It is used as a model organism for studying filamentous growth because of the small size of the genome, its haploidy and ease of culturing. We have followed the migration of GFP-labelled nuclei in multinucleate hyphae of Ashbya gossypii. For the first time we could demonstrate that the mode of long range nuclear migration consists of oscillatory movements of nuclei with, on average, higher amplitudes in the direction of the growing tip. We could also show that mitotic division proceeds at a constant rate of 0. 64 microm/minute which differs from the biphasic kinetics described for the yeast Saccharomyces cerevisiae. Furthermore we were able to identify the microtubule-based motor dynein as a key element in the control of long range nuclear migration. For other filamentous fungi it had already been demonstrated that inactivating mutations in dynein led to severe problems in nuclear migration, i.e. generation of long nuclei-free hyphal tips and clusters of nuclei throughout the hyphae. Some cells cultured in vitro have multiple nuclei. Since cultured cells are used in various fields of science, including tissue engineering, the nature of the multinucleated cells must be determined. However, multinucleated cells are not frequently observed. In this study, a method to efficiently obtain multinucleated cells was established and their morphological properties were investigated. A novel synthetic retinoid, 6-[3-(1-adamantyl)-4-hydroxyphenyl]-2-naphthalene carboxylic acid (CD437), is a selective ligand of the RARgamma nuclear receptor. We examined the in vitro effects of CD437 and found that CD437 induces S phase arrest within 24 to 48 h, followed by cell death, in the p53-negative Hep3B and the p53-positive HepG2 human hepatoma cell lines. Based on observations of cellular and nuclear fragmentation, chromatin condensation, and DNA fragmentation, the CD437-mediated cell-killing effect appears to be due to apoptosis. On morphological examination, a number of CD437-treated cells were found to have increased 5- to 10-fold in size and persisted as single giant cells without cell division, while the remainder underwent nuclear division (multiple nuclei) but were unable to complete cytokinesis, and finally all died by apoptosis. Some of the clones permanently grow and continuously produce large amounts of virus after infection with cytopathic (HTLV-III) variants of these viruses. SUMMARY: Radioactive phenyl mercuric chloride (labelled with 203Hg) was used to measure -SH groups in the mycelium and in the medium. Tritiated phenyl mercuric chloride was used as a quantitative cytological reagent. The changes in -SH concentration of the fungus were not always directly related to the changes in total S in Aspergillus niger, Penicillium chrysogenum 49.133, Eremothecium ashbyii and Candida albicans. A peak in -SH concentration occurred in the cells of these organisms during the early growth phase. Extracellular -SH compounds were produced by all these organisms at some time during development. Cytological studies showed differences in the distribution of -SH compounds in the cell wall and cytoplasm. E. ashbyii and C. albicans had a strong -SH reaction in the cell wall, A. niger and P. chrysogenum 49.133 a weak one. Sulphydryl compounds were most concentrated in areas of hyphal bud formation in E. ashbyii but were not concentrated at the hyphal tip.

Phytophagous insects with wide host ranges often exhibit host-associated genetic structure. We used microsatellite analysis to assess the population structure of the cotton aphid, Aphis gossypii Glover (Hemiptera: Aphididae), a serious pest on many economically important crops worldwide. We sampled aphids from five host plant species in Iran and detected strong population subdivision, with an overall multilocus FST of 0.191. The matrix of pairwise FST values indicated that differentiation between populations collected from different hosts was significantly stronger than between populations from the same hosts. Host-associated differentiation was further supported by Bayesian clustering analyses, which grouped all samples from cotton together with aubergine, and all samples from cucumber together with pumpkin and hibiscus. Background: The limited within-breed genetic heterogeneity and an enrichment of disease-predisposing alleles have made the dog a very suitable model for the identification of genes associated with risk for specific diseases. Canine mammary cancer is an example of such a disease. However, the underlying inherited risk factors for canine mammary tumours (CMTs) are still largely unknown. In this study, 52 single nucleotide polymorphisms (SNPs) in ten human cancer-associated genes were genotyped in two different datasets in order to identify genes/alleles associated with the development of CMTs. The first dataset consisted of English Springer Spaniel (ESS) CMT cases and controls. ESS is a dog breed known to be at increased risk of developing CMTs.
