Our findings represent evidence of enhanced degradation of elastin outside of the lung in COPD, and we believe that systemic elastin degradation may be a novel systemic feature of COPD. Furthermore, we have found increased expression of MMP-9 in the skin of patients with COPD, a protease implicated in the pathogenesis of emphysema, and these data suggest a role in systemic elastin degradation in COPD.. Mice deficient in the macrophage elastase matrix metalloproteinase-12 (MMP-12) do not develop cigarette smoke induced emphysema or show high levels of macrophage accumulation in the lung on exposure to cigarette smoke. However, monocyte migration to the lung in MMP-12 deficient mice can occur in the presence of an appropriate stimulus. This study used an in vivo model to investigate that stimulus.. While characterizing developmental changes in aortic wall composition in sheep, we observed very rapid accumulation of elastin and collagen in the immediate perinatal period. Thoracic aortic elastin content increased by 41%, and collagen content increased by 49% in approximately 1 week, between 140 days gestation and 3 days postpartum (term = 145 days). Even larger changes were observed in the abdominal aorta. Elastin content increased by 66%, and collagen increased by 57%. The pronounced increase in wall tissue accumulation near birth preceded a marked postnatal increase in arterial pressure. We propose that this elastin and collagen accumulation is a preadaptive response in preparation for the later increase in pressure. Chronic obstructive pulmonary disease (COPD) is a common lung disease with cigarette smoking as the major etiological factor, but only 15% of smokers develop COPD. Destruction of lung elastin observed in COPD is mediated by many enzymes, including cysteine, serine, and matrix metalloproteinases (MMP). The contribution of these enzymes to the lung elastolytic load, released from alveolar macrophages collected from nonsmokers, healthy smokers, and COPD patients, was examined by radiolabeled elastin as substrate in the presence of specific enzyme inhibitors. The activity of MMP was further examined by zymography and Western blotting. COPD macrophages degraded more elastin than either of the other groups. Elastolysis was greatest in the initial 24 h. Through the 72-h culture period, the contribution to elastolysis of serine elastases decreased, MMP increased, and cysteine elastases remained constant. The increased release of elastolytic enzymes in COPD subjects may explain why some smokers develop COPD.

The low-density lipoproteins (LDL) of human serum and very low-density lipoproteins (VLDL) of rabbit serum were incubated with bovine tendon collagen and with porcine aortic elastin. Cholesterol analysis of the elastin residues revealed a significant ability of the elastin to bind the lipoprotein cholesterol. RGD (arginine-glycine-aspartic) containing elastin-like proteins (RGD-ELP), in the structure of TGPG[VGRGD (VGVPG)6]20WPC, was treated to HEK293 cells to construct in vivo-mimic 3-dimensional (3D) spheroid cells. The constructed 3D cells had increased extracellular matrices (ECMs). To identify the induced functions of 3D cells with ECMs, genomic profiles changed in 3D cells compared to the conventional 2-dimensional (2D) cells were analyzed. In HEK293 cells exposed RGD-ELP for 72 h, the total up- and down-regulated genes (824 genes) were selected to be differentially expressed genes. Further analysis by bioinformatics showed that top functions induced in 3D cells were cell proliferation, cell-to-cell signaling, cell differentiation and movement, and connective tissue development and function.
