A proteinaceous elicitor of the plant defense reaction known as the hypersensitive response was isolated from Erwinia amylovora, the bacterium that causes fire blight of pear, apple, and other rosaceous plants. The elicitor, named harpin, is an acidic, heat-stable, cell-envelope-associated protein with an apparent molecular weight of 44 kilodaltons. Harpin caused tobacco leaf lamina to collapse and caused an increase in the pH of bathing solutions of suspension-cultured tobacco cells. Washington State produces about 70% of total fresh market apples in the United States. One of the primary goals of apple breeding programs is the development of new cultivars resistant to devastating diseases such as fire blight. The overall objective of this study was to investigate high-throughput phenotyping techniques to evaluate fire blight disease symptoms in apple trees. In this regard, normalized stomatal conductance data acquired using a portable photosynthetic system, image data collected using RGB and multispectral cameras, and visible-near infrared spectral reflectance acquired using a hyperspectral sensing system, were independently evaluated to estimate the progression of fire blight infection in young apple trees. Biological properties and the genetic basis for biosynthesis of the Germination Arrest Factor (GAF) produced by the soil bacterium Pseudomonas fluorescens are only partially understood. ARS scientists at Corvallis, Oregon, worked with Oregon State University collaborators and demonstrated that GAF also exhibits a rather specific antimicrobial activity against Erwinia amylovora, the pathogen that causes fire blight in apples and pears. This group also demonstrated that certain amino acids reverse GAF inhibition of bacterial growth, an observation which suggests that GAF affects the activity of aminotransferases, enzymes that are involved in plant nitrogen metabolism. Pseudomonas fluorescens EPS62e was selected during a screening procedure for its high efficacy in controlling infections by Erwinia amylovora, the causal agent of fire blight disease, on different plant materials. In field trials carried out in pear trees during bloom, EPS62e colonized flowers until the carrying capacity, providing a moderate efficacy of fire-blight control. The putative mechanisms of EPS62e antagonism against E. amylovora were studied. EPS62e did not produce antimicrobial compounds described in P. fluorescens species and only developed antagonism in Kings B medium, where it produced siderophores. AcrAB-TolC is the major multidrug efflux system in Enterobacteriaceae recognizing structurally unrelated molecules including antibiotics, dyes, and detergents. Additionally, in Escherichia coli it mediates resistance to bile salts. In the plant pathogen Erwinia amylovora AcrAB-TolC is required for virulence and phytoalexin resistance. Exchange analysis of AcrAB-TolC was conducted by complementing mutants of both species defective in acrB or tolC with alleles from either species. The acrB and tolC mutants exhibited increased susceptibility profiles for 24 different antibiotics. All mutants were complemented with acrAB or tolC, respectively, regardless of the taxonomic origin of the alleles. Importantly, complementation of E. amylovora mutants with respective E. coli genes restored virulence on apple plants. It was concluded that AcrAB and TolC of both species could interact and that these interactions did not yield in altered functions despite the divergent ecological niches, to which E. coli and E.

Cotoneaster is an ornamental shrub valued for showy flowers, berries and architecture as well as the ability tolerate adverse conditions under which other taxa fail. Cotoneaster is a highly diverse genus of over 400 species, of which few are available in the US nursery trade. Some species commercially available have been identified as potentially invasive in the state of Oregon and also are susceptible to the bacterial disease fire blight. Cotoneaster selections with reduced fertility and disease resistance would be desirable characteristics for low input landscape plants. Several previous works seem to indicate that flagellar and hrp genes of Erwinia amylovora are counter-regulated. In the present paper, this hypothesis is confirmed by the analysis of several parameters linked to flagella synthesis in E. amylovora grown in hrp- or non hrp-inducing conditions. Moreover, amongst the various hrp mutants tested, the hrpL regulatory mutant only was more motile, produced more flagellins and showed more flagella than the wild-type strain when cultivated on a hrp-inducing medium. A plasmid carrying hrpL introduced into a hrpL mutant decreased its motility and flagellin production. These results strongly suggest that flagellar synthesis of E. amylovora is under the negative control of HrpL, a positive regulator of hrp genes.

Do we recommend biological materials for fire blight management? Overall, the answer is generally yes. There are several important considerations to consider. In our research orchards, the system is challenged with a very high level of inoculum to examine fine differences in product performance. These inoculum levels are much higher than would be present in most commercial orchards. Hence, we expect all programs would perform even better in a commercial setting. In addition, combinations of products seem to be the best: for example, pairing a defense inducer applied at bloom with a protectant material at bloom to control blossom blight, with follow up defense inducer applications for shoot blight. We also expect efficacy of biological materials to improve in the future. We ordered 12 so we could plant 3 around each quarter of the wall - the picture below gives you an impression. We chose this variety since they are disease resistant and the blossom looks gorgeous. Im not sure how long it will take for them to get going; Ill be sure to post some photos of any blossom they treat us with in spring 2020, all being well they survive the planting process.
