The ability of the computer to idealize the irregular synaptic contacts as solid disks by modifying the information gleaned from scanning the preparation allows the stereological principle of Weibel & Gomez to be used to calculate the number of E‐PTA stained synaptic contacts per unit volume of tissue. The mitochondrial derivative of the sperm cell of the Gastropod Helix is endowed with a compartment loaded with glycogen (Personne et André, 1964). The aim of this work is to establish whether this mitochondrial glycogen is synthesized in the mitochondrial derivative itself or elsewhere in the cell. For this purpose, living sperm were first incubated in a medium containing glucose 1-phosphate, then fixed, sectioned, and stained specifically for polysaccharides by the phosphotungstic acid technique or the periodic acid-thiosemicarbazide-silver proteinate technique. Comparison with controls shows that a synthesis of glycogen occurred during the time of incubation. It can be inferred from this result that an amylophosphorylase system controlling the metabolism of polysaccharides is present in the mitochondrial derivative itself. The most widely used staining procedure in recent electron microscope studies of virus-infected plants has involved the use of uranyl acetate, either during dehydration or as a post-stain, together with lead citrate as a post-stain (Arnott & Smith, 1968; Milne & de Zoeten, 1967). Following the recent demonstration that the appearance of tobacco mosaic virus in thin section could be altered by prolonging the duration of dehydration staining with uranyl acetate (Cocking & Pojnar, 1968a), the use of other heavy-metal stains for dehydration staining, both alone and in combination with uranyl acetate, has been investigated. A double-staining procedure using phosphotungstic acid and uranyl acetate has been developed and the potential of this procedure in electron-microscope studies of virus infection is discussed. Phosphotungstic acid has been known for many years to give high contrast when used as a tissue stain during dehydration (Huxley, 1958).
Polyaniline shows great potential and promises wide application in the biomedical field thanks to its intrinsic conductivity and material properties, which closely resemble natural tissues. Surface properties are crucial, as these predetermine any interaction with biological fluids, proteins and cells. An advantage of polyaniline is the simple modification of its surface, e.g., by using various dopant acids. An investigation was made into the adhesion, proliferation and migration of mouse embryonic fibroblasts on pristine polyaniline films and films doped with sulfamic and phosphotungstic acids. In addition, polyaniline films supplemented with poly (2-acrylamido-2-methyl-1-propanesulfonic) acid at various ratios were tested. Results showed that the NIH/3T3 cell line was able to adhere, proliferate and migrate on the pristine polyaniline films as well as those films doped with sulfamic and phosphotungstic acids; thus, utilization of said forms in biomedicine appears promising.
