Pulmonary alveolar proteinosis (PAP) is a rare disease of unknown aetiology characterized by accumulations of lipoproteinaceous material within the alveoli. Eosinophil accumulation is a distinctive feature of lung allergic inflammation. Here, we have used a mouse model of OVA (ovalbumin)-induced pulmonary eosinophilia to study the cellular and molecular mechanisms for this selective recruitment of eosinophils to the airways. OBJECTIVE: Nicotinic acid (NA) treatment has been associated with benefits in atherosclerosis that are usually attributed to effects on plasma lipoproteins. The NA receptor GPR109A is expressed in monocytes and macrophages, suggesting a possible additional role for NA in modulating function of these immune cells. We hypothesize that NA has the potential to act directly on monocytes to alter mediators of inflammation that may contribute to its antiatherogenic effects in vivo. METHODS AND RESULTS: In human monocytes activated by Toll-like receptor (TLR)-4 agonist lipopolysaccharide, NA reduced secretion of proinflammatory mediators: TNF-α (by 49.2±4.5%); interleukin-6 (by 56.2±2.8%), and monocyte chemoattractant protein-1 (by 43.2±3.1%) (P|0.01). Methods and Results-Contrast-enhanced ultrasound perfusion imaging and molecular imaging of endothelial P-selectin expression in the proximal hindlimb were performed at 1, 3, and 21 days after arterial ligation in wild-type and db/db mice. Ligation reduced muscle blood flow to ≈0.05 mL/minute per gram in both strains. Significant recovery of flow occurred only in wild-type mice (60%-65% of baseline flow). On molecular imaging in db/db mice, baseline P-selectin signal was 4-fold higher in db/db compared with wild-type mice (P,0.01) but increased minimally at day 1 after ischemia, whereas signal increased approximately 10-fold in wild-type mice (P,0.01). Immunohistology of the hindlimb demonstrated severely reduced monocyte recruitment in db/db mice compared with wild-type mice. Previously, we showed that a chemokine CCL2 recruits IMs to metastatic sites where they differentiate to MAMs (Qian et al., 2011). In this study, we revealed a novel role for CCL2 as a trigger of a prometastatic chemokine cascade involving CCL3 signaling via CCR1 that is required for efficient metastasis. These data illustrate a signaling relay that amplifies the pathology already in the system by promoting retention of recruited monocytes that stimulate tumor cell establishment at the metastatic site.. Our in vivo and in vitro results indicate that CCL2 can increase CCL3 expression in MAMs at the metastasis site.
