An enzyme linked immunosorbent assay (ELISA) was developed to detect and quantify Edwardsiella ictaluri in asymptomatic carrier channel catfish Ictalurus punctatus and was compared to conventional culture on agar. Tissue homogenization, using 0.5% Triton X-100 in 0.05 M phosphate buffered saline (pH 7.2), clarified tissue slurries, improved filterability and permitted live bacteria to be filtered from 1 g or more of channel catfish organ tissue. Treatment of tissues with Tween 20 or trypsin did not allow improved filtration or bacterial counts. Interpretive Summary: Technical Abstract: Two major problems in the channel catfish (Ictalurus punctatus) aquaculture industry have been high disease losses to enteric septicemia of catfish (ESC), caused by the bacterium Edwardsiella ictaluri and columnaris disease, caused by the bacterium Flavobacterium columnare. Methods to control these diseases include antibiotic therapy, vaccinations, and management strategies such as taking the fish off feed. Evasion of complement-mediated killing is a common phenotype for many different types of pathogens, but the mechanism is still poorly understood. Most of the clinic isolates of Edwardsiella tarda, an important pathogen infecting both of human and fish, are commonly found serum-resistant. To explore the potential mechanisms, we applied GC-MS based metabolomics approaches to profile the metabolomes of E. tarda EIB202 in the presence or absence of serum stress. We found that TCA cycle was greatly enhanced in the presence of serum. The QRT-PCR and enzyme activity assays validated this result. Furthermore, exogenous succinate that promoting TCA cycle increased serum resistance, while TCA cycle inhibitors (bromopyruvate and propanedioic acid) that inhibit TCA cycle, attenuated serum resistance. Moreover, the enhanced TCA cycle increased membrane potential, thus decreased the formation of membrane attack complex at cell surface, resulting serum resistance. The hybrid from the mating of a female channel catfish (Ictalurus punctatus) with a male blue catfish (Ictalurus furcatus) exhibits heterosis for various performance traits which would make it attractive to the commercial aquaculture industry. However, the widespread use of the hybrid in the aquaculture industry has been impeded by reproductive isolating mechanisms that prohibit the adequate production of fry. The primary research objective was to evaluate various genotypes of channel catfish females to determine if certain genotypes with blue catfish produced increased numbers of fry when hybridized. Fry output and other reproductive traits of seventeen genotypes of channel catfish females when crossed with blue catfish males were evaluated.
