Genetic diversity and genomic rearrangements are a driving force in bacterial evolution and niche adaptation. We sequenced and annotated the genome of Lactobacillus johnsonii DPC6026, a strain isolated from the porcine intestinal tract. Although the genome of DPC6026 is similar in size (1.97mbp) and GC content (34.8%) to the sequenced human isolate L. johnsonii NCC 533, a large symmetrical inversion of approximately 750 kb differentiated the two strains. Comparative analysis among 12 other strains of L. johnsonii including 8 porcine, 3 human and 1 poultry isolate indicated that the genome architecture found in DPC6026 is more common within the species than that of NCC 533. Most of the Dictyostelium ribosomal nontranscribed spacer DNA has been mapped using Eco R1, Hind III, and Sal I restriction digests and recombinant plasmids containing parts of the rDNA (ribosomal DNA). The repeat units are at least 42 kb (1 kilobase = 1000 base pairs = 1 kb) long; of this, 8 kb codes for the 36S rRNA precursor. The repeat units give a homogeneous pattern of Sal I, Eco R1, and Hind III restriction fragments. The repeats could be linked by a heterogeneous region in the nontranscribed spacer, not tandemly repeated, or linked in a more complicated structural arrangement. The total rDNA comprises at least 18% of the nuclear DNA.
In hopes of protecting springflows and future water supplies, the Edwards Aquifer Authority declared Stage III restrictions Wednesday for the first time, as the pool below San Antonio continues to drop. On June 13, when the EAA declared Stage II restrictions, all industrial, municipal and agricultural pumping permits that tap the Edwards below San Antonio were reduced by 30 percent of their authorized amounts. The EAA made the declaration because the 10-day rolling average of the J-17 monitoring well at Fort Sam Houston dropped below 640 feet above sea level. The move is part of the EAAs management plan to assure the San Marcos and Comal springs keep flowing and that water from the aquifer will be available next year for use if the current two-year drought continues.

DNA Restriction-Modification Enzymes: Systems consisting of two enzymes, a modification methylase and a restriction endonuclease. They are closely related in their specificity and protect the DNA of a given bacterial species. The methylase adds methyl groups to adenine or cytosine residues in the same target sequence that constitutes the restriction enzyme binding site. The methylation renders the target site resistant to restriction, thereby protecting DNA against cleavage.

Chloramphenicol acetyltransferase (or CAT) is a bacterial enzyme (EC 2.3.1.28) that detoxifies the antibiotic chloramphenicol and is responsible for chloramphenicol resistance in bacteria. This enzyme covalently attaches an acetyl group from acetyl-CoA to chloramphenicol, which prevents chloramphenicol from binding to ribosomes.
