Cryptands are a family of synthetic bi- and polycyclic multidentate ligands for a variety of cations. The Nobel Prize for Chemistry in 1987 was given to Donald J. Cram, Jean-Marie Lehn, and Charles J. Pedersen for their efforts in discovering and determining uses of cryptands and crown ethers, thus launching the now flourishing field of supramolecular chemistry. The term cryptand implies that this ligand binds substrates in a crypt, interring the guest as in a burial. These molecules are three-dimensional analogues of crown ethers but are more selective and strong as complexes for the guest ions. The resulting complexes are lipophilic. The most common and most important cryptand is N[CH2CH2OCH2CH2OCH2CH2]3N; the formal IUPAC name for this compound is 1,10-diaza-4,7,13,16,21,24-hexaoxabicyclo[8.8.8]hexacosane. The focus of this thesis is the incorporation of the 1,2-bis(pyridinium)ethane motif into new [2]rotaxane ligands for the purpose of developing new metal organic rotaxane frameworks (MORFs). Both new stoppers that can act as ligands as well as new crown ethers with appended donors were synthesized and these components combined to create new [2]rotaxane ligands and eventually new MORFs. Chapter 2, looks at the synthesis and coordination chemistry of a [2]rotaxane containing terpyridine stoppers. The [2]rotaxane was shown to form mixed ligand complexes with RuII, AgI, ZnII, and PdII. The complexes were studied in solution by NMR, and UV/vis spectroscopy and mass spectrometry and in the solid state by X-ray crystallography. This invention describes a new concept of flexible template-directed microporous partially pyrolyzed polymeric membranes which have greatly improved performance in separation of gas pairs compared to their precursor polymeric membranes. Organic hosts, such as crown ethers, cyclodextrins (CDs), calixarenes (CXs), and spherands, or polymeric additives, such as poly(ethylene glycol) (PEG) and polyvinylpyrrolidone (PVP) were used as the micropore-forming templates. Micropore-forming template/polymer blend membranes comprising organic micropore-forming templates embedded in a polymer matrix were prepared by dissolving the organic micropore-forming templates in the polymer solution followed by solution-casting and solvent evaporation or solvent exchange. Low-temperature selectively pyrolyzing micropore-forming templates in the micropore-forming template/polymer blend membranes at a nitrogen flow resulted in the formation of flexible microporous partially pyrolyzed polymeric membranes.

This dissertation focuses on studies of mechanically interlocked molecules (MIMs) specifically [2]rotaxanes in three principle areas: (1) creating [2]rotaxane linkers to incorporate into metal-organic frameworks (MOFs); (2) studying the rate of shuttling motion in solution and finally (3) investigating the shuttling motion inside the MOF. Chapter 1 provides a brief introduction to MIMs, rotaxanes, MOFs and all previous studies on dynamic motions of MIMs in MOFs. Chapter 2 describes a [2]rotaxane linker with donor atoms attached to both the axle and the wheel. The linker contains four carboxylate groups attached to a rigid, H-shaped axle and two carboxylate units appended to a crown ether wheel. In the resulting Zn-based MOF, three independent 3-periodic frameworks (threefold interpenetration) are interconnected only by virtue of the threading of their individual components in the rotaxane linker. The enzyme Purple Acid Phosphatase (PAP) is an important target for the development of new anti-osteoporotic drug leads. The major goal of this thesis is to better emulate the synergy that occurs between the primary and secondary coordination sphere within the active site of phosphatase enzymes, such as PAP. This was perceived by development of synthetic methods for new biomimetics, i.e. low-molecular weight metal complexes. Therefore, asymmetric dinucleating ligands which mimic the peptide backbone of the active site of PAP including the ability to form hydrogen bonds with a phosphoester substrate or a nucleophile were designed and synthesized. Using these ligands, more accurate model systems for the enzyme active site were achievable as they combine the two essential structural features known to influence the catalytic activity towards the hydrolysis of phosphoesters, i.e. the asymmetry of the dinuclear active site and the capacity for hydrogen bond formation. Kinetics of Cr(III) ions transport through a bulk liquid membrane containing dinonylnaphthalenesulfonic acid (DNNSA) as a carrier, flowing over aqueous phases, has been examined. Special attention has been paid to the effect of the membranes velocity flow on the chromium concentration decrease in a feed phase. For the description of relationships of chromium(III) concentration in particular phases with the time, a model based on the assumption of consecutive first-order reactions was proposed. Satisfactory compatibility of experiments and model results have been obtained both for the membrane flow velocities below 0.0034 m·s-1 when the interfaces begin to fluctuate slightly and for low initial Cr(III) concentration in the feed phase.

The synthesis of a peptide-containing lasso molecular switch by a self-entanglement strategy is described. The interlocked [1] rotaxane molecular machine consists of a benzometaphenylene[25]crown-8 (BMP25C8) macrocycle surrounding a molecular axle. This molecular axle contains a tripeptidic sequence and two molecular stations: a N-benzyltriazolium and a pH-sensitive anilinium station. The tripeptide is located between the macrocycle and the triazolium station, so that its conformation can be tailored depending on the shuttling of the macrocycle from one station to the other. At acidic pH, the macrocycle resides around the anilinium moiety, whereas it shuttles around the triazolium station after deprotonation. This molecular machinery thus forces the lasso to adopt a tightened or a loosened conformation.

Physiologic concentration in amniotic fluid (AF) of several metabolites has not been established with certainty. In this study, we initially assayed purines, pyrimidines, and amino compounds in 1,257 AF withdrawn between the 15th and the 20th week of gestation from actually normal pregnancies (normal gestations, normal offspring). Results allowed to determine physiologic reference intervals for 45 compounds. In these AF, not all purines and pyrimidines were detectable and uric acid (238.35 +/- A 76.31 mu mol/l) had the highest concentration. All amino compounds were measurable, with alanine having the highest concentration (401.10 +/- A 88.47 mu mol/l). In the second part of the study, we performed a blind metabolic screening of AF to evaluate the utility of this biochemical analysis as an additional test in amniocenteses. Hydrogen bonding underpins the properties of a vast array of systems spanning a wide variety of scientific fields. From the elegance of base pair interactions in DNA to the symmetry of extended supramolecular assemblies, hydrogen bonds play an essential role in directing intermolecular forces. Yet fundamental aspects of the hydrogen bond continue to be vigorously debated. Here we use dynamic force microscopy (DFM) to quantitatively map the tip-sample force field for naphthalene tetracarboxylic diimide molecules hydrogen-bonded in two-dimensional assemblies. A comparison of experimental images and force spectra with their simulated counterparts shows that intermolecular contrast arises from repulsive tip-sample interactions whose interpretation can be aided via an examination of charge density depletion across the molecular system. For 3.10b the sirglet of the phenyl protons at 7.15 ppm (in the absence of shift reagent) was monitored.
