Bacteria of the Chlamydiales order are known abortive agents, therefore cases of bovine abortion from three representative regions of Switzerland were investigated in this study. Particularly Chlamydiaceae as well as the Chlamydia-like organisms Waddlia and Parachlamydia were of interest, especially because of their possible zoonotic potential. Placenta samples (n=343) were tested for these bacteria by different PCR-methods, immunohistochemistry and serology for Chlamydia (C.) abortus. In 67.3% of the 343 cases a necrotizing and/or purulent placentitis was found histologically. By real-time PCR, 0.9% (3/343) of the cases were positive for Waddlia, 13.4% (46/343) positive for Parachlamydia and 14.6% (50/343) positive or questionable positive for Chlamydiaceae. Of these samples, confirmation by immunohistochemistry was possible in 2/3 cases for Waddlia, 25/46 for Parachlamydia and 4/50 for Chlamydiaceae. Six consecutively hatched cohorts and one cohort of pre-hatch eggs of farmed barramundi (Lates calcarifer) from south Australia were examined for Chlamydia-like organisms associated with epitheliocystis. To identify and characterise the bacteria, 59 gill samples and three pre-hatch egg samples were processed for histology, in situ hybridisation and 16S rRNA amplification, sequencing and comprehensive phylogenetic analysis. Cases of epitheliocystis were observed microscopically and characterised by membrane-enclosed basophilic cysts filled with a granular material that caused hypertrophy of the epithelial cells. In situ hybridisation with a Chlamydiales-specific probe lead to specific labelling of the epitheliocystis inclusions within the gill epithelium. Two distinct but closely related 16S rRNA chlamydial sequences were amplified from gill DNA across the seven cohorts, including from pre-hatch eggs. Environmental chlamydiae belonging to the Parachlamydiaceae are obligate intracellular bacteria that infect Acanthamoeba, a free-living amoeba, and are a risk for hospital-acquired pneumonia. However, whether amoebae harboring environmental chlamydiae actually survive in hospital environments is unknown. We therefore isolated living amoebae with symbiotic chlamydiae from hospital environments. One hundred smear samples were collected from Hokkaido University Hospital, Sapporo, Japan; 50 in winter (February to March, 2012) and 50 in summer (August, 2012), and used for the study. Acanthamoebae were isolated from the smear samples, and endosymbiotic chlamydial traits were assessed by infectivity, cytokine induction, and draft genomic analysis. From these, 23 amoebae were enriched on agar plates spread with heat-killed Escherichia coli. Two contemporary effects of humans on aquatic ecosystems are increasing temperatures and increasing nutrient concentrations from fertilizers. The response of organisms to these perturbations has important implications for ecosystem processes. We examined the effects of phosphorus (P) supply and temperature on organismal carbon, nitrogen and phosphorus (C, N and P) content, cell size and allocation into internal P pools in three strains of recently isolated bacteria (Agrobacterium sp., Flavobacterium sp., and Arthrobacter sp.). We manipulated resource C:P in chemostats and also manipulated temperatures from 10 to 30ºC. Dilution rates were maintained for all the strains at approximately 25% of their temperature-specific maximum growth rate to simulate low growth rates in natural systems. Under these conditions, there were large effects of resource stoichiometry and temperature on biomass stoichiometry, element quotas, and cell size. Each strain was smaller when C-limited and larger when P-limited.

The effect of the fish holding system (cages or ponds) on the activity of alkaline phosphatase (AP) and the brush border enzymes: maltase, leucine amino peptidase (LAP) and γ-glutamyl transferase (γ-GT) was examined. The enzymatic activity was tested in three intestinal sections: the pyloric caeca, upper and lower intestine. All the tested enzyme activities were influenced by an interaction between the intestinal section and the strain or cross of silver perch. Training and account setup are usually completed in two sessions. The first session is an introduction and lecture on the operation of one of our spectrometers, sample handling, safety, data collection and storage, and some data processing. We also set up your account so that you can reserve instrument time, and log into the instruments. You are also assigned a door code for 24/7 access to the facility. During the second session you will bring your own sample to run, and practice your skills. You can ask questions or seek further training at that point. NMR users at WSU are mostly self-sufficient-users run their own samples and experiments, collect and file data, and do data processing independently.
