Rapid destruction of hepatocytes after hepatocyte transplantation has hampered the application of this procedure clinically. The instant blood-mediated inflammatory reaction (IBMIR) is a plausible underlying cause for this cell loss. The present study was designed to evaluate the capacity of low molecular weight dextran sulfate (LMW-DS) to control these initial reactions from the innate immune system. Fresh and cryopreserved hepatocytes were tested in an in vitro whole-blood model using ABO-compatible blood. The ability to elicit IBMIR and the capacity of LMW-DS (100 mu g/ml) to attenuate the degree of activation of the cascade systems were monitored. Hemorrhagic shock was induced by removal of up to 50% blood volume over 20 min until mean arterial pressure (MAP) decreased to 35-40 mmHg. Animals were left in shock at 32°C for 60 min. Resuscitation was commenced with small boluss (0.3 ml or ~3% of shed blood) injected into the femoral vein and Lead II ECG, MAP and heart rate (HR) were monitored.. Results: 0.3 ml of 7.5% saline at resuscitation led to a drop in MAP to about 10 mmHg lasting for 4 min, increased to around 60 mmHg and then slowly declined to 40 mmHg over the next hour. No arrhythmias were detected. Curiously, 0.3 ml of 7.5% NaCl/6% Dextran-70 led to a 1.5 fold increase in the QRS amplitude, ventricular arrhythmias and cardiac instability after 10 min, and an increase in HR of 20 to 40 bpm. Although hydroxyapatite is commonly used as a scaffold for bone regeneration, sponges may be suitable because of the adaptability to the defect. To use as a scaffold, the fiber of sponge would be coated with any adhesive to storage stem cells in the sponges. Fiber in the structure of commercially available sponges was coated by immersion in dextran solution and air dried. After seeding of rat bone marrow cells (rBMCs), the sponges were implanted subcutis of rats for estimate osteogenesis in vivo. The level of osteocalcin was 25.28 ± 5.71 ng/scaffold and that of Ca was 129.20 ± 19.69 μg/scaffold. These values were significantly high- er than those in sponges without dextran coating (p < 0.01). It was thought that rBMCs could be stored on the shelf by dextran deposition in the fiber of the sponge. The goal of the present work was to determine if higher valency preparations of Rituximab could be used to simulate hyper-cross-linking events using formulations that are applicable in vivo. In the present study, the production of dextran from date extract and sucrose as carbon sources by the bacterium Leuconostoc mesenteroides NRRL B512 (f) was investigated. In comparison to blue dextran (Mw≈2000 Da), dextran molecular weight was reduced (Mw2000 KDa). Flow behavior indices of dextran solutions from date extracts were investigated and rheological parameters were evaluated at concentrations of 1.5, 2.5, 5, together with 10% total solids at 25˚C using a Brookfield rotational viscometer as well as a cone and plate geometry. The experimental results followed the power law model for the best fit and the values of flow behavior index (n) were less than unity (0.49 .93) at all concentrations, revealing the shear thinning nature of the produced dextran. Furthermore, dependence of apparent viscosity on concentration was confirmed using the power law model.
In this study, the release of recombinant human interleukin-2 (rhlL-2) from methacrylated dextran (dex-MA) and (lactate-)hydroxyethyl methacrylated dextran (dex-(lactate-)HEMA) hydrogels with varying crosslink density was investigated. We used verapamil to examine the effect of calcium entry blockade on microvascular transport of macromolecules in ischemia-reperfusion injury. The rat cremaster muscle was splayed, placed in a Lucite intravital chamber, and suffused with bicarbonate buffer. The clearance of fluorescein isothiocyanate-conjugated dextran (FITC-dextran 150) was measured as an index of microvascular transport. After determination of baseline data (clearance of FITC-dextran 150, 3.0 +/- 0.5 microliters/5 min/g), the muscle was made ischemic for 2 hours by clamping its vascular pedicle and subsequently was reperfused for 2 hours. Ischemia-reperfusion produced a marked increase in FITC-dextran clearance. After a peak of 12 +/- 2-fold increase observed in the first 15 minutes into reperfusion, FITC-dextran 150 clearance decreased in magnitude and stabilized at about sixfold above baseline. There are several experimental models describing in vivo eosinophil (EO) migration, including ip injection of a large volume of saline (SAL) or Sephadex beads (SEP). The aim of this study was to investigate the mechanisms involved in the EO migration in these two models. Two consecutive injections of SAL given 48 hr apart, induced a selective recruitment of EO into peritoneal cavity of rats, which peaked 48 hr after the last injection. SEP, when injected ip, promoted EO accumulation in rats. The phenomenom was dose-related and peaked 48 hr after SEP injection. To investigate the mediators involved in this process we showed that BW A4C, MK 886 and dexamethasone (DXA) inhibited the EO migration induced by SAL and SEP.
