denna oönskade egenskap. Inom detta projekt kommer vi att öka förståelsen för mjölkproteiners funktionalitet genom att bestämma de exakta aminosyrasekvenserna och variationerna av kaseinvarianterna. Denna information är idag inte tillgänglig på grund av brister i analysmetoder. Projektet kommer därför att ge en unik position för att bidra till en gemensam världsomfattande förståelse av mjölkproteiners funktionalitet och förekomsten av icke-koagulerande mjölk. Detta är viktigt för industriella tillämpningar inom mejeri- och avelssektorn för att förhindra förekomsten av icke-koagulerande mjölk, men också för selektion av mjölk för specifik användning och hälsoeffekter. 锕系元素是第89号元素锕到103号元素铹共15种放射性元素的统称。錒系元素也属于过渡元素,只是錒系元素的外层和次外层的电子构型基本相同,新增加的电子則大都填入了从外侧数第三个电子层(即5f电子层)中,所以錒系元素又可以称为5f系。为了区别于元素周期表中的d区过渡元素,故又将锕系元素及镧系元素合称为内过渡元素。由于锕系元素都是金属,所以又可以和镧系元素统称为f区金属。锕系元素用符号An表示。 1789年德国馬丁·克拉普羅特从沥青铀矿中发现了铀,它是被人们认识的第一个锕系元素。其后陆续发现了钍、锕和镤。铀以后的元素(即超鈾元素)都是在1940年后用人工核反应合成的,屬於人工合成元素,不過也有部分超鈾元素最初是通過人工合成的方式發現,但是後來在自然界中,也發現有痕跡量的存在,例如錼和鈽等。 ...
