Detection of hepatitis B virus (HBV) serological markers in dried blood spot (DBS) samples by enzyme immunoassay (ELISA) has not yet been fully optimized. In this study, the ability to detect three HBV markers (HBsAg, anti-HBc, and anti-HBs) was evaluated in DBS samples using a modified commercial ELISA. Matched serum and DBS samples were obtained from individuals with or without a past history of HBV infection. Sera samples were tested according to the manufacturers instructions, but for DBS testing, paper diameters, elution buffer, volume of input sample, and cut-off values were evaluated to optimize the assay. Stability studies were done on DBS stored at for up to 180 days at different temperatures. The absorbance values that yielded the maximum sensitivity and specificity were determined based on the area under the ROC curve (AUROC) and chosen as the cut-off value. In this study, we compared the QFT and the IP-10 tests for the diagnosis of LTBI in a cohort of 6th grade school children from an urban setting in Finland. IP-10 was detected in both plasma samples and dried plasma spots and analysed using pre-set test algorithms; the two IP-10 methods were 100% concordant. Two children were deemed IP-10 and QFT positive. The QFT test showed 97% agreement with both the IP-10 tests. The children were followed up for 4 years and no new cases were identified, rendering a very high negative predictive value for both QFT and IP-10 tests.. Mice are anesthetised prior to stimulation with clotting agents such as thrombin or collagen, and radiolabelled platelets are tracked in real-time, instead of the standard approach of studying clot formation in conscious mice using death as an end-point (with its associated pain and suffering). The refined model has scientific benefits and in addition to reducing the severity level of the procedures, reduces the number of animals used per study from 200 to 30 mice. Despite this there has been relatively low uptake of the refined model and the mortality model continues to be reported in the scientific literature from a range of labs worldwide.
