In neonatal, weanling, and adult chimeric atria and ventricles, irregularly formed patches of various sizes rather than highly dispersed cardiomyocytes were observed. Most of the smaller patches and single cardiomyocytes were found in spatial neighborhood of large patches. Although telomere length is genetically determined, mouse embryonic stem (ES) cells with telomeres of twice the normal size have been generated. Here, we use such ES cells with hyper-long telomeres, which also express green fluorescent protein (GFP), to generate chimaeric mice containing cells with both hyper-long and normal telomeres. We show that chimaeric mice contain GFP-positive cells in all mouse tissues, display normal tissue histology and normal survival. Both hyper-long and normal telomeres shorten with age, but GFP-positive cells retain longer telomeres as mice age. Chimaeric mice with hyper-long telomeres also accumulate fewer cells with short telomeres and less DNA damage with age, and express lower levels of p53. Reciprocal hybrids showing different phenotypes have been well documented in previous studies, and many factors accounting for different phenotypes have been extensively investigated. However, less is known about whether the profiles of small RNAs differ between reciprocal hybrids and how these small RNAs affect gene expression and phenotypes. To better understand this mechanism, the role of small RNAs on phenotypes in reciprocal hybrids was analysed. Reciprocal hybrids between Solanum lycopersicum cv. Micro-Tom and S. pimpinellifolium line WVa700 were generated. Significantly different phenotypes between the reciprocal hybrids were observed, including fruit shape index, single fruit weight and plant height. What is the issue?. It has recently been recognised that a specific brand of medical device used in open-heart surgery, known as a heater-cooler unit, has the potential to be contaminated with an uncommon environmental bacterium called Mycobacterium chimaera (M. chimaera), which grows in the units water tank.. The water and the heater-cooler unit itself never comes into contact with the patients blood or body fluids. M. chimaera can become airborne from the heater-cooler device, and can make contact with a patients wound during open heart surgery, potentially resulting in infection. Activation of A2A adenosine receptors (A2ARs) protects kidneys from ischemia-reperfusion injury (IRI). A2ARs are expressed on bone marrow-derived (BM-derived) cells and renal smooth muscle, epithelial, and endothelial cells. To measure the contribution of A2ARs on BM-derived cells in suppressing renal IRI, we examined the effects of a selective agonist of A2ARs, ATL146e, in chimeric mice in which BM was ablated by lethal radiation and reconstituted with donor BM cells derived from GFP, A2AR-KO, or WT mice to produce GFP→WT, A2A-KO→WT, or WT→WT mouse chimera. We found little or no repopulation of renal vascular endothelial cells by donor BM with or without renal IRI. ATL146e had no effect on IRI in A2A-KO mice or A2A-KO→WT chimera, but reduced the rise in plasma creatinine from IRI by 75% in WT mice and by 60% in WT→WT chimera. Hi netters! I have a copy of mulfold for the Mac. A limitation of this version: one can only fold a 300 base sequence. I, however, have a sequence of ~4000 that I want to fold to see if the start codon or SD domain could potentially interact with bases more distant than the neighboring ~150 bases (and the effect of mutations in these more distant domains on this interaction). Is there anyway I could do this without constructing some chimeric sequence between the initiation region and the distant region of interest (I dont think the results I would get from this would be very realistic)? Do you thing it is reasonable (thermodynamically) for a such a distant region (say ~500 bases away) to base pair with the initiation region?
