In the present study, the production of dextran from date extract and sucrose as carbon sources by the bacterium Leuconostoc mesenteroides NRRL B512 (f) was investigated. In comparison to blue dextran (Mw≈2000 Da), dextran molecular weight was reduced (Mw2000 KDa). Flow behavior indices of dextran solutions from date extracts were investigated and rheological parameters were evaluated at concentrations of 1.5, 2.5, 5, together with 10% total solids at 25˚C using a Brookfield rotational viscometer as well as a cone and plate geometry. The experimental results followed the power law model for the best fit and the values of flow behavior index (n) were less than unity (0.49 .93) at all concentrations, revealing the shear thinning nature of the produced dextran. Furthermore, dependence of apparent viscosity on concentration was confirmed using the power law model.
In this study, the release of recombinant human interleukin-2 (rhlL-2) from methacrylated dextran (dex-MA) and (lactate-)hydroxyethyl methacrylated dextran (dex-(lactate-)HEMA) hydrogels with varying crosslink density was investigated. Hydrogels derived from dex-MA are stable under physiological conditions (pH 7 and 37degreesC), whereas dex-HEMA and dex-lactate-HEMA hydrogels degrade due to the presence of hydrolytically sensitive esters in the crosslinks of the gels. In this work, intramolecular and intermolecular associations under the effect of shear flow of dilute aqueous alkaline solutions of dextan, hydroxyethylcellulose (HEC), and the hydrophobically modified analogue (HM-2-HEC) in the presence of a chemical cross-linking agent were characterized with the aid of viscometry and rheo-small angle light scattering (rheo-SALS) methods. The picture that emerges at short times during the cross-linking reaction at a constant shear rate is that HEC coils contract because of intramolecular cross-linking; whereas the HM-2-HEC species show an incipient association and the dextran molecules are unaffected due to their compact structures. Calcium kinetics and its intracellular mobilization are important in all biological processes. We used verapamil to examine the effect of calcium entry blockade on microvascular transport of macromolecules in ischemia-reperfusion injury. The rat cremaster muscle was splayed, placed in a Lucite intravital chamber, and suffused with bicarbonate buffer. The clearance of fluorescein isothiocyanate-conjugated dextran (FITC-dextran 150) was measured as an index of microvascular transport. After determination of baseline data (clearance of FITC-dextran 150, 3.0 +/- 0.5 microliters/5 min/g), the muscle was made ischemic for 2 hours by clamping its vascular pedicle and subsequently was reperfused for 2 hours. Ischemia-reperfusion produced a marked increase in FITC-dextran clearance. There are several experimental models describing in vivo eosinophil (EO) migration, including ip injection of a large volume of saline (SAL) or Sephadex beads (SEP). The aim of this study was to investigate the mechanisms involved in the EO migration in these two models. Two consecutive injections of SAL given 48 hr apart, induced a selective recruitment of EO into peritoneal cavity of rats, which peaked 48 hr after the last injection. SEP, when injected ip, promoted EO accumulation in rats. The phenomenom was dose-related and peaked 48 hr after SEP injection. Pharmacological manipulation of the ventrolateral pontine reticular formation (vlPRF) of rats has an anticonvulsant effect in the maximal electroshock model of epilepsy. This study presents three anatomical experiments that determine the efferent projections from this region likely to mediate this anticonvulsant effect. We assessed material-tissue interactions with a prototypical adhesive material based on dendrimer/dextran and colon as a model tissue platform. Adhesive materials have high sensitivity to changes in their environment and can be exploited to probe and quantify the influence of even subtle modifications in tissue architecture and biology.
