A year ago today we were filled with anticipation, hope, and mostly fear. Fear of the unknown and of what was to be in the days to come. We prayed, for Amelia to be safe and for the donors recovery. The emotions of that day are still there, yet very hard to describe them. Having been through it twice you would think that it would be easy to describe. I hope and pray that somewhere overseas that the donor knows how grateful we are for what they have done for us and will always be apart of our lives. The donor will be remembered as Amelia marks every milestone in her life be it big or small, because no matter she would not have been able to reach any of them had it not been for the amazing donation of their marrow. In the past year we have already begun to be thank for the donors donation. Amelia has done remarkably well well since we have returned home, we had a bump or two but just a few! Its not perfect though, thats what the Beta bit is all about I guess. The history of this novels title is quite a chequered one! Whilst writing, it was called The Edge of the Map and was submitted to the publisher with that title. Its mentioned in the novel a couple of times and sums up many of the themes. But it was felt that it didnt have the right appeal for the audience, so then we had to think up another. The Orphan Myth was indeed an early possibility - well spotted! But again it was generally felt that it had a mournful sound to it and therefore didnt really suit the narrative - and had those awkward ph/th sounds! We went through many ideas, with just about everyone in the know chipping in different suggestions, mostly revolving around mermaids and caves. We ALMOST finished the new Golden Gate Bridge today. The entire deck is done, all of the shingles are done, the guardrails are up along the top. We have to finish guardrails along the climb to the top, and the decent down. Also, the large berm at the bottom of the decent need construction, but all of the materials are in place. We also need to walk the trial and clean up some tree growth. Its going to be a legendary trial! The bridge is absolutely awesome! Its so bid and tall, that the first part is actually a 60 climb. The new jumps trail should be a big hit with everyone! 2 doubles and a table-top jump right before the tunnel section....I cant wait to ride it! Amelia came home one month after she received her new heart. She celebrated her first birthday last October, which was a milestone she would not have met without her new heart. We have watched her learn to sit up, stand, speak her first words, and play with her big brother in the comfort of our home. Looking at her today, you would never know that she had at one time been so sick. We know what a miracle she is, and what a miracle her new heart is. As weve tried to show, even those cases that are considered single gene causation usually arent quite. A gene is a long string of nucleotides with regions of different (or no known) function. Many different variants of that gene can arise by mutation so either the CFTR nor BRCA1 gene as a whole actually causes cystic fibrosis or breast cancer. Instead, what seems to be meant in these cases is that the disease in someone carrying the variant has the disease because (literally) of that variant. Of course, that implies that the rest of the gene is working fine, or at least is not responsible for the disease. That is, only the particular short deletion or nucleotide substitution causes the disease and no other factors are involved. The answer is that it is far less surprising, as a generality. With some few notable exceptions, all the cells in your body have the same genome. This means that while each cell is of a particular type largely because it uses a specific set, but not all, of the genes in the genome. There are, so to speak, blood genes, stomach genes, and so on. Gene expression is based on the physical packaging of chromosome regions and the presence of proteins specific to the cell type, that bind to DNA in regions near to, and that cause the expression of the specifically used genes. But since with few exceptions all the genes still exist in all cells, if one changed these regulatory traits (packaging and so on of DNA, presence of regulatory proteins), one could make the cell do something else. To that end, a lot of stuff in the movie just feels cobbled together much in the manner of Barbossas wooden leg (which, amusingly, doubles as a flask). For some reason, Blackbeard wields a magical sword that lets him control his ship - causing its ropes and tethers to spring to life and attack his enemies, and its cannons to shoot blazing streams of hellfire. How can Blackbeard do all this? Who knows. How and why does he have a colleciton of plundered ships (including The Pearl) magically shrunken inside glass bottles? Who knows? And what was the point of Keith Richards poppoing up again as Jacks father, then mysteirously disappearing, then having no further relevance to the plot? Again - a big WTF type of moment. It is tempting to view what we see today as examples of stages on the way from ordinary two-sex plants to this kind of ones, as if a kind of inevitability. Rather, and here we repeat a figure from the last part of our recent series on natural selection, the evolution of what we see today could have had so many local reasons as to appear essentially random, relative to a straight development to todays system. The idea of an essentially straight path from there to here, so tempting, is in fact in many ways a teleological view of evolution that assumes, at the least, that the same kinds of selection, mutation, ecology and so forth are serially occurring everywhere there are these plants. I have never seen more joyful parents. They are pleased with her choice in Culpepper and if there is any friction between the families it was undetectable. They all were having a great time.
