The present study developed and compared the solid phase extraction and LC-MS/MS analytical approaches for determination of phytoestrogens (8 flavonoids - biochanin A, coumestrol, daidzein, equol, formononetin, genistein, naringenin, apigenin - and 5 sterols - ergosterol, β-sitosterol, stigmasterol, campesterol, brassicasterol) and cholesterol in water. The method was used for analyses of samples collected in stagnant water bodies dominated by different cyanobacterial species. The purpose of this study was to examine the effects of phytoestrogens on luteinizing hormone (LH) and follicle-stimulating hormone (FSH). Phytoestrogens are found in soybean based foods. The rats in a previous study were fed varying amounts of phytoestrogens, euthanized, and their pituitaries were preserved for further research, which is what was used in this study. In the previous study there was an increase in spermatogenesis for the low and high phytoestrogen diet group compared to the pre-treatment and medium group. The LH and FSH levels in the pituitaries were measured and the results were then analyzed to see if there was a correlation in LH and FSH levels to the phytoestrogen diet groups. A coomassie stain was done to ensure that the differences that were seen between the groups were because of varying amounts of phytoestrogens and not varying amounts of protein being loaded into the electrophoresis gels. We evaluated the utility of whole embryo culture for the screening technique of the potency of endocrine active chemicals in rat embryos. Rat embryos on embryonic day 11 (ED11) were exposed to two estrogenic chemicals, estradiol benzoate (EB) and diethylstilbestrol (DES), for 42 hr in culture. The treated embryos showed some morphological defects in the cardiovascular system, such as edema around umbilical vessels (EB- and DES-treated groups), enlargement of the heart (EB) and hemorrhage of the head (DES) at high doses. To determine the mediation of estrogen receptors to these morphological defects, we also studied the expression of estrogen receptor a (ER-a) in embryos both by immunohistochemistry and by Western blotting. We report here the effects of oral micronized estradiol and soy phytoestrogens on uterine weight, choline acetyltransferase (ChAT) and nerve growth factor (NGF) mRNAs in the frontal cortex and hippocampus of ovariectomized young and retired breeder rats. Within each age category, 15 bilaterally ovariectomized rats were randomized equally into three groups: control (OVX), estradiol (E2), and soy phytoestrogens (SBE).

Twenty male hamsters were used for experimentation and animals were divided into a PE (n = 10) and a PE Free group (n = 10) for a period of 3 weeks. Animals were sacrificed, perfused and brains removed for subsequent protein extraction and immunohistochemisty.
