Polymer ceramic composites obtained as the result of a mineralization process hold great promise for the future of tissue engineering. Simulated body fluids (SBFs) are widely used for the mineralization of polymer scaffolds. In this work an exhaustive study with the aim of optimizing the mineralization process on a poly(L-lactic acid) (PLLA) macroporous scaffold has been performed. We observed that when an air plasma treatment is applied to the PLLA scaffold its hydroxyapatite nucleation ability is considerably improved. However, plasma treatment only allows apatite deposition on the surface of the scaffold but not in its interior. When a 5 wt % of synthetic hydroxyapatite (HAp) nanoparticles is mixed with PLLA a more abundant biomimetic hydroxyapatite layer grows inside the scaffold in SBF. PMID: 10212540 Two hydroxyapatite-coated implants were retrieved after 12 months of loading because of a fracture of the abutments. The specimens were treated to obtain thin ground sections, and a microprobe chemical analysis was done under a scanning electron microscope equipped with an energy-dispersive x-ray analysis and cathodoluminescence system. Under light microscopy, close contact between the bone and the hydroxyapatite coating was seen, with no gaps at the interface. In some areas of the coating a reduction of the coating thickness could be observed, along with the presence of some detached hydroxyapatite particles embedded in newly formed bone. Modern day tissue engineering and cellular therapies have gravitated toward using stem cells with scaffolds as a dynamic modality to aid in differentiation and tissue regeneration. Mesenchymal stem cells (MSCs) are one of the most studied stem cells used in combination with scaffolds. These cells differentiate along the osteogenic lineage when seeded on hydroxyapatite containing scaffolds and can be used as a therapeutic option to regenerate various tissues. In recent years, the combination of hydroxyapatite and natural or synthetic polymers has been studied extensively. Due to the interest in these scaffolds, this review will cover the wide range of hydroxyapatite containing scaffolds used with MSCs for |i|in vitro|/i| and |i|in vivo|/i| experiments. Further, in order to maintain a progressive scope of the field this review article will only focus on literature utilizing adult human derived MSCs (hMSCs) published in the last three years.

Said crystalline hydroxyapatite is prepared by the solid phase reaction wherein a mixture of a calcium phosphate, having a Ca/P ratio of 1.5 to 1.67 by mole, and a stoichiometric amount or more of a calcium compound is heated at a temperature of 500 to 1,000 C. Said calcium compound is at least one compound selected from the group consisting of CaCO3, CaO, Ca(OH)2, CaF2, Ca(COO)2, Ca(NO3)2 and CaCl2.

Special attention is paid to the role of vitamin D in the division and differentiation of osteoblasts and thus, in its role in osteogenesis. In this regard it was interesting to observe the local effects of the active form of vitamin D3 in osteogenesis, as a part of the system for local delivery. Adult subcutaneous fat tissue is an abundant source of multipotent cells. Previous studies from our laboratory have shown that, in vitro, adipose-derived adult stem (ADAS) cells express bone marker proteins including alkaline phosphatase, type I collagen, osteopontin, and osteocalcin and produce a mineralized matrix as shown by alizarin red staining. In the current study, the ADAS cell ability to form osteoid in vivo was determined. ADAS cells were isolated from liposuction waste of three individual donors and expanded in vitro before implantation. Equal numbers of cells (3 x 10(6)) were loaded onto either hydroxyapatite/tricalcium phosphate (HA-TCP) cubes or the collagen/HA-TCP composite matrix, Collagraft, and then implanted subcutaneously into SCID mice. After 6 weeks, implants were removed, fixed, and demineralized and sectioned for hematoxylin and eosin staining. Osteoid formation was observed in 80% of HA-TCP implants loaded with ADAS cells. Bone defects due to trauma or disease have led to the need for biomaterials as substitutes for tissue regeneration and repair. Herein, we introduce a porous triphasic ceramic-coated hydroxyapatite scaffold (HASi) for such applications. Interestingly, in the degradation experiments with isotonic buffer, HASi showed a significant release of silica with the disappearance of the tricalcium phosphate phase. Furthermore, the material also exhibited cytocompatibility with cultured bone marrow-derived mesenchymal stem cells of human origin. The material chemistry, together with the favorable cellular characteristics, indicates HASi as a promising candidate for critical-size bony defects, which still remains a formidable clinical challenge in the orthopedic scenario.

In this study one of classical antibiotic compound nitrofurantoin was studied as micro and nano particles in electrochemical analysis by cyclic voltammetric technique using glassy carbon electrode in blood medium to observe the oxidative effect on the blood components. It was found oxidation - reduction current peaks of nitrofurantoin in blood medium at +1 and -0.75 V respectively. The oxidation current peak was disappeared for the nitrofurantoin nano particles in blood medium, but for micro particles is still oxidize the blood. Local bisphosphonate release versus hydroxyapatite coating for stainless steel screw fixation in rat tibiae
Implant fixation in bone can be improved by a coating that delivers bisphosphonates locally, or by a hydroxyapatite (HA) coating. In this study, we compared these different types of coatings. For mechanical testing, 30 rats were assigned into three groups, and similar screws were implanted bilaterally in the proximal tibiae. The rats received screws that were either uncoated, coated with nano-crystalline hydroxyapatite or coated with a bisphosphonate releasing protein matrix. After 4 weeks, one screw was subjected to pull-out testing, and the contra-lateral one to torsion testing. For morphology, 30 rats were assigned to similar treatment groups, but received only one screw each. Bisphosphonates enhanced the pull-out force by 41% (P = 0.02) compared to controls, HA increased the pull-out force although not significantly. In order to obtain the properties of the sintered as-dried calcium phosphate with [Ca]/[P] = 1.50, the characteristics of sintered pellets have been investigated using X-ray diffraction (XRD), inductively coupled plasma-mass spectrometry (ICP-MS), Fourier-transform infrared (FT-IR) spectra, Vickers hardness indentation and scanning electron microscopy (SEM). When the pellet samples were sintered between 700 °C and 1200 °C for 4 h, the hydroxyapatite (Ca10(PO4)6(OH)2, HA) still maintained the major phase, accompanied with the rhenanite (NaCaPO4) as the secondary phase and β-tricalcium phosphate (β-Ca3(PO4)2, β-TCP) as the minor phases. Deformation of Nanocrystalline Metals under Nanoscale Contact
The nanoindentation of a columnar grain boundary (GB) network in nanocrystalline Al has been examined by atomistic simulation. Our intent was to gain fundamental understanding on the relationship between structure evolution at GBs and incipient plasticity for indenter tips significantly larger than the average grain size. The nanoindentation simulations were performed by quasicontinuum method at zero temperature. A GB network made of vicinal and high-angle |110| tilt GBs was produced by generating randomly-oriented 5-nm grains at the surface of a 200 nm-thick film. The major findings of this investigation are that (1) nanocrystalline GB networks profoundly impact on the nanoindentation response and cause significant softening effects at the tip/surface interface; (2) GB movement and deformation twins are found to be the predominant deformation modes in columnar Al, in association with shear band formation by GB sliding and intragranular slip, and crystal growth by grain rotation and coalescence; and (3)

The dynamics of nanocrystalline materials has attracted a lot of scientific interest during the last decade due to the striking differences observed for their atomic vibrations relative to the bulk counterparts. These anomalies are the enhancement of their density of phonon states (DOS) at low and high energies and broadening of the phonon peaks [1]. In addition, the energy dependence of the low-energy part of their phonon DOS has been a source of long-standing debates. The experimental results are contradictory, reporting a linear dependence [2], a power law with n = 1.33 [3], and a quadratic (Debye-like) behaviour ([4, 5], and references therein). Flynn in the CTBA Stakes @ Del Mar today...she set the track record @ Pleasanton in her debut, smoking 5 furlongs in 21.55, 44.06, 55.92 (note the final furlong in under 12 seconds)...Pleasanton does run fast, but i checked the charts from thatr day, and this filly ran considerably faster than everyone else on the card...two hporses fromn that race have already returned to win, and shes trained by Steve Miyadi, who is hitting a whopping 32% from 245 starters (i guess you can add him to the list of super-trainers)...shes 3/1 on the morning line, but the fact shes coming out of a maiden claimer @ Pleasanton should keep the price fair...i like this filly a lot today.... ...
