The impact of 2,4-dinitrophenol (DNP), cadmium and N-ethyl-maleimide (NEM) shock loads on the composition of the soluble fraction of activated sludge mixed liquor was analyzed by liquid chromatography-mass spectrometry (LC-MS). Fresh mixed liquor was divided in four different batches and subjected to different conditions: no addition of toxin (control) and spike additions of cadmium, DNP or NEM. Samples were taken from each reactor 0, 45 minutes, 3 and 5 hours after the chemical shock. The preliminary results obtained from the LC-MS analysis indicate that there was a significant release of materials from the floc structure into the bulk liquid upon exposure of mixed liquor to cadmium and NEM stress. In the present study, we report the first phenotypic characterization of mice deficient for GPAT3, one of two microsomal GPAT enzymes identified so far at the molecular level. Our data show that total and NEM-sensitive GPAT activity was significantly reduced in Gpat3−/− mice compared with wild-type controls in both subcutaneous and visceral fat depots, whereas NEM-resistant GPAT activity in adipose tissue remained unchanged. In contrast, in liver, although the total GPAT activities were comparable between wild-type and Gpat3−/− littermates, the NEM-sensitive (presumably microsomal) activity was decreased by 30% due to an unexpected increase in NEM-resistant GPAT activity in this tissue. Liquid chromatography-tandem mass spectrometry was described for the determination of mertansine in rat plasma. 50-µL rat plasma sample was pretreated with 25 µL of 20 mM tris-(2-carboxyethyl)-phosphine, a reducing reagent, and further vortex-mixing with 50 µL of 50 mM N-ethylmaleimide for 3 min resulted in the alkylation of thiol group in mertansine. Alkylation reaction was stopped by addition of 100 µL of sildenafil in acetonitrile (200 ng/mL), and following centrifugation, aliquot of the supernatant was analyzed by the selected reaction monitoring mode.
