BACKGROUND: The current study was carried out to determine whether the aqueous-ethanolic extract or the butanolic fraction of nigella sativa (NS) seeds could prevent or reduce calculi aggregation in experimental calcium oxalate nephrolithiasis in Wistar rats. MATERIALS AND METHODS: Male Wistar rats were randomly divided into 5 groups: group A received tap drinking water for 28 days. Groups B, C, D and E received 1% ethylene glycol for induction of calcium oxalate (CaOx) calculus formation for 28 days. Rats in groups C, D and E also received aqueous-ethanolic extract of NS, N-butanol fraction and N-butanol phase remnant of NS, respectively, in drinking water at a dose of 250 mg/kg for 28 days. Urine concentration of oxalate, citrate, and calcium on days 0, 14, and 28, and also serum concentration of magnesium and calcium on days 0 and 28, were measured. A novel water-soluble chitosan (CS) derivative,methoxy poly(ethylene glycol)-O-chitosan- polyethylenimine(mPEG-O-CS-PEI), was synthesized by grafting polycationic polyethylenimine (PEI) and methoxy poly (ethylene glycol) (mPEG) onto chitosan, the copolymer was characterized and confirmed by 1H-NMR and FT-IR spectra. The particle size and zeta potential of mPEG-O-CS-PEI/plasmid complexes were respectively 65 nm and +28.5 mV at the mass ratio of 20:1. Agarose gel electrophoresis confirmed that DNA was retained completely by the copolymer nanoparticles, indicating good DNA package of mPEG-O-CS-PEI nanoparticles. We describe size-selective semipermeable poly(ethylene glycol) (PEG) films which avoid the nonspecific absorption of large proteins but permit the passage of small target molecules. The size threshold for permeation through the PEG films on indium-tin oxide surfaces was characterised using cyclovoltammetry and redox-active probes of 0.3 and 0.6 nm diameter. The permeation was dependent on the molecular weight of PEG and the different conformational preferences of the polymer chains. PEG 5000 D with a looped and dynamically changing structure provided a porous and easily permeable meshwork for the passage of small molecules.
