A novel water-soluble chitosan (CS) derivative,methoxy poly(ethylene glycol)-O-chitosan- polyethylenimine(mPEG-O-CS-PEI), was synthesized by grafting polycationic polyethylenimine (PEI) and methoxy poly (ethylene glycol) (mPEG) onto chitosan, the copolymer was characterized and confirmed by 1H-NMR and FT-IR spectra. The particle size and zeta potential of mPEG-O-CS-PEI/plasmid complexes were respectively 65 nm and +28.5 mV at the mass ratio of 20:1. Agarose gel electrophoresis confirmed that DNA was retained completely by the copolymer nanoparticles, indicating good DNA package of mPEG-O-CS-PEI nanoparticles. The transfection of L-02 cells proved that mPEG-O-CS-PEI/ plasmid nanoparticles only had weak toxicity on the growth of cells compared with that of chitosan. The result of real time quantitative PCR and GFP expression imaging showed that the transfection efficiency of mPEG-O-CS-PEI was significantly higher than PEI and Lipofectin in L-02 cells (P|0.05). We describe size-selective semipermeable poly(ethylene glycol) (PEG) films which avoid the nonspecific absorption of large proteins but permit the passage of small target molecules. The size threshold for permeation through the PEG films on indium-tin oxide surfaces was characterised using cyclovoltammetry and redox-active probes of 0.3 and 0.6 nm diameter. The permeation was dependent on the molecular weight of PEG and the different conformational preferences of the polymer chains. PEG 5000 D with a looped and dynamically changing structure provided a porous and easily permeable meshwork for the passage of small molecules. In contrast, parallel aligned and helical PEG 500 chains represented a denser molecular sieve which is only permeable for small molecules 0.3 nm in size. The activities of methanol, ethanol, 2-propanol, and 1-butanol in poly(ethylene glycol) methacrylate (Mn = 360) solutions have been measured by the isopiestic method at 298.15 K. Sodium iodide and calcium chloride were used as the isopiestic standards for the calculation of activities.
