Its isoenzymatic profile and immunological properties led us to conclude that the increased activity was not due to the expression of an isoform unrelated to the HEX A and B present in other human tissues. Measurement of enzyme levels in the three distinct epididymal regions revealed that in any given sample, activity was higher in the caput than in the corpus or cauda. Moreover, we found a striking correlation between β-HEX activity in the caput region and concentrations of blood testosterone, suggesting a possible involvement of the hormone in modulating enzyme expression. In our previous studies, we showed that a finasteride-induced DHT deficiency may cause changes in the morphology of the seminiferous epithelium without any morphological alteration of the epididymis. In this study, we demonstrated the constitutive immunoexpression of inducible nitric oxide synthase (iNOS) in the testis and epididymis of Wistar rats treated with finasteride for 28 days (the duration of two cycles of the seminiferous epithelium) and 56 days (the duration of one spermatogenesis). Methodology: We evaluated and compared paraffin-embedded tissue samples of 20 ovarian cancer patients with 10 samples from healthy individuals. Adult male rats were injected with TSAA-291 (16 beta-ethyl-17 beta-hydroxyestr-4-en-3-one), a steroidal antiandrogen, daily for 30 days at two doses, viz., 10.0 and 25.0 mg/kg. The treatment caused a dose-dependent reduction in the weights of testis, epididymis, and other accessory sex glands. In the animals that received the high dose, 50% of the rats showed spermatogenic arrest in about 20% of the seminiferous tubules and Leydig cell morphology was normal. The levels of glycerylphosphorylcholine and sialic acid were significantly reduced in the cauda epididymis of all the treated rats. There was a reduction in the number of motile spermatozoa in the cauda epididymis of rats treated with high dose of the antiandrogen. This was accompanied by a reduced number of females exhibiting spermatozoa in their vaginal smears. A combination treatment of the antiandrogen and androgen appears to be a promising approach for fertility regulation in the male.
