Exposure to high concentrations of persistent organochlorines may cause fetal toxicity, but the evidence at low exposure levels is limited. Large studies with substantial exposure contrasts and appropriate exposure assessment are warranted. Within th
Every year, 250 million people are infected with malaria, resulting in 880,000 deaths, primarily to African children under age five. Many malaria-endemic countries use Indoor Residual Spraying (IRS), the application of dichlorodiphenyl trichloroethane (DDT) or pyrethroid insecticides to interior walls of homes, to control malaria. Concerns regarding the unintended consequences of chronic, high-level DDT exposure have resulted in considerable international debate. For example, the Stockholm Convention on Persistent Organic Pollutants aims to ban DDT, while the World Health Organization and the U.S. Agency for International Development promote and/or fund IRS, including with DDT. DDT and pyrethroids are neurotoxicants in insects and animals. Human studies, primarily conducted in Western populations with low to moderate levels of exposure, suggest that prenatal exposure to DDT and its breakdown product, DDE, may adversely affect childrens neurodevelopment. DDT/E exposure has also been related to ...
dichlorodiphenyltrichloroethane is understood for its pre-judicious result on the setting and life. However, the new kind was found to be more practical against insects and in smaller amounts, probably minimizing its environmental impact.. In continued to explore the crystal structure of insecticde, the researchers began learning fluorinated varieties of dichlorodiphenyltrichloroethane, swapping out atomic number 17 atoms for halogen.. They ready 2 solid varieties of the compound a mono fluoro and a difluoro analog and tested them on fruit flies and mosquitoes, as well as dipterous insect species that carry protozoal infection, black vomit, Dengue, and Zika.. The solid varieties of fluorinated dichlorodiphenyltrichloroethane killed insects a lot of quickly than did DDT; the difluoro analog, referred to as DFDT, killed mosquitoes 2 to fourfold quicker, the researchers aforesaid.. A Hydrogenomonas cleaved one of the rings of p,p-dichlorodiphenylmethane, a product of DDT metabolism, to yield p-chlorophenylacetate and further metabolized the latter compound. Products of microbial degradation of other diphenylmethanes were also identified. Substituents on the methylene-carbon and para-chloro substitution are critical factors governing resistance of DDT and related compounds to aerobic metabolism and decomposition by the bacterium. ...

In this study, the primary goal is to examine the relationship between DDT (dichlorodiphenyltrichloroethane) levels and the odds of loss of clinically-recognized pregnancies amongst women in Limpopo, South Africa. The initial protocol (which will be referred to as Part 1) planned to enroll 2,400 non-pregnant women, draw blood for measurement of DDT exposure, and follow 1,200 pregnant participants to ascertain occurrence of miscarriage. An initial two-year pilot of 850 non-pregnant women was proposed to evaluate field procedures, recruitment strategies and the reproducibility of DDT levels. Of the 444 women enrolled in the first 10 months of the pilot of Part 1, only 27 have become pregnant. Although the field procedures have been well executed, the number of pregnancies has been low. Based upon the experience to date, the approach taken so far is unlikely to generate enough pregnancies to monitor the occurrence of miscarriages. Therefore, the revised protocol (referred to as Part 2) will modify ...
One or more organochlorine compounds were detected in all women in the LIBCSP, although median levels were as much as 10-fold lower than those reported earlier in the 20th century. Multivariate models reconciled the different directions of correlations of DDE and PCB with BMI that were found in simple or age-adjusted correlations (Tables 1 and 2; Fig. 2), in a manner consistent with the pharmacokinetic models. When both BMI and weight gain were included in multivariate models, the direction of the associations was identical for DDE and PCB. Therefore, the statistical models corrected the apparent positive association of DDE with BMI-gain (e.g., Fig. 3) to a negative coefficient in the model (e.g., Table 4), whereas at the same time, after multivariate adjustment, the inverse correlation of PCBs with BMI became positive.. From predicted organochlorine model shown in Fig. 3, highest DDE levels were in women who were heaviest, had the least adult weight gain, and did not lactate. Lowest levels were ...

What is 1,1-Dichloroethylene? 1,1-Dichloroethylene (1,1-DCE) is a man-made substance used to make fire retardant fibers and polyethylene food wraps. It is a clear, colorless, flammable liquid with a mild, sweet smell. Dichloroethylene evaporates quickly at room temperature and can pollute the air. When 1,1-DCE gets into water or on soil it evaporates quickly. However, in groundwater or soil, it breaks down very slowly. Most 1,1-DCE in the environment results from industrial use and improper disposal.
BACKGROUND: We performed a cross-sectional study of associations between personal characteristics and lipid-adjusted serum concentrations of certain PCB congeners and chlorinated pesticides/metabolites among 323 pregnant primiparous women from Uppsala County (age 18-41 years) sampled 1996-1999. METHODS: Extensive personal interviews and questionnaires about personal characteristics were performed both during and after pregnancy. Concentrations of organochlorine compounds in serum lipids in late pregnancy were analysed by gas chromatography. Associations between personal characteristics and serum levels of organochlorine compounds were analysed by multiple linear regression. RESULTS: Participation rate was 82% (325 of 395 women). Serum concentrations of PCB congeners IUPAC no. 28, 52, 101, 105 and 167, and o, p-DDT and -DDE, p, p-DDT and -DDD, oxychlordane, and gamma- and alpha-HCH were in many cases below the limit of quantification (LOQ). No statistical analysis of associations with personal ...
OBJECTIVES: Since the metabolism of the organochlorine pesticide dichlorodiphenyltrichloroethane (DDT) is not fully known in humans, we evaluated if circulating levels of a major breakdown product of DDT, p,p-DDE, were related to genome-wide genetic and methylation variation in a population-based sample. METHODS: In the population-based Prospective Investigation of the Vasculature in Uppsala Seniors (PIVUS) study (1016 subjects all aged 70), circulating levels of p,p-DDE were analyzed by high-resolution chromatography coupled to high-resolution mass spectrometry (HRGC/HRMS). Genetic variants were genotyped and imputed (1000 Genomes reference, March 2012 release). Methylation sites were assayed using the Illumina HumanMethylation450 array in whole blood. A genome-wide association study (GWAS) approach was applied. RESULTS: Evidence for genome-wide significant association with p,p-DDE levels was observed only for a locus at chromosome 19 corresponding to the CYP2B6 gene (lead SNP rs7260538). Subjects
The impact of persistent organochlorine pollutants (POPs)[4], of which PCBs and DDT are two, on human fertility is still unknown and there are limited and contradictory findings so far as to whether PCBs and DDT/DDE damage human sperm. This study, which is part of a wide-ranging project known as INUENDO[5], set out also to see whether these two POPs damage sperm by altering its chromatin integrity. (Chromatin is the DNA and associated proteins that make up a chromosome ...

TY - JOUR. T1 - The organochlorine o,p-DDT plays a role in coactivator-mediated MAPK crosstalk in MCF-7 breast cancer cells. AU - Bratton, Melyssa R.. AU - Frigo, Daniel E.. AU - Chris Segar, H.. AU - Nephew, Kenneth P.. AU - McLachlan, John A.. AU - Wiese, Thomas E.. AU - Burow, Matthew E.. PY - 2012/9. Y1 - 2012/9. N2 - Background: The organochlorine dichlorodiphenyltrichloroethane (DDT), a known estrogen mimic and endocrine disruptor, has been linked to animal and human disorders. However, the detailed mechanism(s) by which DDT affects cellular physiology remains incompletely defined. Objectives: We and others have shown that DDT activates cell-signaling cascades, culminating in the activation of estrogen receptor-dependent and -independent gene expression. Here, we identify a mechanism by which DDT alters cellular signaling and gene expression, independent of the estrogen receptor. Methods: We performed quantitative polymerase chain reaction array analysis of gene expression in MCF-7 breast ...
Organochlorines are lipophylic molecules that accumulate in the fat where they remain for years. During weight loss, they are mobilized and their concentration increases in blood. The present work tests, in transgenic estrogen-reporter mice (ERE-tK-LUC), whether this increase is sufficient to modulate the estrogen receptors (ERs) in the whole body. Three weak estrogens were studied: p,p DDT [1,1,1-trichloro2,2-bis(p-chlorophenyl) ethane], p,p DDE [1,1-dichloro-2,2-bis(p-chlorophenyl)ethylene], and betaBHC [beta-benzene-hexachloride]. Dose-dependent analysis of reporter expression (luciferase) were performed in tissues of acutely treated mice. A body snap of ER activation was obtained. All these chemicals modulated the reporter, although with a different efficiency and depending upon the tissue analyzed. Induction was confirmed in the liver by determining the expression of the endogenous progesterone receptor (PR) gene, at the dose and time point at which the luciferase gene was maximally ...
We are constantly exposed to these toxins in our own lifestyles but they are also found in the soil and within the animals and animal products we eat. Aside from overly acidifying the bodys blood when these toxins are released, th accumulation of these toxins can also depress the function of thyroid which greatly affects metabolism on its own. The Physical Activity Sciences Laboratory in Canada did a study on this here:http://toxsci.oxfordjournals.org/content/67/1/46.full where it showed: Organochlorine compounds are released from body fat into the bloodstream during weight loss. Because these compounds may impair thyroid status, which is implicated in the control of resting metabolic rate (RMR), the aim of this study was to determine if the augmentation in plasma organochlorine concentrations might be associated with the decrease in serum T(3) concentration and RMR observed in response to body weight loss. Plasma organochlorine concentrations, serum T(3) concentration, and RMR were measured ...
DDT, DDE, and DDD was once widely used to control insects on agricultural crops and insects that carry diseases like malaria and typhus, but is now used in only a few countries to control malaria. Technical-grade DDT is a mixture of three forms, p,p-DDT (85%), o,p-DDT (15%), and o,o-DDT (trace amounts). All of these are white, crystalline, tasteless, and almost odorless solids. Technical grade DDT may also contain DDE (1,1-dichloro-2,2-bis(p-chlorophenyl)ethylene) and DDD (1,1-dichloro-2,2-bis(p-chlorophenyl)ethane) as contaminants. DDD was also used to kill pests, but to a far lesser extent than DDT. One form of DDD (o,p-DDD) has been used medically to treat cancer of the adrenal gland. Both DDE and DDD are breakdown products of DDT.

Chromosomal abnormalities contribute substantially to reproductive problems, but the role of environmental risk factors has received little attention.,We evaluated the association of polychlorinated biphenyl (PCB) and dichlorodiphenyldichloroethylene (p,p-DDE) exposures with sperm sex-chromosome disomy.,We conducted a cross-sectional study of 192 men from subfertile couples. We used multiprobe fluorescence in situ hybridization (FISH) for chromosomes X, Y, and 18 to determine XX, YY, XY, and total sex-chromosome disomy in sperm nuclei. Serum was analyzed for concentrations of 57 PCB congeners and p,p-DDE. Poisson regression models were used to calculate incidence rate ratios (IRRs) for disomy by exposure quartiles, controlling for demographic characteristics and semen parameters.,The median percent disomy was 0.3 for XX and YY, 0.9 for XY, and 1.6 for total sex-chromosome disomy. We observed a significant trend of increasing IRRs for increasing quartiles of p,p-DDE in XX, XY, and total ...

Mitotane (o,p′-DDD) is the standard treatment for advanced adrenocortical carcinoma (ACC). Monitoring of plasma mitotane levels is recommended to look for a therapeutic window between 14 and 20mg/L, but its positive predictive value requires optimization. We report the case of an ACC patient with a history of dyslipidemia treated with mitotane in whom several plasma mitotane levels ,30mg/L were found together with an excellent neurological tolerance. This observation led us to compare theoretical or measured o,p′-DDD and o,p′-DDE levels in a series of normolipidemic and dyslipidemic plasma samples to explore potential analytical issues responsible for an overestimation of plasma mitotane levels. We demonstrate an overestimation of mitotane measurements in dyslipidemic patients. Mitotane and o,p′-DDE measurements showed a mean 20% overestimation in hypercholesterolemic and hypertriglyceridemic plasma, compared with normolipidemic plasma. The carcinogenic potential of environmental xenoestrogens (i.e., organochlorines) is a matter of controversy. Their pathogenic role as promoters in breast cancer has been previously suggested. In Europe, despite their prohibition since the 70s, residues persist in soil and rivers resulting in a widespread contamination of the general population. In this study, we have compared the serum levels of p,p-1,1-dichloro-2, 2-bis (4-chlorophenyl) ethylene (DDE) and hexachlorobenzene (HCB) in 231 women at the time of breast cancer discovery and in 290 agematched healthy controls. p,p-DDE was found in 76.2% of cases and in 71.1% of controls but HCB was present only in 12.6% of cases (29 from 231) and in 8.9% of controls ...

The formal charge distributions in and the dipole moments of some organophosphines and arsines have been calculated, and the dipole moments of (p-chlorophenyl)dichlorophosphine (2.28 D) and (p-bromophenyl)dichlorophosphine (2.04 D) have been determined in benzene at 35° C. The differences between the observed and the calculated moments are explained in terms of dπ---pπ back-bonding and hyperconjugative effects in alkylhaloarsines. The mesomeric effects operating in the aromatic systems are evaluated by comparing the moments with those for the corresponding aliphatic systems. In unsaturated compounds the differences are attributed to mesomeric effects involving the expansion of arsenic valence shell.. ...
