In the present study we show that protein kinase C (PKC) plays an important role in APAP-induced liver injury through JNK-dependent and independent pathways. Treatment of primary mouse hepatocytes with two different broad-spectrum PKC inhibitors (Ro-31-8245, Go6983), protected against APAP hepatotoxicity without inhibiting JNK activation. The main objective of this study was to characterize release profile of a water soluble drug model from polyvinyl alcohol (PVOH) cryogels made at different polymer concentrations and thicknesses. Background. PVOH is a hydrophilic linear polymer, and its aqueous solutions can form a gel when exposed to repeated freeze-thaw cycles. The cryogels prepared as such possess unique mechanical, elastic and swelling properties which can be applicable for pharmaceutical and biomedical applications. Methods. Aqueous PVOH solutions were prepared by dissolving the polymer in deionized water at 90oC under mechanical mixing. In this study, we have developed and validated a simple, accurate and sensitive gas chromatography-mass spectrometry (GC-MS) method for simultaneous quantification of 18 fatty acids in rat serum, including both non-esterified (NEFA) and esterified (EFA) fatty acids, and subsequent analysis of fatty acid metabolic profiles. This novel method was used to evaluate the serum levels of fatty acids from vehicle- and acetaminophen (APAP)-treated rats. Recent surveys of various aquatic habitats have found detectable concentrations of several organic wastewater contaminants (OWCs). The effects of these OWCs on aquatic organisms are relatively unknown. We studied the effects of ecologically-relevant concentrations of two OWCs, acetaminophen and triclosan, on the behavior, growth, and survivorship of Xenopus laevis tadpoles. Xenopus laevis decreased activity with increased concentrations of acetaminophen, but acetaminophen had no effect on startle response, final mass, nor suvivorship. Triclosan had a negative effect on tadpole activity, but no effect on startle response. High concentrations of triclosan had a negative effect on final mass and intermediate concentrations had a positive effect on final mass relative to controls. Triclosan did not affect survivorship.
