Nowadays the industrial chemistry reactions rely on green technologies. Enzymes as lipases are increasing its use in diverse chemical processes. Epoxidized fatty acid methyl esters obtained from transesterification of vegetable oils have recently found applications as polymer plasticizer, agrochemical, cosmetics, pharmaceuticals and food additives. In this research article, grapeseed, avocado and olive oils naturally containing high percents of mono and poly unsaturations were used as starting materials for the production of unsaturated fatty acid methyl esters. The effect of lauric acid as an active oxygen carrier was studied on epoxidation reactions where unsaturated fatty acid methyl esters were converted to epoxy fatty acid methyl esters using immobilized Candida antarctica Lipase type B as catalyst and hydrogen peroxide as oxygen donor at mild temperature and pressure conditions. After this study it was confirmed by 1H NMR, 13C NMR and GC-MS that the addition of lauric acid to the enzymatic

Glutathione S-transferase (GST) isoenzymes of human pancreas were purified, characterized and evaluated for their possible role in the metabolism of ethanol. Human pancreas has at least two GST isoenzymes belonging to the Alpha class (pI 8.8 and 8.1), one belonging to the Mu class (pI 6.4) and one belonging to the Pi class (pI 4.9). During the purification of GSTs from pancreas as well as from heart, liver, lung, brain and muscle, the fatty acid ethyl ester synthase (FAEES) activity was monitored in order to evaluate the role of GSTs in metabolism of ethanol, as suggested in earlier studies. Both t.l.c. and h.p.l.c. were used to identify ethyl oleate in reaction mixtures to monitor FAEES activity. During the purification of GSTs with the use of affinity chromatography on GSH linked to epoxy-activated Sepharose 6B, FAEES and GST activities from each of these tissues segregated independently. Purified GST isoenzymes from these tissues did not exhibit any FAEES activity. Antibodies raised against ...

Microbial production of fatty acid derived chemicals and fuels is currently of great interest due to the limited resources and increasing prices of petroleum and petroleum-based products. The development of Saccharomyces cerevisiae as a fatty acid ethyl ester (FAEE) cell factory would represent an opportunity for biodiesel production due to its successful history in the biotechnology area. However, fatty acid (FA) biosynthesis is highly regulated and usually not high enough for developing an efficient production process. In S. cerevisiae, FAs are degraded by beta-oxidation and a large fraction is utilized to synthesize steryl esters (SEs) and triacylglycerols (TAGs), which are not essential for the cell. Here, by eliminating nonessential FA utilization pathways, we developed a metabolic engineering strategy resulting in a S. cerevisiae strain that can overproduce FAs and in turn use these for producing FAEEs (biodiesel). Compared to the wild-type, there is an about 3-fold increase in free FA content in
Omega-3 acid ethyl esters are the omega-3 fatty acids eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA), i.e., the ester part, attached at one end to an ethanol molecule - the ethyl part. These FDA-approved prescription products are used in combination with changes in diet to lower blood triglyceride levels in adults with severe (≥ 500 mg/dL) hypertriglyceridemia. This was the first fish oil-derived product to be approved for use as a drug. The first approvals came in Europe in 2001. The first approval in the US came in 2004. These prescription products have been tested in clinical trials. In the US, omega-3 ethyl esters are also manufactured and sold as dietary supplements. Omega-3 acid ethyl esters are used in addition to changes in diet to reduce triglyceride levels in adults with severe (≥ 500 mg/dL) hypertriglyceridemia. In the European markets and other major markets outside the US, omega-3 acid ethyl esters are indicated for hypertriglyceridemia as a monotherapy, or in ...

A fatty acid methyl ester (FAME) can be created by an alkali catalyzed reaction between fats or fatty acids and methanol. The molecules in biodiesel are primarily FAMEs, usually obtained from vegetable oils by transesterification. Since every microorganism has its specific FAME profile (microbial fingerprinting), it can be used as a tool for microbial source tracking (MST). ...

Ester formation depends on the amount of fusel alcohols and organic acids in the wort, but also on the activity of alcohol acetyltranferase enzymes (ATAase I and II), which in turn depends greatly on the yeast strain. Esters in the fermentation can be classified into two groups: The acetate esters (acetate+alcohol) and the ethyl esters (ethanol+fatty acid). The acetate esters are usually formed in greater amounts, but the ethyl esters can be very aromatic even in low concentrations. Common descriptors for the aromas of esters are listed in the table below. The short chain fatty acid esters (C6, C8) are formed early in the fermentation, the medium chain esters (C10,C12) quite evenly throughout the fermentation and the longer chain esters (C16) mostly at the cell-death phase. Increased cell growth usually results in lower levels of esters, due to lower levels of free fatty acids in the wort, as fats are used to build cell membranes. Organic acids are formed throughout the fermentation and at high ...

Honey and agave syrup are high quality natural products and consist of more than 80% sugars. They are used as sweeteners, and are ingredients of cosmetics or medical ointments. Furthermore, both have low water content, are often liquid at room temperature and resemble some known sugar-based deep eutectic solvents. Since it has been shown that it is possible to synthesize sugar esters in these deep eutectic solvents, in the current work honey or, as vegan alternative, agave syrup are used simultaneously as solvent and substrate for the enzymatic sugar ester production. For this purpose, important characteristics of the herein used honey and agave syrup were determined and compared with other available types. Subsequently, an enzymatic transesterification of four fatty acid vinyl esters was accomplished in ordinary honey and agave syrup. Notwithstanding of the high water content for transesterification reactions of the solvent, the successful sugar ester formation was proved by thin-layer chromatography

Wax esters occur widely among bacteria, plants and mammals. Due to their special properties, they have many applications such as lubricants, cosmetics, pharmaceutical products, ink, polishes and candles. They are also considered as potential biofuels and biolubricants in the future. Wax ester producing enzymes synthesize wax esters from alcohols and fatty acyl coenzyme A thioesters. There are three gene families encoding enzymes capable of synthesizing wax esters. Two of them exist in plans; the jojoba-like wax synthase (WS), and the bifunctional wax synthase/diacylglycerol acyl transferase (WS/DGAT). We investigated the phylogenetic relationships among and between WS and WS/DGAT, based upon primary sequence homology of the encoded proteins. Nine candidate genes were chosen for experimental characterization in yeast and Arabidopsis seed. In yeast heterologous expression system, three of the expressed gene products were detected immunologically, Arabidopsis WS At5g55340, a maize WS and a moss WS.

This paper reports the retention data of fatty acid methyl esters on a novel ionic liquid column, SLB-IL111, which has higher polarity than commercially available columns for gas chromatography (GC). The fatty acids analyzed using GC include branched-chain saturated fatty acids, C16-C22 monounsaturated fatty acid cis/trans and positional isomers, and polyunsaturated fatty acids commonly found in natural products. Fatty acids of sardine oil were also analyzed using GC. GC was carried out on a 100-m column under isothermal conditions within the range 120-170°C, using helium as a carrier gas. The retention data of fatty acid methyl esters were represented by equivalent chain length (ECL) values calculated from the retention times of each fatty acid related to those of saturated fatty acids with an even carbon number. This is novel information, obtained on an SLBIL111 column being operated at various temperatures with a single GC system. The results show that the column temperature greatly affected ...
Iso-amyl acetate (the banana ester) production is increased by all the factors that increase esters in general in beer fermentations. Higher gravity worts, lower pitching rates, lower aeration rates and higher fermentation temperatures all promote ester production.. The factors that stimulate 4VG are largely the same as it is a yeast by-product just as iso-amyl acetate is - higher gravity worts, lower pitching rates, lower aeration rates and higher fermentation temperatures.. In addition, open fermentations have been found to stimulate both ester production and 4VG production in hefe-weizens fermented with a hefe-weizen specific yeast strain. Open fermentation also allows the brewer to skim the scum and top-crop the yeast, both of which are traditional practices. (Many commercial breweries have switched to cylindro-conical fermenters, however.) At home, fermenting in a bucket and opening it during the most active part of fermentation can be done. The bucket is not a shallow fermenter, as a ...
0028] Based on the results, it can reasonably be projected that soluble N.sup.α-long chain alkanoyl dibasic amino acid alkyl ester salts will provide synergistic anti-microbial activity when combined with a glycerol mono-fatty acid ester. It would seem that in order to obtain the practical benefits of the synergism between an N.sup.α-long chain alkanoyl dibasic amino acid alkyl ester salt and a glycerol mono-fatty acid ester, the solubility of the N.sup.α-long chain alkanoyl dibasic amino acid alkyl ester salt would need to be sufficient to release a concentration of N.sup.α-long chain alkanoyl dibasic amino acid alkyl ester cation which is greater than microbial inhibitory concentration of that ion in water at RT. Based on the data, it would seem that the absolute minimum solubility required would be approximately 5 ppm in water at RT. Desirably, however, the solubility of the N.sup.α-long chain alkanoyl dibasic amino acid alkyl ester salt should be such that the amount of cation released ...

Fatty acids are a widely studied group of lipids of sufficient taxonomic diversity to be useful in defining microbial community structure. The extraordinary resolution of glass capillary gas-liquid chromatography can be utilized to separate and tentatively identify large numbers of fatty acid methyl esters derived from the lipids of estuarine detritus and marine benthic microbiota without the bias of selective methods requiring culture or recovery of the microbes. The gas-liquid chromatographic analyses are both reproducible and highly sensitive, and the recovery of fatty acids is quantitative. The analyses can be automated, and the diagnostic technique of mass spectral fragmentation analysis can be readily applied. Splitless injection on glass capillary gas chromatographic columns detected by mass spectral selective ion monitoring provides an ultrasensitive and definitive monitoring system. We studied the seasonal lipid dynamics of Calanus finmarchicus from Loch Etive, a semi-enclosed fjordic environment on the west coast of Scotland, in relation to the diapause behavior of this species. Levels of total lipid and triacylglycerol in individual copepodid stage V (CVs) exhibited clear seasonal trends, with highest levels in June and progressively decreasing through late summer to winter. Wax esters in CVs were more variable, but followed a similar pattern. An index of the level of unsaturation of wax esters, effectively an indicator of the levels of polyunsaturated fatty acids in the wax ester reserves, were highest in midsummer and declined progressively through autumn and winter. Fatty acid analysis of total lipid in the CVs confirmed that the amounts of polyunsaturated fatty acids in the wax esters decreased, while amounts of saturated and monounsaturated fatty acids remained comparatively stable. Polyunsaturated wax esters are selectively catabolized by C. finmarchicus during ...

There has been a common understanding that wax esters are poorly absorbed by humans, partly due to outbreaks of the purgative effect named keriorrhea, associated with consumption of oilfish (Ruvettus pretiosus) and escolar (Lepdocybium flavobrunneum). Fillets from these fish species contain up to 20% fat, where 90% of the fat comes as wax esters, resulting in a typical intake of more than 30 000 mg wax esters from one single meal. Orange roughy (Hoplostethus atlanticus) is an attractive food fish with 5,5% fat, where 90% of the fat comes as wax esters. Consumption of this fish gives no unpleasant adverse effects, most likely due to the relatively low fat content that provides approximately 10 000 mg wax ester per 200 grams serving of fish.. In 2015 a randomized, two-period crossover human study, showed that EPA and DHA from oil extracted from the small crustacean Calanus finmarchicus was highly bioavailable and the study concluded that oil from C. finmarchicus could serve as a relevant source of ...

This study has demonstrated the importance of NOME in mediating acute pancreatic toxicity and inflammation induced by alcohol, damage arising primarily from Ca2+-dependent mitochondrial dysfunction in PACs. Many theories have been proposed to explain the detrimental effects of alcohol in AP,22 ,23 including direct actions on Ca2+-release mechanisms,24 OME-sensitised mitochondrial dysfunction25 and formation of toxic FAEEs.26 The association between alcohol and fat in pancreatic damage is particularly intriguing; diets rich in corn oil and alcohol induce chronic pancreatic injury in rats.24 Epidemiological studies suggest that high fat diets may be linked to development of acute and chronic alcoholic pancreatitis,27 while hypertriglyceridaemia is an independent risk factor for both.28 The present data highlight the importance of NOME in mediating acute PAC damage leading to AP. It is likely that under conditions of OME inhibition, available alcohol and fatty acid from triglyceride hydrolysis are ...
A prodrug approach for local and sustained diclofenac action after injection into joints based on ester prodrugs having a pH-dependent solubility is presented. Inherent ester prodrug properties influencing the duration of action include their pH-dependent solubility and charge state, as well as susceptibility to undergo esterase facilitated hydrolysis. In this study, physicochemical properties and pH rate profiles of 3 diclofenac ester prodrugs differing with respect to the spacer carbon chain length between the drug and the imidazole-based promoiety were determined and a rate equation for prodrug degradation in aqueous solution in the pH range 1-10 was derived. In the pH range 6-10, the prodrugs were subject to parallel degradation to yield diclofenac and an indolinone derivative. The prodrug degradation was found to be about 6-fold faster in 80% (vol/vol) human plasma as compared to 80% (vol/vol) human synovial fluid with 2-(1-methyl-1H-imidazol-2-yl)ethyl 2-(2-(2,6 ...
The main objective of this study is to evaluate the clinical utility of meconium analysis for fatty acid ethyl esters as a universal screening tool intended for the detection of newborns at risk for fetal alcohol spectrum disorder. This will be accomplished by assessing the rate of voluntary participation in a nonanonymous neonatal screening program and by determining the logistics of implementing the necessary follow-up and interventions as part of routine care. Additionally, this study will determine the predictive value of fatty acid ethyl ester-positive meconium with regard to neurodevelopmental delays. This is an ongoing prospective cohort study. Written informed consent is sought from all Grey Bruce women delivering at participating birthing sites. Collected meconium samples are tested for fatty acid ethyl esters by headspace-solid-phase microextraction followed by gas chromatography-mass spectrometry. Children with positive results are followed up through an existing public health program

Electron beam- (EB-) induced graft polymerization of acrylic acid and the subsequent immobilization of arginine-glycine-aspartic acid (RGD) peptide onto nanopatterned polycaprolactone with parallel grooves is reported. A high concentration of carboxylic groups was introduced onto the polymer substrate by EB-induced polymerization of acrylic acid. In the coupling of the RGD peptide to the carboxylated polymer surface a three-step peptide immobilization process was used. This process included the activation of surface carboxylic acid into an active ester intermediate by use of 1-(3-dimethylaminopropyl)-3-ethylcarbodiimide hydrochloride (EDC) and N-hydroxysuccinimide (NHS), the introduction of disulfide groups by use of 2-(2-pyridinyldithio)ethanamine tie hydrochloride (PDEA), and final immobilization of the peptide via a thioldisulfide exchange reaction. The extent of coupling was measured by UV spectroscopy. A preliminary study of the in vitro behavior of keratinocytes (NCTC 2544) cultured on the ...

Laurate esters of catechin and condensed tannins were synthesized by both transesterification and conventional acylation techniques. Transesterification was developed as an alternative route to prepare flavonoid fatty acid esters as a prerequisite to advancing potential applications. Resorcinol, catechin, and two condensed tannins were transesterified with vinyl esters, and the resulting products were compared to those formed with lauroyl chloride. The esterified products, including catechin pentalaurate, were characterized both chemically and thermally. Transesterification produced partially substituted derivatives, with a preference for substitution at the catechol ring. Melt features were identified at or below 50°C, which were dependent upon the level of substitution. In addition, the tannin laurates showed thermal stability to at least 100°C and moderate retention of antioxidant properties ...

Precision Engineered Creatine Ethyl Ester & Creatine Ketoglutarate is a unique creatine formula, providing fantastic value for money as well as excellent absorption, effectiveness and bioavailability. Creatine Ethyl Ester is the most absorbable form of creatine available. The ethyl ester has a delivery system that bypasses the need for creatine transporters and is delivered directly into the muscle cells. Once the creatine ethyl ester enters the muscle, the ethyl ester is simply removed leaving creatine in its salt form.
This thesis is built on the concepts of constitutional dynamic chemistry and dynamic kinetic resolution, where reversible covalent reactions are used to generate dynamics and kinetically controlled reactions are employed for resolution. The thesis is divided into two parts:. The first section focuses on the study of dynamic systemic resolution, a concept derived from constitutional dynamic chemistry. Three projects are addressed in this section: 1) lipase-catalyzed resolution of a double parallel dynamic system involving both hemiacetal formation and nitroaldol reaction; 2) resolution of a dynamic α-iminonitrile system through a silver-catalyzed 1,3-dipolar cycloaddition process; 3) resolution of a dynamic imine system via organogelation. Both external and internal selection pressures are applied for the resolution of complex dynamic systems.. The second section explores the asymmetric synthesis of two types of heterocycles through dynamic kinetic resolution. In the first example, a series of ...

Metabolism Information for Isodecyl Benzoate Isodecyl Benzoate (IDB) is an alkyl benzyl ester, also referred to as a carboxylic ester (R-C(O)-OR) produced by reaction with benzoic acid and isodecyl (C10) alcohol. An important feature is that such esters are poorly soluble in water. Esters are reactive with water and can hydrolyze under either acidic or basic conditions. The expected hydrolysis of IDB would be a reverse of the esterification process (generally catalyzed with esterase) and would produce benzoic acid and primary C10 alcohol. Benzoic acid can be further metabolised to hippuric acid and benzoyl glucuronide and excreted in the urine. Some benzoic acid may also be excreted in the urine. The primary C10 alcoholcan be further metabolised by mitochondrial beta-oxidation or by cytochrome P450 mediated omega and omega-minus-one oxidation (may be followed by beta-oxidation). The C10 alcohol can undergo various oxidative steps to yield other smaller alcohols, ketones, aldehydes, or ...

An ester is a carbonyl with an ether group on the carbon atom. The group can be written as COOR. They can be made by reacting a carboxylic acid with an alcohol with some acid. Esters are a very important functional group. They can be found in many substances. They are very common in fats and oils. They sometimes have a very strong, nice smell. They can be used to make perfumes. Polyesters are found in plastics. Esters can also do many reactions. It is very easy to change the ether part of the molecule with another similar one. It is also possible to reduce the ester to a ketone or an alcohol, which can do many different reactions. Nucleophilic addition is also a good reaction, though a bit more difficult than with ketones. There are many ways to find out if a molecule has an ester group. Infrared spectroscopy gives very sharp signals, which are different from other carbonyls. Carbon NMR spectroscopy has similar characteristics. ...
