Epichlorohydrin (ECH) is a DNA bifunctional alkylating agent that has been shown to form DNA inter-strand cross-links in vitro at the N7 position of guanines at the 5-GNC and 5-GC consensus sequences. We used the quantitative polymerase chain reaction (QPCR) to monitor the formation of in vivo ECH lesions in 6C2 chicken erythroid progerutor cells. Three distinct loci were investigated in order to determine the effect of ECH reactivity in different levels of chromatin condensation. The sites consisted of an open locus of the nuclear genome, a closed locus of the nuclear genome, and a naked locus of the mitochondrial genome. We found that ECH alkylation was preferentially targeted towards the nuclear sites and that chromatin structure had no effect upon ECH reactivity. Repair assays of the three sites revealed that there appears to be no repair of ECH lesions in the nuclear genome.
A comprehensive disposition and metabolism study of epichlorohydrin (ECH) has not been previously reported. In this study, male Fischer 344 rats were dosed (6 mg/kg) orally with [2-14C]ECH (98% radiochemically pure) as an aqueous solution and killed after 3 days. Approximately 38% of the radioactive dose was exhaled as CO2, 50% was excreted as metabolites in the urine, and 3% was present in the feces. Radioactivity in tissues accounted for the remainder of the administered dose. When expressed per gram of tissue, radioactivity was highest in liver, kidney, and forestomach. The half-life of initial elimination of radioactivity in both the urine and exhaled air was about 2 hr, indicating that ECH was rapidly absorbed and metabolized. The major metabolites in the urine were identified as N-acetyl-S-(3-chloro-2-hydroxypropyl)-L-cysteine and alpha-chlorohydrin, about 36 and 4% of the administered dose, respectively. The aim of this thesis was to prepare an insoluble sulphated polysaccharide and explore its potential as a rapid column chromatographic method for the isolation and separation of lipoproteins directly from serum. Regenerated cellulose crosslinked with formaldehyde was found to be relatively inert towards esterification. A degree of sulphation of 0.5 milliequivalents per gram could be obtained but the product had poor hydraulic properties. Epichlorohydrin crosslinked regenerated cellulose was found to be readily sulphated so long as the matrix was highly crosslinked (50-100% v/w). The reactivity of the matrix was retained at low levels of crosslinking if hydroxypropyl groups were introduced with propylene oxide. The hydroxypropylated regenerated celluloses were readily sulphated up to four milliequivalents per gram with minimal degradation of the cellulose chain. The aim of this work is to investigate synthesis conditions of water insoluble β-cyclodextrin (β-CD) nanospheres that could be useful as an adsorbent to remove very low concentration of p-nitrophenol (p-NP) from water. The effect of several parameters on β-CD polymer synthesis such as β-CD/epichlorohydrin (EPI) mole ratio, reaction temperature and NaOH concentration was investigated. The FTIR-ATR analysis confirmed the formation of β-CD polymer structure with the addition of EPI by preserving the basic β-CD structural units. To determine the optimal synthesis conditions, performances of the synthesized β-CD nanospheres were determined by measuring the adsorption efficiencies of p-NP. On the basis of these results, the optimal synthesis conditions of β-CD nanospheres were 65°C, 40% NaOH (w/w) concentration and β-CD/EPI=1/55 mole ratio. To enhance solubility and the performance of a plant gum (from south peach tree of China, SP gum) as a drilling fluid additive, we explored the potential of Modified SP (MSP) gum by modifying it with epichlorohydrin. The modification conditions, such as the mass ratio of the crosslinking agent, pH and temperature, were optimized and the results show that 56.40 g/100g mass ratio of the crosslinking agent to SP gum with pH=10 at the temperature of 50°C lead to the Modified SP (MSP) gum showed the well performance in the water based drilling fluids.It was discovered that drilling fluids modified with 0.5%wt MSP demonstrated more excellent rheology and lower filtration than that of SP treated drilling fluids, which should be contributed to the good clay inhibition obtained from swelling experiment and high inhibitive to the hydration and swell of the mud supported by mud ball immersing experiments .

The performance of a new anion exchanger prepared from raw cassava straw (RCS), for the removal of nitrate from aqueous solutions was evaluated in this study. The cassava straw was modified by epichlorohydrin in the presence of pyridine. The influencing factors, adsorption kinetics, and thermodynamics model of nitrate adsorption onto the modified cassava straw (MCS) were studied. The results showed that the zeta potentials of RCS and MCS were -20.5 mV and +37.3 mV, nitrogen contents (N %) of RCS and MCS were 0.43 and 4.96%, respectively. The current study shows that BH4, known as a cofactor of eNOS, has a glucose-lowering effect in diabetic mice. The BH4-to-BH2 ratio was found to be decreased in various tissues of mice in the diabetic state, indicating deterioration of eNOS bioactivity by eNOS uncoupling. Previous studies have shown that impairment of eNOS function is involved in glucose dysmetabolism and insulin resistance (4,5), which lends support to the notion that alleviation of eNOS dysfunction such as by supplementation of BH4 ameliorates glucose dysmetabolism and insulin resistance. In addition, we found that supplementation of BH4 increased dimerization of eNOS and NO production in the liver of diabetic mice, which strongly suggests alleviation of eNOS dysfunction by recoupling of eNOS. Simultaneously with the restoration of eNOS activity, BH4 elicited a glucose-lowering effect in these mice. No such glucose-lowering effect by BH4 appeared in diabetic mice lacking eNOS.
