Dacryoadenitis is infection of the lacrimal gland results in diffuse homogeneous enlargement, which can sometimes compress the globe. Most common organisms are Staphylococcus aureus, mumps, infectious mononucleosis, and influenza virus.
Pathophysiology of canaliculitis. As has been mentioned above, canaliculitis occurs due to infection of the canaliculi. This infection is often brought on by bacteria though there could be alternative aetiology such as fungi and viruses as well. The most common cause of canaliculitis is Actinomyces Israelii. However, the other causes can include aspergillus, Canada albicans and even herpes simplex and herpes zoster virus.. Infection by these organisms results in the formation of tiny stones that are rich in sulphur. These stones form little pockets that unfortunately are not affected by the antimicrobial properties of tears.. History and examination. ...

Purpose. To investigate the microstructure of the lacrimal canaliculus and the characteristics of lacrimal canalicular diseases by 80-MHz ultrasound biomicroscopy (UBM).. Methods. This study included 33 participants: 20 normal subjects (40 eyes), 2 patients with chronic lacrimal canaliculitis (4 eyes), 10 patients with chronic dacryocystitis (16 eyes), and 1 patient with lacrimal punctum atresia (2 eyes). All participants underwent 80-MHz UBM; disease-specific features were noted.. Results. On 80-MHz UBM of the lacrimal canaliculi (vertical section) in normal subjects, low echo of the lacrimal canalicular lumen and high echo of the lacrimal canalicular wall were observed. The uniform low echo near the wall was the mucosal epithelium. The outermost layer of medium-to-high echo was the subepithelial elastic fibrous layer. In the horizontal section, the lumen was continuous. Two linear high echoes parallel to the canalicular wall could be observed at the center of the lacrimal canaliculus, which ...

We detected serum autoantibodies in healthy and diseased NOD mice targeted against membrane-bound proteins found in the lacrimal glands of mice, rat, and rabbits. As disease in the NOD mouse model progressed with age, we observed an increase in autoantibody titer as well as a shift in IgG isotypes from non-complement-fixing (IgG1, IgG3) to complement-fixing (IgG2A, IgG2B) isotypes. This shift appeared to be age and gender dependent: female NOD mice had higher titers of the complement-fixing immunoglobulins, IgG2A and IgG2B. It is regularly reported that male NOD mice develop more severe dacryoadenitis than females, whereas female mice develop more severe disease in other organs.; Flow cytometry showed that the ratios of CD4+ T cells to CD8+ T cells were 2:1 in the lacrimal glands of both male and female animals with disease. However, within the CD4+ cell populations, the female mice appeared to have a larger subpopulation of CD4+CD25+ FoxP3 expressing cells. These results suggest that ...

Acute Myeloid Leukemia (AML) is a heterogeneous group of malignant hemopathies characterized by clonal proliferation and medullary accumulation of myeloid precursors which frequently results in hematopoietic insufficiency with or without leukocytosis. The World Health Organization classification considers that patients with a blasts percentage ,20% in bone marrow and / or peripheral blood have acute leukemia (1).. The incidence of AML has been estimated at around 30 cases per million inhabitants per year with a median age at diagnosis around 67 (2). During the past four decades AML treatment has been based on the combination of an anthracycline (daunorubicin or idarubicin) and cytarabine, the 3+7. During this time there have been numerous and unsuccessful attempts to improve the 3+7. A better understanding of the AML biology and treatment resistant mechanisms have allowed new agents to develop.. During this abstract and presentation, we will discuss FLT3 mutated AML and FLT3 inhibitors.. The ...

Numerous eye complications are associated with that relating to diabetes mellitus, of which the dry eyes Disease is one of the most typical as a result of improvement in the tear proteins from that relating to the healthy people .Diabetes could damage certain nerves in your body. In the eyes, such damage can block the device that controls tear secretion. When this occurs, the lacrimal glands fail to produce sufficient tears, resulting in dry eyes. Insulin deficiency is another symptom associated with diabetes. Aside from controlling blood sugar, insulin comes with a major effect, on several glands in your body. In the eyes, lacrimal gland metabolism is affected by insulin. If you find low insulin in your body, the biomechanical balance of the eyes is disrupted resulting in ocular dryness. Another reaction of diabetes is lacrimal gland inflammation thats on account of abnormal lacrimal secretion. When this gland is inflamed, tear secretion is affected, which leads to dry eyes. Remedial ...
In this study, we have demonstrated that Aire-deficient mice represent a model for autoimmune dry eye and have further characterized the autoimmune response. Of note, previous work in the model has suggested that the Aire-deficient autoimmune response to the salivary and lacrimal glands is directed against the known autoantigen α-fodrin (7). To carefully assess the Ags potentially targeted in the model, we performed immunoblot analysis with sera from multiple NOD and BALB/c Aire-deficient mice on lacrimal gland extracts and identified a novel molecular target of 18 kDa that proved to be OBP1a. Interestingly, OBP1a is expressed in the thymus in an Aire-dependent fashion, suggesting that defective thymic expression of OBP1a may play a role in the spontaneous dacryoadenitis that develops in the Aire-deficient mouse model.. Previous work on the Aire-deficient model has suggested that α-fodrin is a likely prominent autoantigen associated with SS; however, our data provides evidence for other ...

Dacryocystocele or timo cyst is a benign, bluish-gray mass in the inferomedial canthus that forms as a result of a narrowing or obstruction of the nasolacrimal duct, usually during prenatal development. The prevalence of dacryocystocele is 1 in 3884 live births. Complications like swelling, watery eyes and infection might occur. While usually filled with sterile mucus, dacryocystoceles occasionally become infected. The nasolacrimal ducts drain the excess tears from our eyes into the nasal cavity. In dacryocystocele there this tube gets blocked on either end and as a result when mucoid fluid collects in the intermediate patent section it forms a cystic structure. The diagnosis can be made prenatally; routine obstetric ultrasound can identify the characteristic hypoechoic lesion inferior and medial to the globe. It is important to distinguish a dacrocystocele from the more serious encephalocele, which is a neural tube defect. A dacryocystocele can be diagnosed postpartum with a non-invasive ...

The purpose of this research was to understand the mechanisms by which regulatory T cells (Treg) prevent the induction of autoimmune disease. First, we wished to elucidate whether regulatory T cells were present in day 3 thymectomized (d3tx) mice, which develop strainand organ-specific autoimmune disease. It was thought that a paucity of Treg secondary to their late ontogeny relative to effector T cells (Teff) was central to disease induction. Contrary to this paradigm, we found that functional Treg were present in d3tx mice as well as normal 3 day old mice. Importantly, they dampened the severity of autoimmune ovarian disease (AOD) and dacryoadenitis (DA) in the endogenous host. Treg present in d3tx mice were distributed such that Treg that suppress AOD were preferentially found in the ovarian draining lymph node (LN). This diseasespecific accumulation could belie a mechanism by which Treg suppress disease in a normal individual, but could also reflect the active autoimmune disease in the host. ...

Dacryocystitis is an infection of the lacrimal sac, secondary to obstruction of the nasolacrimal duct at the junction of lacrimal sac. The term derives from the Greek dákryon (tear), cysta (sac), and -itis (inflammation). It causes pain, redness, and swelling over the inner aspect of the lower eyelid and epiphora. When nasolacrimal duct obstruction is secondary to a congenital barrier it is referred to as dacrocystocele. It is most commonly caused by Staphylococcus aureus and Streptococcus pneumoniae. The most common complication is corneal ulceration, frequently in association with S. pneumoniae. The mainstays of treatment are oral antibiotics, warm compresses, and relief of nasolacrimal duct obstruction by dacryocystorhinostomy. ...

Purpose: Sox11, a transcription factor expressed in trigeminal and dorsal root ganglion neurons early in development, is induced in adult sensory neurons following nerve injury. As a promotor of nerve regeneration, we hypothesized that Sox11 may also be expressed during dry eye in order to increase trophic support to the cornea provided by corneal afferent neurons. The present study examined Sox11 expression and examined corneal epithelial integrity and mechanical sensitivity in Sox11 conditional knockout animals following lacrimal gland excision.. Methods: Male C57BL/6 mice were obtained from Jackson Labs and Sox11 conditional knockout (Sox11CKO) mice were generated using Cre-LoxP-mediated recombination under the control of a NaV1.8 promoter. Under isoflurane, unilateral lacrimal gland excision was performed. Either the left exorbital lacrimal gland, or both the exorbital and intraorbital lacrimal glands were excised. For sham surgeries, incisions were made and glands were partially exposed on ...

1. Segments of mouse pancreatic or exorbital lacrimal gland were superfused with saline solutions. Under visual control two micro-electrodes were inserted into neighbouring cells within the same acinus or into neighbouring acini. Cell to cell electrical coupling was assessed by injecting rectangular current pulses through one electrode and measuring the electrotonic potential change in the same cell (V(1)) and in the neighbouring cell (V(2)). Acetylcholine (ACh) was added locally to impaled acini by micro-ionophoresis from an extracellular micropipette.2. Exposure of the tissues to a Krebs solution equilibrated with 100% CO(2) caused a rapid increase in the size of electrotonic potential changes in the current injection cell and disappearance of the electrotonic potential changes in a neighbouring acinus or cell. This electrical uncoupling of previously coupled cells was rapidly reversible upon return to a solution equilibrated with 95% O(2) and 5% CO(2).3. Reduction of electrical intercellular ...
