Confocal microscopy and in situ hybridization procedures have led to enormous progress in the visualization of the spatial organization and dynamics of transcription and RNA‐processing machinery in eukaryotic cells. It is well established that the nucleus is a highly organized structure composed of many different territories, subdomains and organelles (Lamond and Earnshaw, 1998). A state‐of‐the‐art account of the nuclear architecture of plant cells was given by P.Shaw (John Innes Centre, Norwich, UK). Mapping of the RNA polymerase I transcription and rRNA processing sites, using different pre‐rRNA, snoRNA and nucleolar protein probes, indicates that the plant cell nucleolus is organized as a series of concentric layers in which transcription and successive rRNA processing reactions occur. Individuals with intellectual disability (ID) often engage in behaviours that are labelled as challenging (challenging behaviours; CB). CBs are actions that may place the individual at risk of harm or exclusion, or may place other people (e.g. carers) at risk of harm [1]. Thus, CBs are defined socially, in terms of their impact. Behaviours labelled as challenging typically include actions such as anger and aggression, self-injurious behaviours (e.g. self-biting, hitting body parts against objects, scratching and gouging), destruction of property, and inappropriate or risky social behaviour. CBs are, by definition, a significant challenge for services and impact negatively on the quality of life of people with ID. CBs are also related to risk of abusive practices (cf. This work combines (i) an siRNA‐based biochemical analysis of RNA and protein constituents of the tri‐snRNP in vivo with (ii) the use of fluorescence microscopy to locate these constituents in the nucleus. Our initial finding is that when the cells are depleted of hPrp31 or hPrp6 protein by RNAi knockdown, the formation of U4/U6.U5 tri‐snRNP is inhibited, and stable U4/U6 di‐snRNPs and U5 snRNPs accumulate in the cell (Figure 2). This demonstrates for the first time in vivo that the U4/U6 protein hPrp31 is essential for the interaction of the U4/U6 di‐snRNP with the 20S U5 snRNP, and is consistent with previous studies performed in vitro (Makarova et al, 2002). We surveyed the landscape of DNA methylation alterations in gastric cancer by analyzing genome-wide CG dinucleotide (CpG) methylation profiles of 240 gastric cancers (203 tumors and 37 cell lines) and 94 matched normal gastric tissues. Cancer-specific epigenetic alterations were observed in 44% of CpGs, comprising both tumor hyper- and hypomethylation. Twenty-five percent of the methylation alterations were significantly associated with changes in tumor gene expression. In addition to defense against foreign DNA, the CRISPR-Cas9 system of Francisella novicida represses expression of an endogenous immunostimulatory lipoprotein. We investigated the specificity and molecular mechanism of this regulation, demonstrating that Cas9 controls a highly specific regulon of four genes that must be repressed for bacterial virulence. Regulation occurs through a protospacer adjacent motif (PAM)-dependent interaction of Cas9 with its endogenous DNA targets, dependent on a non-canonical small RNA (scaRNA) and tracrRNA. The limited complementarity between scaRNA and the endogenous DNA targets precludes cleavage, highlighting the evolution of scaRNA to repress transcription without lethally targeting the chromosome.
