TY - JOUR. T1 - Mesenchymal stem cells generate distinct functional hybrids in vitro via cell fusion or entosis. AU - Sottile, Francesco. AU - Aulicino, Francesco. AU - Theka, Ilda. AU - Cosma, Maria Pia. PY - 2016/11/9. Y1 - 2016/11/9. N2 - Homotypic and heterotypic cell-to-cell fusion are key processes during development and tissue regeneration. Nevertheless, aberrant cell fusion can contribute to tumour initiation and metastasis. Additionally, a form of cell-in-cell structure called entosis has been observed in several human tumours. Here we investigate cell-to-cell interaction between mouse mesenchymal stem cells (MSCs) and embryonic stem cells (ESCs). MSCs represent an important source of adult stem cells since they have great potential for regenerative medicine, even though they are also involved in cancer progression. We report that MSCs can either fuse forming heterokaryons, or be invaded by ESCs through entosis. While entosis-derived hybrids never share their genomes and induce ...
A cell-in-cell process identifies the invasion of one living cell into another homotypic or heterotypic cell. the target cells was the common hallmark during the early stage of all cell-in-cell processes which resulted in the accumulation of reactive oxygen species and subsequent mitochondrial injury of encapsulated killer or non-cytotoxic immune cells. However internalized killer cells mediated rapid bubbling of the vacuoles with the subsequent degranulation of GzmB inside the vacuole of the target cells and underwent the reuptake of GzmB by killer cells themselves. The confinement of GzmB inside the vacuole surpassed the lysosome-mediated cell death occurring in heterotypic or homotypic entosis processes resulting in a GzmB-triggered caspase-dependent apoptotic cell-in-cell death of internalized killer cells. On the contrary internalized killer cells from GzmB-deficient mice underwent a typical non-apoptotic entotic cell-in-cell death similar to that of non-cytotoxic immune cells or tumor ...

Project A4: Mechanisms of post‐transcriptional regulation of plakophilin 1 (PKP1) expression and its role in carcinogenesis. Tina studied Biochemistry at the Martin-Luther-University of Halle-Wittenberg. As a PhD student she investigates the posttranscriptional regulation of p120-cell contact proteins by microRNAs. The members of the p120-catenin subfamily of armadillo proteins are essential for cell-cell contact formation. Besides this function in cell-cell adhesion, they participate in various signaling pathways. Their deregulated expression with up- as well as down-regulation has been connected to cancer and metastasis. Because of this it is important to understand how their expression is controlled at the post-transcriptional level by microRNAs and RNA-binding proteins. For example the RNA-binding protein FMRP associates with and inhibits the translation of the p0071 mRNA. In this way FMRP can regulate the actin cytoskeleton in fibroblasts and neuronal cells via the armadillo protein ...

The liver is our largest internal organ and it plays major roles in drug detoxification and immunity, where the ingestion of extracellular material through phagocytosis is a critical pathway. Phagocytosis is the deliberate endocytosis of large particles, microbes, dead cells or cell debris and can lead to cell-in-cell structures. Various types of cell endocytosis have been recently described for hepatic epithelia (hepatocytes), which are non-professional phagocytes. Given that up to 80% of the liver comprises hepatocytes, the biological impact of cell-in-cell structures in the liver can have profound effects in liver regeneration, inflammation and cancer. This review brings together the latest reports on four types of endocytosis in the liver -efferocytosis, entosis, emperipolesis and enclysis, with a focus on hepatocyte biology.
Apart from determining proliferation and migration of keratinocytes within the hair follicle, Rac1 is essential for structure formation of the hair shaft, as shown, for the first time, by our results. In the absence of Rac1 from all differentiating hair follicle compartments, the hair has an abnormal composition, with obvious disturbances of the surface and medulla structure. How could Rac1 regulate structure formation of the hair shaft? It is known that cells of the medulla are tethered to the cortex by cell-cell contacts that contain Dsc2 (Johns et al., 2005; Cai et al., 2009). Considering the role of Rac1 in cell-cell contact formation in vitro (Braga et al., 1997; Takaishi et al., 1997), it is possible that the absence of Rac1 prevents the formation of these cortex-medulla contacts. This could result in the observed chaotic arrangement of medulla cells.. Contacts between cortex and medulla cells form during hair shaft development and are initiated by the formation of pseudopodia in certain ...

To determine whether a synthetic GPCR can enhance activation of killing response, we transfected T cells with a synthetic GPCR from last years team (part# BBA_K209400), that recognizes the small synthetic molecule called CNO. Initially we wanted to use a killing assay to measure the level of immune killer cell activation but unfortunately this assay did not work out (please click here for a detailed explanation). So to measure the level of activation of our T cells we used an assay that measures T cell activation through the production of interleukin 2 (IL-2). IL-2 is a signaling molecule that is expressed by activated T cells. In our assay non-transfected and transfected T cells were incubated with and without CNO (synthetic GPCRs ligand). After incubation the levels of IL-2 produced (presence of IL-2 in supernatant) were measured by ELISA assay. You can see our results in the graph below. The first and second bars (from the left to the right) on the graph below represent non-transfected T ...

Despite expression of immunogenic polypeptides, tumors escape immune surveillance by engaging T cell checkpoint regulators and expanding Tregs, among other mechanisms. What orchestrates these controls is unknown. We report that free C3d, a fragment of the third component of complement, inside tumor cells - or associated with irradiated tumor cells and unattached to antigen - recruits, accelerates, and amplifies antitumor T cell responses, allowing immunity to reverse or even to prevent tumor growth. C3d enhances antitumor immunity independently of B cells, NK cells, or antibodies, but it does so by increasing tumor infiltrating CD8+ lymphocytes, by depleting Tregs, and by suppressing expression of programmed cell death protein 1 (PD-1) by T cells. These properties of C3d appear specific for the tumor and dependent on complement receptor 2, and they incur no obvious systemic toxicity. The heretofore unrecognized properties of free C3d suggest that protein might determine the effectiveness of ...

PY - 2011/6/14. Y1 - 2011/6/14. N2 - Cadherins play a key role in the dynamics of cell-cell contact formation and remodeling of junctions and tissues. Cadherin-cadherin interactions are gated by extracellular Ca2+, which serves to rigidify the cadherin extracellular domains and promote trans junctional interactions. Here we describe the direct visualization and quantification of spatiotemporal dynamics of N-cadherin interactions across intercellular junctions in living cells using a genetically encodable FRET reporter system. Direct measurements of transjunctional cadherin interactions revealed a sudden, but partial, loss of homophilic interactions (τ = 1.17 ± 0.06 s-1) upon chelation of extracellular Ca2+. A cadherin mutant with reduced adhesive activity (W2A) exhibited a faster, more substantial ...

We were quite surprised to find cellular structures that are essentially as old as the organism they reside in, says Salk Vice President, Chief Science Officer Martin Hetzer, senior author and professor. This suggests even greater cellular complexity than we previously imagined and has intriguing implications for how we think about the aging of organs, such as the brain, heart and pancreas. Most neurons in the brain do not divide during adulthood and thus experience a long lifespan and age-related decline. Yet, largely due to technical limitations, the lifespan of cells outside of the brain was difficult to determine. Isotope imaging of different cells inside an islet of Langerhans within the pancreas. Older cells have a yellow-to-pink color scheme, while younger cells exhibit a blue-to-green color pattern. Isotope imaging of different cells inside an islet of Langerhans within the pancreas. The scientific question that drives our research group is how genetic information is transmitted among the diverse cellular constituents of a tumor, and how that impacts on the heterogeneity and plasticity of cancer cells. We want to understand how do mutations arise in cancer cells and how do they spread in cancer cell subpopulations. We want to understand how mutant or aberrantly expressed molecules impact on the interaction between cancer cells, and between cancer and non-cancer cells. We want to understand why cancer cell subpopulations fluctuate over time, and how does it influence, and is influenced, by clinical events such as therapy and disease progression.. We realize that tumor genetic heterogeneity is likely a key determinant of our understanding of tumor development and evolution, and that such diversity among cancer cells inside a tumor is often a confounder in tumor evaluation for diagnosis and treatment. Hence, the study of genetic dynamics of cancer cell populations provides a ...
Our brains are the seat of all we are. Each idea and activity we perform is a yield of our mind. So naturally the prospect of a burden striking the cerebrum can be alarming. Cerebrum growth is an uncommon yet destroying type of disease representing 2% of all malignancy cases around the world. Cerebrum disease alludes to the unusual development and division of cells inside the mind. Mind tumors can be either favorable or destructive and harmful cerebrum tumors are additionally part into essential mind tumors that begin in the cerebrum and optional tumors that begin somewhere else in the body and spread (metastasize) to the cerebrum. Regardless of whether amiable or a threatening tumor can build the volume of the cerebrum which makes weight in the tight skull space. The hard skull is amazingly hard and unbending. Any infringement in this tight space increments intracranial weight which can prompt mind harm, trance state, and even demise. Kinds Of Brain Tumors The main real order of kinds of ...
Blockage from the metastasis procedure remains a substantial clinical problem requiring innovative therapeutic techniques. N6L inhibited Matrigel invasion of MDA-MB-435 cells inside a revised Boyden chamber model. This is associated with a rise in TIMP-3 within the cell tradition medium with out a modification in TIMP-3 mRNA manifestation suggesting its launch from cell surface area and/or extracellular matrix. This can be described by our proven N6L discussion with sulfated glycosaminoglycans and therefore the managed bioavailability of glycosaminoglycan-bound TIMP-3. The Rabbit Polyclonal to OR4A16. implication of TIMP-3 in N6L-induced inhibition of cell invasion was evidenced by siRNA silencing tests showing that the increased loss of TIMP-3 manifestation abrogated the result of N6L. The inhibition of tumor cell invasion by N6L proven in this research furthermore to its previously founded inhibitory influence on tumor development and angiogenesis shows that N6L represents a guaranteeing ...
The eukaryotic kingdom appears not to have contained very many of these TCS genes to start with, and the animal kingdom may just have lost the very few it possessed. This makes sense from the point of view of cellular control because while TCSs are very useful in the small genomed and non-nuclear membrane containing bacteria, its less clear how useful they are in eukaryotes as a whole. Introducing a membrane around the nucleus makes it harder for proteins to get in and bind to the DNA, and introducing systems of membranes inside a far bigger cell makes it harder for a simple two-component system to sense whats going on. Added to which, cells inside a multicellular organism dont really need to sense whats going on, they get told whats going on by the surrounding cells and circulating hormones ...

Our study compares the efficacy of two methods of administration of MAPC for the treatment of ARDS in a sheep model. We evaluated the early distribution and retention of MAPCs delivered by EB and intravenous route, using PET/CT to track the cells in vivo. Despite showing different cell biodistribution, the therapeutic benefit of MAPC was similar by either EB or intravenous route of administration.. This study evaluates for the first time the feasibility of using endobronchial [18F]-FDG as a way to assess MAPC biodistribution. By this route of administration, we observed a trapping of labelled cells inside the lung with no systemic distribution. In ARDS, the equilibrium between the lung interstitial tissue and the capillary vessels is disrupted in early stages of the disease29 and the possible migration of the cells would hypothetically occur in the early stages. The permanence of the cells inside the lower airways after EB delivery observed in this study may be due to multiple factors: the ...

The process of tumour growth and of developing metastases leads to a kind of cellular stress in some cancer cells. Cells inside the tumour are cut off from oxygen and nutrients due to the lack of blood vessels. The tumour cannot grow and metastasise. Reinheckel found higher than normal cathepsin L (CTSL) levels in breast cancer cells, even though they were in an obviously stressed state. He therefore wondered how cells managed to produce such high cathepsin L levels despite high cancer stress conditions. In the absence of oxygen or nutrients, tumour cells are prompted to initiate a programme that shuts down new protein synthesis in order to save material and energy. The cells achieve this by inhibiting the translation of mRNA into protein. Under stress conditions, newly formed ribosomes fail to associate with cytoskeletal mRNA, preventing the formation of new actin molecules. Moreover, cells that are under stress form granular aggregations in which the mRNA can be temporarily wrapped to protect ...
Researchers have developed a new technique that enlists T cells to ferry chemotherapy drugs into tumors and that improves the efficiency of drug delivery.. Cancer cells can elude chemotherapy by hiding out in lymph nodes and other protected locations. Even if a small amount of drug makes it to these refuges, it may not permeate the tissue to reach the tumor cells inside.. Researchers have tried to overcome these problems by having nanoparticles transport chemotherapy drugs, but not all tumors have the leaky blood vessels that enable the particles to leave the bloodstream and disperse into the tumor.. Another approach uses immune cells as carriers, relying on T cells tendency to home in on lymph nodes and the ability of T cells and macrophages to infiltrate tumors. For instance, researchers have inserted liposomes containing doxorubicin into macrophages, which then transported the liposomes into tumors in mice; the macrophage-delivered drug slowed the tumors growth. A limitation of this ...

The PDEFoam method is an extension of the PDERS method, which divides the multi-dimensional phase space in a finite number of hyper-rectangles (cells) of constant event density. This foam of cells is filled with averaged probability-density information sampled from a training event sample.. For a given number of cells, the binning algorithm adjusts the size and position of the cells inside the multidimensional phase space based on a binary-split algorithm, minimizing the variance of the event density in the cell. The binned event density information of the final foam is stored in binary trees, allowing for a fast and memory-efficient classification of events..
