Engineered Auxotrophy Auxotrophy is the inability of an organism to synthesize a particular organic compound required for its growth. In synthetic biology this is used to create an organism that cannot grow when it leaves the lab or a production facility, because it would be unlikely to encounter the necessary metabolite in nature. This has been a strategy in research for over 70 years, made famous in Beadle and Tatums Nobel prize-winning work on the one gene-one enzyme hypothesis. Examples of engineered auxotrophy 1991 Conditional-Suicide Containment System for Bacteria Which Mineralize Aromatics - Here they have a Pseudomonas that degrades benzoates for bioremediation, and then dies when substrate is no longer present [5]. 2003 Biological containment of genetically modified Lactococcus lactis for intestinal delivery of human interleukin 10 - Here they have engineered a Lactococcus that produces il-10 for the treatment of Crohns disease, this strain requires thymidine for full viability as it ...

Unaffordable healthcare. Unsustainable fuels and industrial chemicals. Environmental pollution. Lack of sufficient food supplies for a rapidly growing population.. There is one thing each of these pressing, real-world problems has in common: their potential solution.. Synthetic biology, a novel branch of science that enables the design and construction of biological parts, devices, and systems as well as the redesign of natural biological systems, is beginning to emerge with innovative solutions to these global challenges. But technology has always raised concerns about safety, including at the dawn of the biotech revolution when numerous naysayers raised fears of dire consequences that never materialized.. Synthetic biology is not proving immune to this common challenge. Because this emerging discipline works with the most elemental properties of life, it raises serious questions about who is doing the work, under what conditions, and under what supervision?. Regulation is one way to foster ...

In contrast to Risk Groups, Biosafety Levels (BSL) prescribe procedures and levels of containment for the particular microorganism or material (including Research Involving Recombinant or Synthetic Nucleic Acid Molecules). Similar to Risk Groups, BSL are graded from 1 - 4. Detailed descriptions of containment practices and biosafety levels can be found in the CDC-NIH Guidelines Biosafety in Microbiological and Biomedical Laboratories.. The majority of work at UVA involves Biosafety Level 2 (BSL-2) practices. BSL-2 containment and practice is suitable for work with agents that are infectious to humans or animals where exposure may result in limited to moderate disease. The routes of exposure to these agents are typically through cuts and breaks in the skin, ingestion, and splashes to the mucous membranes (eyes, nose, and mouth). These agents or materials include:. ...

A pump housing that contains a pump that draws fuel from an underground storage tank containing fuel to deliver to fuel dispensers in a service station environment. The pump is coupled to a double-walled fuel pipe that carries the fuel from the pump to the fuel dispensers. The double-walled fuel piping contains an inner annular space that carries the fuel and an outer annular space that captures any leaked fuel from the inner annular space. The outer annular space is maintained through the fuel piping from the pump to the fuel dispensers so that the outer annular space can be pressurized by a pump to determine if a leak exists in the outer annular space or so that fuel leaked from the inner annular space can be captured by a leak containment chamber in the pump housing.

A medical cost containment system for ensuring that the anticipated cost savings from utilization review (UR) agreements are actually realized. The UR agreements are essentially contractual agreements that specify the type and quantity of medical treatments relating to a specific claim resulting from a specific injury. Each UR agreement comprises a claim number, a procedure code describing the particular medical service authorized, and some indication as to dates or quantity of service authorized. All of the UR agreements are stored in a UR database. When a medical bill is received by the insurance company, the bill is entered in to the computer. The cost containment system searches all UR agreements in the UR database which have the same claim number as the claim number on the bill. For each item in the medical bill, the system finds the UR agreement which most closely matches the procedure code in the line item and various other criteria, such as the dates of treatment. The system then checks to

In the GM microbe, the lethal functions associated with the GM trait are not active. However, if this trait is transferred to a different microbe, the associated lethal functions become active and the recipient microbe dies (gene containment). Lethal functions can also be activated in the GM microbe upon completion of the biotechnological process by a specific signal from the environment, such as root exudates, temperature, or toxic pollutants (biological containment ...
Previously known as Ebola hemorrhagic fever because of the internal and external bleeding that can occur, the illness has an incubation period between exposure and the onset of clinical illness that ranges from 2-21 days. Humans are not infectious until they develop symptoms which typically mimic a flu-like illness with the sudden onset of fever, fatigue, muscle pain, headache and sore throat. This is followed by vomiting, diarrhea, rash, impaired kidney and liver function, and in some cases bleeding. Leukopenia (low white blood cell counts), thrombocytopenia (low platelet counts), and elevated liver enzymes can be seen. A definitive diagnosis can be made using specialized testing of blood samples (e.g. ELISA, RT-PCR, electron microscopy or cell culture) under maximum biological containment conditions ...

Research with highly pathogenic infectious agents is limited to the very few laboratories worldwide with the required biosafety levels (level 4), none of which are found in Spain. "Greater investment in such facilities is required in order to be able to adequately respond to threats that will almost certainly appear in the future", noted Rafael Delgado, a microbiologist from the Hospital Universitario 12 de Octubre Research Institute, during his presentation as part of the scientific seminar organised today by the Biomedical Research Institute (IBI) in the framework of the BIOCAPS project.. Experts in various infectious diseases met today in the AFundación building in Vigo to discuss the latest progress related to the prevention, diagnosis, treatment and development of vaccines against infectious diseases, with a special emphasis on recent events.. This is the case of Ebola, for example, the virus for which was responsible for the latest pandemic risk only a few months previously. With regard ...
Four Biosafety Levels (BSLs) are described which consist of combinations of laboratory practices and techniques, safety equipment, and laboratory facilities. Each combination is specifically appropriate for the operations performed, the documented or suspected routes of transmission of the infectious agents, and for the laboratory function or activity.. The recommended biosafety level(s) for the organisms in Section VII (Agent Summary Statements) represent those conditions under which the agent can ordinarily be safely handled. The laboratory director is specifically and primarily responsible for assessing risks and for appropriately applying the recommended Biosafety Levels. Generally, work with known agents should be conducted at the biosafety level recommended in Section VII. When specific information is available to suggest that virulence, pathogenicity, antibiotic resistance patterns, vaccine and treatment availability, or other factors are significantly altered, more (or less) stringent ...

BSL-3: Processing human or animal material in endemic zones or in non-endemic areas in emergency circumstances; stringent aerosol containment practices, autoclaving waste, decontamination of work areas, and control of egress of material from the laboratory. Other cultures, cells, or similar biological material that could potentially harbor RVFV should not be used in a RVFV laboratory and subsequently removed. Diagnostic or research studies outside endemic areas should be performed in a BSL-3 laboratory. Personnel also must have additional respiratory protection (such as a PAPR) or be vaccinated for RVFV. In addition, for research conducted in non-endemic areas, the USDA may require full BSL-3Ag containment. The USDA may require enhanced ABSL-3 or ABSL-3 facilities and practices for working with RVFV in the United States. ...

Computational or mathematical modeling work is perhaps not the most glamorous of scientific jobs, and is certainly not the most visible. Whenever emerging diseases, epidemics and other events associated with public health are concerned in real life, people tend to think in terms of direct action - physicians and laboratory personnel for diagnosis and treatment, public health officials for strategy decisions related to prevention and interventions, and even lawmakers for official promulgation of policy and provision of funding for relevant activities. However, a nations state of preparedness for potential public health disasters, such as an infectious disease outbreak, depends on a lot more - much of which is required to be done in the background, without fanfare.. This is where bioinformaticians and computational scientists come in; their role is to: (a) aggregate knowledge from various fields, such as medicine, microbiology, and immunology; (b) collect and gather data from epidemiological and ...

The stratigraphic survey which was conducted in October 2015 identified a particular area of interest and identified a number of sub surface anomalies that were considered worthy of further investigation. These were further investigated by a test excavation in November/December 2016 and in January/February 2017. Test trenches were dug revealing two large structures. One structure appears to be a large sewage containment system or septic tank that had been decommissioned and filled with rubble and debris and then covered with top soil. The second structure is a long structure which is divided into 20 chambers. The Commission has not yet determined what the purpose of this structure was but it appears to be related to the treatment/containment of sewage and/or waste water. The Commission has also not yet determined if it was ever used for this purpose. ...

When an investigator applies to an agency for funds to support work on recombinant DNA molecules, the statement certifying the physical containment rating would be appended to the grant proposal. The group reviewing the research proposal would determine whether the certified level of physical containment was adequate for the real or potential biohazard associated with the proposed experiments. Entering into the assessment of adequacy would be the type of DNA to be cloned, the scale of the proposed bacterial cultures to be used and most importantly the adequacy of the biologic barriers to be employed. If the reviewing group concludes that the experimental design and physical containment rating is commensurate with their and the investigators estimate of the risk, the grant would be processed for scientific merit in the usual fashion. If there is some question of the adequacy of the containment capability, the experimental design or the investigators estimate of the risks, the matter would be ...

Air Master Systems Flammable Liquid Storage Cabinet is a "cabinet-within-a-cabinet" design with fully welded 18ga interior and exterior units. Both cabinets are completely powder coated inside and out, offering greater protection against corrosion than the standard double panel construction. This design creates 1.5" of airspace on all four sides as well as top and bottom for heat resistance up to 2400 degrees F.. The interior of the cabinet contains one fully adjustable shelf and a 2" deep removable drip pan to capture any spills or leaks inside the cabinet. Exterior depth of the cabinet is 18" and interior depth is 14". A 2" x 2" 12ga support angle is shipped with each cabinet for countertop installation.. Doors have a continuous hinge and lever type handles with a hidden 3-point lock mechanism. If a self-closing feature is required, a hydraulic closure will be attached to the doors and a custom closing system attached to the interior cabinet. The back of the cabinet has two venting holes that ...

Methods of improving the pressure containment integrity of subterranean well bores are provided. The methods include pumping a fracture sealing composition into the well bore that rapidly converts into a high friction pressure sealing composition which is impermeable, deformable, extremely viscous and does not bond to the faces of fractures. Thereafter, the fracture sealing composition is squeezed into one or more natural fractures or into one or more new fractures formed in the well bore to thereby increase the pressure containment integrity of the well bore. The methods also include the prediction of the expected increase in pressure containment integrity.

Epidemic model looked at 3 contributing factors. Three waves of the deadliest influenza pandemic in history, known as the Spanish flu, hit England and Wales in 1918, just as World War 1 was coming to an end.. Why flu arrives in multiple waves like this is the focus of a study by McMaster University researchers who discovered three contributing factors: the closing and opening of schools, temperature changes and â most importantly â changes in human behavior.. "We found all three factors were important in 1918 but that behavioral responses had the largest effect," said David Earn, an investigator with the Michael G. DeGroote Institute for Infectious Disease Research, and a professor in McMasters Department of Mathematics and Statistics.. The study appears in the July 10 online issue of the Proceedings of the Royal Society.. The researchers did not measure behavioral change directly; instead, their model showed that the three waves could only be explained if people reduced infectious contact ...
