The beginnings of this partnership stretch directly to Louis Pasteur and his establishment of the Pasteur Institute in 1888. Pasteurs success with rabies vaccine, and then the discovery of diphtheria antitoxin in the 1890s, inspired the foundation of many similar organizations around the world dedicated to producing public health products. In 1894, the Ontario Board of Health began importing a commercial supply of diphtheria antitoxin from the U.S., and also established the Ontario Vaccine Farm to provide smallpox vaccine. For the next decade, concerns grew in Canada about the price and quality of imported diphtheria antitoxin. There was also pressure from the Canadian Public Health Association, and others, on the federal government to assume responsibility for producing the antitoxin, or at least regulate its quality. In 1910, a rabies outbreak in Ontario also brought calls for the establishment of a Pasteur Institute affiliated with the University of Toronto, but little developed.. Meanwhile, ...
Accurate determination of diphtheria toxin antibodies is of value in determining the rates of immunity within broad populations or the immune status of individuals who may be at risk of infection, by assessing responses to vaccination and immunization schedule efficacy. Here we report the results of an external quality assessment (EQA) study for diphtheria serology, performed within the dedicated surveillance network DIPNET. Twelve national laboratories from 11 European countries participated by testing a standard panel of 150 sera using their current routine method: Vero cell neutralization test (NT), double-antigen enzyme-linked immunosorbent assay (ELISA; DAE), dual double-antigen time-resolved fluorescence immunoassay (dDA-DELFIA), passive hemagglutination assay (PHA), toxin binding inhibition assay (ToBI), and in-house or commercial ELISAs. The objective of the study was not to identify the best assay, as the advantages and drawbacks of methods used were known, but to verify if laboratories ...
In Brazil, until 2004, the immunization policy against diphtheria involved childhood vaccination with no official routine booster dose administered after 15 years of age. This study assessed functional antibody levels against diphtheria among blood donors. A total of 140 blood samples were collected, and diphtheria antitoxin levels were evaluated by Vero cell neutralization test. The mean age of the population was 34 years old (range: 18-61 years); 37.8% females and 62.2% males. Overall, 30.7% (95%, CI: 23.4-38.7) individuals presented neutralizing antitoxin antibody titers < 0.01 IU/ml; 42.1% (95%, CI: 34.1-50.4) showed values between 0.01-0.09 IU/ml and, 27.1% (95%, CI: 20.2-34.9) had ≥ 0.1 IU/ml. In the subgroup of individuals with history of diphtheria immunization during childhood (85%), a number of 28.5% showed unprotective levels of circulating neutralizing antibody (< 0.01 IU/ml). Despite the continuous progress of immunization programs directed to Brazilian population, currently ...

This ECDC gap analysis demonstrates that there are potentially significant gaps in diphtheria diagnostic capacity within the EU/EEA in terms of surveillance, specialised laboratory diagnostics, expertise and availability of diphtheria antitoxin (DAT). The results highlight the importance of sound surveillance systems for diphtheria and laboratory personnel training, as well as the need to ensure access to immunisation and high vaccination coverage rates.. ...
Task 5. Test at cistinase (Pizy test). In the column of gelose with cistin by a prick sow the studied culture and put a test tube in a thermostat. In 1 day a medium is blaking on motion a prick, on the depth of 1 cm view a brown cloud appears from a surface. Difteroidy does not form a cloud .. Task 6. Test at urease (Zaksa test). Determination of urease is made sowing on media with urea. As an indicator in a nutrient media, bring in phenolic red. A nutrient medium with culture Corynebacterium spp. put on 30 minutes in a thermostat (37 C). For this time an urea fissions with formation of ammonia and an medium becomes red (a test is positive, because changes).. Task 7. The indirect hemagglutination test determine the antitoxin antibodies concentration in blood serum. It is made according to the formula: X=10*A/B, where X is content of diphtheria antitoxin in assayed serum (IU), 10- titer of control serum (IU/ml), A - maximal dilution of assayed serum with positive result, B - maximal dilution of ...
What were you doing seven years ago? Me, I was living in Washington, DC and enjoying my first "real" job out of college. Not yet married, but dating my now husband. Schick? They were designing the next best thing in razors - the Schick Hydro Silk™, launching at the end of the month. After seven years of testing, Schick says they have made the shaving experience better. This is welcome news. Razors have come a long way. My young nieces will likely never know the hacking my knees and ankles suffered. (I have bad vision - shower without contacts on. Mistakes happen(ed). I dont want to talk about it.). Im hoping after a few more months with the Tria, I wont have to use this on my underarm or bikini line, but my legs see a lot of razor time, especially in the summer. Ive been using the Hydro Silk for a while now, and wanted to share my thoughts.. Schick infused this razor with a moisturizing serum that activates with water to prevent skin from being stripped of essential moisture and the ...

Overall, the company believes there is alignment with the FDA on its panel of analytical methods and the proposed assay qualification/validation plans. Analytical methods are used to assess how reliably and consistently the key product characteristics can be determined in order to ensure patients receive safe and effective product.. In the meeting minutes, the FDA made a request that the company complete implementation of its potency assay qualification plan, presented in the meeting, prior to initiation of upcoming clinical studies. This assay utilizes the Delta 7 mouse model, which has been used historically to assess AVXS-101 potency but now incorporates additional elements to make it acceptable to global regulatory authorities. Specifically, the agency stated, "We believe that your potency assay development and qualification plans have the potential to result in an acceptable potency assay, and we are encouraged that the assay includes reference controls that allow for calculation of ...

Cell Based Assays - Cell based assay are preferred when the biopharmaceutical contains multiple functional domains which will all need to be intact in order for the product to be effective. For example, if the biopharmaceutical protein contains a membrane translocation domain and a transcription factor domain, a cell based assay would be able to show the activity of both functional domains, the transcription factor domain by using a reporter gene and the membrane translocation domain because the protein would need to be able to enter the cell to activate the gene. Custom cell based assays could be developed by PBL depending on your needs. The staff at PBL would be happy to discuss your product with you and help determine what type of cell based assay would work best for your product ...

In 1826, Pierre Bretonneau, a French physician, called the disease diphtérite and distinguished diphtheria from scarlet fever. In 1883, Swiss-German pathologist, Edwin Klebs, identified the bacterium that causes diphtheria and in 1884, Friedrich Loeffler, first cultivated the bacterium, which resulted in its first name of Klebs-Loeffler bacterium (it is now called Corynebacterium diphtheria). In 1890 Kitasato Shibasaburo, a Japanese physician and bacteriologist, and Emil von Behring, a German physiologist who would win the Nobel Prize in Medicine in 1901 for his work on diphtheria, successfully immunized guinea pigs with a heat-treated diphtheria toxin. On Oct. 16, 1894 two Cincinnati physicians successfully treated a two-year-old girl with a diphtheria antitoxin. On Dec. 4, 1894 the New York City Board of Health regulated the purity and potency of diphtheria antitoxin. In 1895 the Mulford Company of Philadelphia (later Merck) and the New York City Health Department started producing and ...
1. The immunological properties of two contrasting types of human antisera, each containing a high titer of diphtheria antitoxin, have been investigated. 2. Sera which contain only non-precipitating antitoxin exhibit most of the properties of atopic reagin-containing sera. This type of antitoxin is capable of sensitizing normal human skin to toxin or toxoid and remains for many weeks in the injected area. It exhibits no Danysz effect, does not fix complement unless very large amounts of serum are used, and can be specifically coprecipitated by addition of precipitating antitoxin and toxin. On the other hand, it is capable of sensitizing guinea pigs to fatal anaphylactic shock. Heating at 56°C. for 4 hours destroys the skin-sensitizing properties and results in almost quantitative conversion to a modified antitoxin which is capable of blocking the wheal and erythema reaction caused by injection of toxoid into sensitized skin. Heating at 56°C. does not result in an appreciable loss of ...
As a devoted employee of the College of Physicians of Philadelphia, I must defend the holdings of Mütter Museum and the Colleges Historical Medical Library. Just yesterday I was donning rubber gloves, assuring our curator that I was up on my tetanus shots, and looking through 19th-century diphtheria antitoxin syringes-some capable of delivering up to seven ounces of horse-derived antitoxin. (Ouch.). But as for specific items, I personally am very fond of our copy of William Heberdens "Some Account Of the Success of Inoculation for the Small-Pox in England and America together with Plain Instructions, By which any Person may be enabled to perform the Operation, and conduct the Patient through the Distemper." The pamphlets introduction is by Benjamin Franklin, who encouraged Heberden to write the pamphlet. Franklins own son had died of smallpox at age four. Theres also an original letter from Louis Pasteur to Emile Roux that is quite curious, and an old Thomas Dover text, and… I could go ...
The complex life cycle of the malaria parasite involves several developmental stages in the mammalian host that provide opportunities for vaccine intervention. The effector arm of the immune response that protects against malaria is specific for each stage of the parasite life cycle. While CD4+ and CD8+ T-cells are required to mediate protection during the liver stage, antibodies play a major role before parasite entry into the liver and during the blood stage. Induction of cytotoxic T-cells or those producing IFNgamma has become a major goal for T-cell-inducing vaccines, and the liver stage is currently one of the preferred targets for malaria vaccine development. T-cells can effectively be primed by several vaccine strategies, including recombinant vectors. A wide range of such vectors is currently available and their use alone or in combination induces high frequencies of antigen-specific T-cells in animal models and humans. Therefore, traditional potency assays, such as the highly sensitive ...

Bacterial toxin-antitoxin loci consist of two genes: one encodes a potentially toxic protein, and the second, an antitoxin to repress its function or expression. The antitoxin can either be an RNA or a protein. For type I and type III loci, the antitoxins are RNAs; however, they have very different modes of action. Type I antitoxins repress toxin protein expression through interacting with the toxin mRNA, thereby targeting the mRNA for degradation or preventing its translation or both; type III antitoxins directly bind to the toxin protein, sequestering it. Along with these two very different modes of action for the antitoxin, there are differences in the functions of the toxin proteins and the mobility of these loci between species. Within this review, we discuss the major differences as to how the RNAs repress toxin activity, the potential consequences for utilizing different regulatory strategies, as well as the confirmed and potential biological roles for these loci across bacterial species.

The concept of "allergy" was introduced in 1906 by pediatricians Viennese Clemens von Pirquet and Bela Schick who were the first to observe how the immune system could also play a harmful role with altered responses after administration of heterologous serum or smallpox vaccine for therapeutic purposes. Pirquet and Schick called this phenomenon "allergy", words from greek ἄλλος, which means "other", and ἔργον meaning "work" . At first all forms of hypersensitivity were classified as allergies, because they thought they were all caused by an altered immune system activation. It became clear later that they were involved many different mechanisms of disease, having in common an abnormal immune system activation. In year 1963, PhilipGell and RobinCoombs proposed a new classification scheme which provided for 4 types of the hypersensitivity reactions, identified as hypersensitivity grade I to IV . With this new-fangled arrangement, the term "allergy" was controlled to merely one type I ...
Visit the AGR in the Balkwill to see artwork by local artists: in main gallery, Brendan Schick: the Metafold. Step through the looking-glass into a shadowy dream space, where things seem very real but are somehow just wrong enough to be unsettling. A silver teapot pours drapery onto a tablecloth, an ornately framed mirror reflects a distorted gingham pattern, fleshy blobs float over twisted sheets. These uncanny images are rooted in nightmare-like trance states Schick experienced in childhood. They also speak of the uncertainty that we experience with our body, and our sense of self. ...

Potency testing is critical for the regulatory approval and lot release of biologics. In particular, FDA and EMA guidelines mandate that customized potency assays be developed and validated prior to clinical trials. This testing is commonly done through in vivo or in vitro bioassays, as advised by global regulatory agencies, to demonstrate product efficacy and manufacturing consistency. Because potency assays are essential in verifying product quality, it is critical to choose a testing partner that understands your development needs ...
