Size-resolved aerosol samples for subsequent quantitative determination of polymer sugars (polysaccharides) after hydrolysis to their subunit monomers (monosaccharides) were collected in surface air over the central Arctic Ocean during the biologically most active summer period. The analysis was carried out by novel use of liquid chromatography coupled with highly selective and sensitive tandem mass spectrometry. Polysaccharides were detected in particle sizes ranging from 0.035 to 10 mu m in diameter with distinct features of heteropolysaccharides, enriched in xylose, glucose + mannose as well as a substantial fraction of deoxysugars. Polysaccharides, containing deoxysugar monomers, showed a bimodal size structure with about 70% of their mass found in the Aitken mode over the pack ice area. This study examined the dopamine-induced astroglial protective function through the activation of the pentose-phosphate pathway (PPP) to reduce reactive oxygen species (ROS). In vitro experiments were performed using striatal neurons and cortical or striatal astroglia prepared from Sprague-Dawley rats or C57BL/6 mice. Insulin binding and 2-deoxy-d-glucose uptake were compared in the soleus, plantaris, and extensor digitorum longus (EDL) muscles, in vitro, since the known biochemical profile of the soleus differs markedly from the other two. The present study showed that insulin binding increased in all three muscles with increasing concentrations of insulin in the range of 0.2- 30 nM. However, the increase in binding of insulin to soleus at each insulin concentration exceeded that observed in the other two muscles (P , 0.05). Differences between the plantaris and EDL were not significant. The quantity of insulin bound at each concentration also increased more rapidly in the soleus than in either the plantaris (P , 0.05) or EDL (P , 0.05). Basal and insulin-stimulated uptake of 2-deoxy-d-glucose was also greater in the soleus than in the other muscles (P , 0.05). We studied the uptake of 2-deoxy-D-glucose (2DG) and the synthesis of its phosphorylated product 2DG-6-phosphate (2DG-6P) by the retinas of the clawed frog (Xenopus laevis) and the bullfrog (Rana catesbeiana). Autoradiographs showed that most of the retinal 2DG uptake is by the photoreceptor layer. The 2DG accumulation by isolated Xenopus retinas was time and concentration dependent. The Kt for transport was 5.05 mM; Vmax was 6.99 X 10(-10) mol . mg-1 tissue wet weight min-1. The Km for 2DG-6P formation was estimated to be 2-3 mM and Vmax to be approximately 4 x 10(-9) mol . mg-1 min-1. 2DG uptake was inhibited competitively by glucose with a Ki of 2.29 mM. Exposure to light reduced 2DG uptake by no more than 10% as compared with dark uptake. BACKGROUND: Hypoxic pulmonary vasoconstriction (HPV) is a defense mechanism to maintain adequate oxygenation. It has been reported that metabolism inhibition augments HPV. The purpose of the present study was, therefore, to determine the effect of metabolism inhibition on HPV in a rabbit model of isolated lung perfusion with exclusion of the influential factors on HPV. METHODS: In adult rabbits, lungs were isolated and perfused with a constant pulmonary perfusate flow. Acid-base status and temperature of perfusate was also constantly maintained. Thirty minutes after, the baseline hypoxic pressor response (HPR) was measured as the difference of pulmonary artery pressure (PAP) between a period of 21% normoxic gas inhalation and that of 3% hypoxic gas inhalation. Background Optical imaging (OI) techniques such as bioluminescence and fluorescence imaging have been widely used to track diseases in a non-invasive manner within living subjects. These techniques generally require bioluminescent and fluorescent probes. Here we demonstrate the feasibility of using radioactive probes for in vivo molecular OI. Methodology/Principal Findings By taking the advantages of low energy window of light (1.2-3.1 eV, 400-1000 nm) resulting from radiation, radionuclides that emit charged particles such as β+ and β− can be successfully imaged with an OI instrument. In vivo optical images can be obtained for several radioactive probes including 2-deoxy-2-[18F]fluoro-D-glucose ([18F]FDG), Na18F, Na131I, 90YCl3 and a 90Y labeled peptide that specifically target tumors. Conclusions/Significance These studies demonstrate generalizability of radioactive OI technique.
