Ribosomal DNA (rDNA) of plants is present in high copy number and shows variation between and within species in the length of the intergenic spacer (IGS). The 45S rDNA of flowering plants includes the 5.8S, 18S and 25S rDNA genes, the internal transcribed spacer (ITS1 and ITS2), and the intergenic spacer 45S-IGS (25S-18S). This study identified six different types of 45S-IGS, A to F, which at 363 bp, 1121 bp, 1717 bp, 1969 bp, 2036 bp and 2111 bp in length, respectively, were much shorter than the reported reference IGS sequences in B. oleracea var. alboglabra. The shortest two IGS types, A and B, lacked the transcription initiation site, non-transcribed spacer, and external transcribed spacer. Respiratory infection in cystic fibrosis (CF) is polymicrobial, but standard sputum microbiology does not account for the lung microbiome or detect changes in microbial diversity associated with disease. As a clinically applicable CF microbiome surveillance scheme, total sputum nucleic acids isolated by a standard high-throughput robotic method for accredited viral diagnosis were profiled for bacterial diversity using ribosomal intergenic spacer analysis (RISA) PCR. Conventional culture and RISA were performed on 200 paired sputum samples from 93 CF adults; pyrosequencing of the 16S rRNA gene was applied to 59 patients to systematically determine bacterial diversity. Two xylose-utilizing yeast strains isolated from rotten wood collected in the rainforest in different mountains of Hainan province, southern China, were studied. Sequence analysis of the large subunit rDNA D1/D2 domain and internal transcribed spacer region revealed that the strains represent a novel anamorphic yeast species, for which the name Candida cellulosicola sp. nov. is proposed; the type strain is HNX16-2T ( = CGMCC 2.3503T = CBS 11952T). Nuclear ribosomal internal transcribed spacer (ITS) DNA sequences are used to estimate the phylogeny of 53 members of Polemoniaceae, representing all but two genera of the family. Fitch parsimony analysis of equal-weighted nucleotide sites result in 1080 minimal-length trees. However, when alignment-ambiguous positions are removed and an II: 10 transition to transversion weighting is imposed only eight trees are found. These data are used to address two issues: I) patterns of diversification in Polemoniaceae, and 2) the circumscription and monophyly of the genus Gilia. Although the monophyly of Polemoniaceae is well supported, relationships inferred among the earliest diverging lineages are altered by character weighting, treatment of indels, and taxon inclusion. In spite of the lack of reliable resolution at the basal nodes, ITS data provide evidence that Gilia, as currently interpreted, is polyphyletic and comprises at least five independent lineages.

A phylogeny of the fungal phylum Basidiomycota. is presented based on a survey of 160 taxa and five nuclear genes. Two genes, rpb2, and tef1, are presented in detail. The rpb2 gene is more variable than tef1 and recovers well-supported clades at shallow and deep taxonomic levels. The tef1 gene recovers some deep and ordinal-level relationships but with greater branch support from nucleotides compared to amino acids. Intron placement is dynamic in tef1, often lineage-specific, and diagnostic for many clades. Introns are fewer in rpb2 and tend to be highly conserved by position.
