Our genetic screen for suppressors that restore defective ERAD in cdc48-10 cells identified Ssz1p. Being a member of the Hsp70 family, we considered the possibility that Ssz1p acted as a cytosolic chaperone that functions in ERAD by either activating or replacing the defective Cdc48p. The ability of Ssz1p to partially restore the impaired 6myc-Hmg2 degradation in ufd1-2 and npl4-1 mutants could have suggested that Ssz1p replaced the entire Cdc48p-Ufd1p-Npl4p complex in its role in ERAD. However, Ssz1p was not an essential ERAD factor and we could not detect any interaction of Ssz1p with the ERAD-M substrate 6myc-Hmg2. The accurate protein folding is vital for homeostasis, but this process is error-prone since it is chemically complicated. Aberrant folding may result in aggregates having a toxic gain of function or may lead to a loss of protein function; therefore, protein misfolding can lead to several pathologies. The ER protein quality control mechanism monitors the fidelity of protein folding. Those proteins that fail to fold or assemble properly are subjected to degradation via a process known as ER-associated degradation (ERAD). The innate immune response is critical for host defense and must be tightly controlled, but the molecular mechanisms responsible for its negative regulation are not yet completely understood. In this study, we report that transporter 1, ATP-binding cassette, subfamily B (TAP1), a virus-inducible endoplasmic reticulum-associated protein, negatively regulated the virus-triggered immune response. In this study, we observed upregulated expression of TAP1 following virus infection in human lung epithelial cells (A549), THP-1 monocytes, HeLa cells, and Vero cells. The overexpression of TAP1 enhanced virus replication by inhibiting the virus-triggered activation of NF-κB signaling and the production of IFNs, IFN-stimulated genes, and proinflammatory cytokines. Defective proteins are cleared from the ER through a process termed ER‐associated degradation, or ERAD. A crucial step in ERAD is the selection and delivery of soluble misfolded proteins in the ER lumen to the membrane‐associated translocation machinery (Fig 2). EDEM1 has been proposed to have a role in this process by recognizing mannose trimming of N‐linked glycans attached to ERAD substrates. D. Hebert (Amherst, MA, USA) showed that EDEM1 recognizes protein‐folding defects directly, independent of the glycosylation status of a protein (Cormier et al, 2009). Nevertheless, EDEM1 seems to bind to the downstream glycosylated membrane‐associated components of the ERAD machinery through its mannosidase‐like domain. Protein quality control pathways monitor the folding of newly synthesized proteins throughout the cell. Irreversibly misfolded proteins are sorted and degraded to neutralize their potential toxicity. In the secretory pathway, multiple strategies have evolved to test the wide diversity of molecules that traffic through the endoplasmic reticulum. The organelle has adapted the use of N-linked glycans to signal protein folding states. Most apicomplexan parasites harbor a relict chloroplast, the apicoplast, that is critical for their survival. While the apicoplast maintains a small genome, the bulk of its proteins are nuclear-encoded and imported into the organelle. Several models have been proposed to explain how proteins might cross the four membranes that surround the apicoplast, however experimental data discriminating these models is largely missing. Here we present genetic evidence that apicoplast protein import depends on elements derived from the ER associated protein degradation (ERAD) system of the endosymbiont. We identify two sets of ERAD components in Toxoplasma gondii, one associated with the ER and cytoplasm and one localized to the membranes of the apicoplast. The mechanisms of protein quality control in hepatocytes in cases of alcoholic hepatitis (AH) including ufmylation, FAT10ylation, metacaspase 1 (Mca1), ERAD (endoplasmic reticulum-associated degradation), JUNQ (juxta nuclear quality control), IPOD (insoluble protein deposit) autophagocytosis, and ER stress are reviewed. The Mallory-Denk body (MDB) formation develops in the hepatocytes in alcoholic hepatitis as a consequence of the failure of these protein quality control mechanisms to remove misfolded and damaged proteins and to prevent MDB aggresome formation within the cytoplasm of hepatocytes. In the present study, we demonstrate that SND1 promotes immune escape of tumor cells through inhibition of MHC-I antigen presentation pathway, leading to impaired antitumor CD8+ T cell response in tumor microenvironment. Physiologically, the nascent unfolded HC of MHC-I would be stabilized by the chaperone calnexin before association with the β2m in ER. Here, we revealed that SND1 physically interacted with the nascent HC of MHC-I molecule in tumor cells. Instead of promoting the assembly of MHC-I molecule, SND1 recruits the nascent HC to VIMP/VCP complex for ERAD pathway. Our goal is to better understand the processes that maintain the fine balance between newly synthesized and old or misfolded proteins, preserving the protein homeostasis and preventing various diseases.
