The intervention arm of the POBASCAM trial provided us with an opportunity to assess the clinical performance of triage strategies for hrHPV-positive women, in a large screening population followed for 4 years within one screening round. Ten triage strategies were evaluated, and most of these met the thresholds for NPV and PPV of 98% and 20%, respectively. The three strategies that showed the best balance between the safety of a strategy (NPV), and the burden of screening on patients and clinicians (PPV and referral rate), were (i) cytology and HPV16/18 genotyping at baseline without repeat testing, (ii) cytology at baseline with repeat cytology testing after 6 months, and (iii) cytology and HPV16/18 genotyping at baseline followed by repeat cytology examination at 6 months.. Im afraid too that immunostaining for CDs is extremely difficult if not impossible, we make a lot of experiments in determining optimal fixation procedure for immunodetection of CDs and other antigens in human cytological specimens and we found as some other investigators that drying of the cells before fixation is detrimental for many antigens especially for surface antigens! it is true that you can detect some robust antigens like CK even in smears previously stained with Giemsa, but this is exception.. Maybe you should try with microwave pretreatment or you can ask clinicians to use spray fixative for fixing the smear or to prepare you the cells suspension.. hope this helps ...

The examination of brush cytology specimens has become an established diagnostic technique in the investigation of patients with suspected pancreatic, bile duct, gallbladder, and ampullary tumours. We have reviewed 448 consecutive brush samples obtained from 406 patients and correlated the findings with pathological and clinical outcomes. To our knowledge, this is the largest series of pancreatico-biliary brush cytology specimens yet reported.. As with previous studies, we found that the brush cytology technique produced cellular samples of good quality in most instances. Only 26 of 489 (5.3%) specimens were considered inadequate for diagnosis. The conventional technique of impression cytology provides a non-invasive method for the evaluation of conjunctival epithelium alterations. Using indirect immunofluorescence procedures two inflammatory markers, class II MHC antigens HLA DR and the receptor to IgE (CD23), were sought in impression cytology specimens obtained from 80 patients. In normal subjects conjunctival epithelial cells did not show any reactivity. Only scattered dendritic cells were found to express class II antigens but not the receptor to IgE. In contrast patients with chronic conjunctivitis of various aetiologies, mainly infectious or allergic, had 40-100% of brightly positive conjunctival cells for one or both antigens. In these cases epithelial cells and goblet cells reacted similarly. Fortunately, this error was only in the manuscript and not in our draft report the USPSTF used for its deliberations. The error was introduced when we omitted proportional cancer results from tables as we were editing to fit within space constraints for dissemination products, including the manuscript.
