Both the control and experimental mice were injected intraperitonally with atropine to reduce peripheral activity. Pilocarpine was dosed along with phosphate buffer against the muscarinic and glutamate receptors to the experimental mice to induce seizures in the mice. The purpose of this investigation was to determine the substrate and inhibitor specificity of the esterases present in a water extract of lyophilized peas, and to determine if pea esterases could be classified according to the criteria established for animal esterases. Esterase activity was determined manometrically using the Gilson differential respirometer. The effect of pH on esterase activity was determined using phenyl acetate, phenyl propionate, tripropionin, and tributyrin as substrates. The pH optima appeared to lie between 6.9 and 7.2, and pH 7 was selected for use in this study. The acetyl, propionyl, and n-butyryl esters of phenol, 2-naphthol-6-SO₃ Na and glycerol were hydrolyzed by the enzyme extract. Long chain esters of 2-naphthol-6-SO₃Na, however, were not hydrolyzed. Because of their biochemical diversity and electrophoretic variability, esterases are widely used as genetic markers in gel electrophoresis assays, yet they remain ill-defined. The aim of this study is to characterise esterases from plants with roles in both normal cellular metabolism as well as in xenobiotic metabolism. A survey of esterase activities in four species of crop plants (Triticum aestivum, Zea mays, Glycine max and Oryza sativa) and Arabidopsis thaliana has shown that most of the esterases were typical carboxylesterases containing a catalytically active serine residue. A new esterase from Thermobifida halotolerans (Thh_Est) was cloned and expressed in E. coli and investigated for surface hydrolysis of polylactic acid (PLA) and polyethylene terephthalate (PET). Thh_Est is a member of the serine hydrolases superfamily containing the -GxSxG- motif with 85-87% homology to an esterase from T. alba, to an acetylxylan esterase from T. fusca and to various Thermobifida cutinases. Thh_Est hydrolyzed the PET model substrate bis(benzoyloxyethyl)terephthalate and PET releasing terephthalic acid and mono-(2-hydroxyethyl) terephthalate in comparable amounts (19.8 and 21.5 mmol/mol of enzyme) while no higher oligomers like bis-(2-hydroxyethyl) terephthalate were detected. Similarly, PLA was hydrolyzed as indicated by the release of lactic acid. Enzymatic surface hydrolysis of PET and PLA led to a strong hydrophilicity increase, as quantified with a WCA decrease from 90.8° and 75.5° to 50.4° and to a complete spread of the water drop on the surface, respectively.

To evaluate a leucocyte esterase test as a predictor of gonorrhoea or chlamydia in asymptomatic Aboriginal males at the Central Australian Aboriginal Congress Male Clinic (Ingkintja), first-void urine samples and clinical information were collected from consecutive asymptomatic males presenting to the Ingkintja in Alice Springs between March 2008 and December 2009.
