A pathophysiologic study of the kidney tubule to optimize organ preservation solutions. Background. Tissue damage at the time of organ transplantation has a negative impact on the subsequent success of the procedure, both in the immediate and longer term. Hypothermia is the principal element used to prolong organ viability ex vivo, but paradoxically also induces cellular edema through inhibition of energy-dependent adenosine triphosphatases (ATPases). This induces an electrolyte imbalance that leads to fluid influx and cell swelling. It is important, therefore, that improvements are made in the preservation of ischemic organs to reduce this injury. Methods. This study has applied a novel in vitro system to model cold and warm ischemic-induced renal tubule swelling that characterizes tissue damage in ischemia/reperfusion injury. Biochemical blockade of ATPases in this system using strophanthidin modeled the effects of energy depletion and induced cell swelling. By measuring such tubule swelling ...

Human serum albumin (HSA) is a mixture of human mercaptalbumin (HMA, reduced form) and nonmercaptalbumin (HNA, oxidized form). We have developed a convenient high-performance liquid chromatographic (HPLC) system for the separation of HSA into HMA and HNA, and studied the mercapt,==,nonmercapt conversion (i.e., dynamic change in the redox state) of HSA. Examination of long-term sample preservation temperature on the redox state of HSA is of fundamental importance for analysis of defense systems against oxidants in humans. The HMA fraction of HSA (f(HMA)) was markedly decreased, i.e., the redox state of HSA samples was more oxidized, when they were kept even at -20 degrees C for 170 days. Moreover, the redox states of five commercial HSA products were analyzed and the results were compared with those for normal control subjects. Surprisingly, marked decreases in f(HMA) value for all commercial HSA products were observed. ...

Introduction. Controlled oxygenated rewarming of cold stored livers by machine perfusion demonstrated a strong protective effect in preclinical studies. Here we report on the first clinical application of this new concept in a pilot series of 6 marginal donor grafts.. Methods. Between 03/2014 and 05/2014 six distally procured livers from DBD-donors, which had been rejected by other transplant centers were subjected to the controlled rewarming protocol before transplantation. Immediately after arrival in our transplant center the hepatic artery and portal vein were cannulated. Gentle rewarming of the graft was effectuated thereafter by pressure controlled oxygenated machine perfusion (MP) via portal vein and hepatic artery (Organ Assist, Groningen, NL) while gradually increasing perfusate temperature up to 20°C. Custodiol-N was used as preservation solution. Real-time biochemical analysis of perfusate parameters was carried out in constant time intervals during the controlled ...
α-Galactosidases are enzymes that act on galactosides present in many vegetables, mainly legumes and cereals, have growing importance with respect to our diet. For this reason, the use of their catalytic activity is of great interest in numerous biotechnological applications, especially those in the food industry directed to the degradation of oligosaccharides derived from raffinose. The aim of this work has been to optimize the recombinant production and further characterization of α-galactosidase of Saccharomyces cerevisiae. The MEL1 gene coding for the α-galactosidase of S. cerevisiae (ScAGal) was cloned and expressed in the S. cerevisiae strain BJ3505. Different constructions were designed to obtain the degree of purification necessary for enzymatic characterization and to improve the productive process of the enzyme. ScAGal has greater specificity for the synthetic substrate p-nitrophenyl-α-d-galactopyranoside than for natural substrates, followed by the natural glycosides, melibiose, raffinose
More than two million American women are breast cancer survivors. Approximately one-third of these women are premenopausal at diagnosis and face issues related to reproduction as they undergo cancer treatment. Ovarian function after breast cancer diagnosis has implications on breast cancer prognosis, choice of adjuvant therapy and reproductive issues such as desire for fertility or concerns about menopause. Therefore, tools to accurately predict ovarian function in breast cancer survivors could significantly impact physicians and patients in counseling, medical and surgical treatment choices, and consideration of fertility preservation options.. The goal of this proposal is to identify pre-chemotherapy hormonal, genetic and ovarian imaging markers that can predict ovarian failure and characterize the course of ovarian function after chemotherapy. The investigators plan to follow a group of young women from breast cancer diagnosis to five years after chemotherapy. The investigators will study the ...

Whole cell and surface proteomes were analyzed together with adhesive properties of the probiotic bacterium Lactobacillus acidophilus NCFM (NCFM) grown on the emerging prebiotic raffinose, exemplifying a synbiotic. Adhesion of NCFM to mucin and intestinal HT-29 cells increased three-fold after culture with raffinose versus glucose, as also visualized by scanning electron microscopy. Comparative proteomics using 2D-DIGE showed 43 unique proteins to change in relative abundance in whole cell lysates from NCFM grown on raffinose compared to glucose. Furthermore, 14 unique proteins in 18 spots of the surface subproteome underwent changes identified by differential 2DE, including elongation factor G, thermostable pullulanase, and phosphate starvation inducible stress-related protein increasing in a range of +2.1 − +4.7 fold. By contrast five known moonlighting proteins decreased in relative abundance by up to −2.4 fold. Enzymes involved in raffinose catabolism were elevated in the whole cell ...

Paraovarian/paratubal cysts constitute 5-20% of all adnexal lesions and typically originate from the paramesonephric or Müllerian duct. The primary epithelial tumors arising from paraovarian cysts account for 25% of the cases, but giant cystadenomas of paraovarian origin are extremely uncommon during childhood and adolescence with very few cases reported in the literature. We present the case of a 15-year-old female that presented with a bulky mass in the abdomen and pelvis. An initial clinical and radiological examination indicated an ovarian cyst measuring ∼25x20 cm. However, explorative laparotomy revealed a giant paratubal cyst that was successfully treated with complete excision using fertility-sparing surgery. Histopathological examination was consistent with a serous cystadenoma. The postoperative course was uneventful and the girl was discharged on the seventh postoperative day. At the follow-up of six months, the patient was doing well. Due to their rarity and enormous size, the ...

Several stochastic simulation algorithms (SSAs) have been recently proposed for modelling reaction-diffusion processes in cellular and molecular biology. In this talk, two commonly used SSAs will be studied. The first SSA is an on-lattice model described by the reaction-diffusion master equation. The second SSA is an off-lattice model based on the simulation of Brownian motion of individual molecules and their reactive collisions. The connections between SSAs and the deterministic models (based on reaction-diffusion PDEs) will be presented. I will consider chemical reactions both at a surface and in the bulk. I will show how the 'microscopic' parameters should be chosen to achieve the correct 'macroscopic' reaction rate. This choice is found to depend on which SSA is used. I will also present multiscale algorithms which use models with a different level of detail in different parts of the computational domain ...

Recent years have seen an explosion of interest in stochastic partial differential equations where the driving noise is discontinuous. In this comprehensive monograph, two leading experts detail the evolution equation approach to their solution. Most of the results appeared here for the first time in book form. The authors start with a detailed analysis of Lévy processes in infinite dimensions and their reproducing kernel Hilbert spaces; cylindrical Lévy processes are constructed in terms of Poisson random measures; stochastic integrals are introduced. Stochastic parabolic and hyperbolic equations on domains of arbitrary dimensions are studied, and applications to statistical and fluid mechanics and to finance are also investigated. Ideal for researchers and graduate students in stochastic processes and partial differential equations, this self-contained text will also interest those working on stochastic modeling in finance, statistical physics and environmental science. ...
The emerging of nanotechnology has increasingly gained expansions and applications in various materials science research and development. However, the exposure to nanoparticles and engineered nanomaterials can lead to adverse biological effects because the small sizes of nanoparticles can enter the human body and deposit in the organs or translocate from the intake area to the secondary organs and can cause inflammation. One of the most used nanoparticles is TiO2, which is commonly found in skin care and household products. It is still unclear how TiO2 nanoparticles are remained in human bodies after exposing. In the present study, we develop a physiologically-based pharmacokinetic (PBPK) model to predict the bio-distribution of TiO2 concentrations in rat tissues. The model is validated with an existing in-vivo study in rats. We also extend our PBPK model to predict cell death caused by TiO2 nanoparticles in the rat liver using a dose-response model. The dose-response model accounts for the interplay
The purpose of this study was to analyze the possible effects of machine perfusion (MP) versus cold storage (CS) on delayed graft function (DGF) and early graft survival in expanded criteria donor kidneys (ECD). As part of the previously reported international randomized controlled trial 91 consecutive heart-beating deceased ECDs--defined according to the United Network of Organ Sharing definition--were included in the study. From each donor one kidney was randomized to MP and the contralateral kidney to CS. All recipients were followed for 1 year. The primary endpoint was DGF. Secondary endpoints included primary nonfunction and graft survival. DGF occurred in 27 patients in the CS group (29.7%) and in 20 patients in the MP group (22%). Using the logistic regression model MP significantly reduced the risk of DGF compared with CS (OR 0.460, P=0.047). The incidence of nonfunction in the CS group (12%) was four times higher than in the MP group (3%) (P=0.04). One-year graft survival was significantly
Fertility and pregnancy issues are of key importance for young breast cancer patients. Despite several advances in the field, there are still multiple unmet needs and barriers in discussing and dealing with these concerns. To address the significant challenges related to fertility and pregnancy issues, the PREgnancy and FERtility (PREFER) study was developed as a national comprehensive program aiming to optimize care and improve knowledge around these topics. The PREFER study is a prospective cohort study conducted across several Italian institution affiliated with the Gruppo Italiano Mammella (GIM) group evaluating patterns of care and clinical outcomes of young breast cancer patients dealing with fertility and pregnancy issues. It is composed of two distinctive studies: PREFER-FERTILITY and PREFER-PREGNANCY. The PREFER-FERTILITY study is enrolling premenopausal patients aged 18-45 years, diagnosed with non-metastatic breast cancer, who are candidates to (neo)adjuvant chemotherapy and not previously

Have you just recently discovered that your milk, eggs, and other refrigerated items seem a little warmer than usual? This is definitely an issue, as specific foods require to be stored at the optimal temperature to avoid ruining. Have you examined the freezer temperature, but it's still cold? If so, you might have a refrigerator stop cooling on refrigerator side problem on your hands.. So what triggers the refrigerator to stop cooling on refrigerator side issue? There are in fact several things that can trigger the refrigerator not cooling issue. Much depends on the make, design, and brand name of your refrigerator, however there are a couple of more common failures that lead to a failing fridge. Repair work range from defrosting refrigerator evaporator coils to replacing the refrigerator thermostat.. We've detailed a few of the most typical factors a refrigerator isn't cooling on refrigerator side for you here. Use this list to repair and see if you can fix it yourself, or click fridge repairs ...
BACKGROUND: The standard preservation solution used during organ procurement and preservation of most organs is the University of Wisconsin (UW) solution. Despite its superiority over other cold storage solutions, the inclusion of hydroxyethyl starch (HES) as one of the components of the UW solution has been both advocated and denied. This study determined whether HES had any effect on red blood cell (RBC) aggregability and correlated aggregation parameters with HES molecular weight. METHODS: Human RBC aggregability and deformability were investigated in vitro, at 4 degrees C, with a laser-assisted optical rotation cell analyzer. The study of RBC aggregation in a binary HES-HES system gave an indication about the nature of HES-RBCs interactions. Bright field microscopy and atomic force microscopy were used to morphologically characterize the aggregates size and form. RESULTS: High molecular weight HES and UW solution had a potent hyperaggregating effect; low molecular weight HES had a hypoaggregating

For freezing ice cream or other confections in a refrigerator's freezer area, a refrigerator tray was a rectangular, shallow, open container. The ice cube tray (often provided with the refrigerator) began, around the mid-to-late thirties, to be fitted with a removable divider so that it doubled as the refrigerator tray.. Ice cube trays with removable dividers are referred to as "refrigerator trays" and were designed specifically for freezing desserts.. The terms "refrigerator tray", "freezer tray", and "ice cube tray" were used somewhat interchangeably for making desserts with these trays, at least during the period after ice cube trays with removable dividers were included with refrigerators and until the included ice cube trays became one-piece plastic trays.. The Carnation Fun to Cook Book, for example, has a recipe for "tropical freeze" on page 40; for the final freeze, the recipe is poured into "two small (or 1 large) refrigerator trays". The drawings accompanying the recipe show shallow, ...
Natural gas is produced through thermogenic and biogenic processing over millions of years and natural gas transmission plants and storage solutions can be as difficult to achieve as the recovery and processing activities. The gas can be partially processed before being piped to the refineries, were the natural gas is further processed to produce pipeline quality gas. Transmission is either by pipelines from the processing plants to the consumers or by conversion of the gas to liquid natural gas and transported in specialist LNG carriers. Storage can be in natural underground caverns or aquifers or depleted oil/gas reservoirs.
The shortage of donor organs for liver transplantation requires investigations into expanding the donor pool. Non-heart beating donor (NHBD) livers are an untapped source of organs. NHBD organs are those that have had a period of warm ischemia prior to cold preservation. A main concern of NHBD livers is that there is a depletion of energy stores during the warm ischemia and subsequent cold storage. This study is to investigate the use of hypothermic machine perfusion to regenerate energy stores in the liver during 5hr storage and after 1hr rewarming in a rat transplantation model. Results will be compared with control. (no warm ischemia) and simple cold stored tissues.. Copyright © 2003 by ASME ...

The use of energy crops and agricultural residues is expected to increase to fulfil the legislative demands of bio-based components in transport fuels. Ensiling methods, adapted from the feed sector, are suitable storage methods to preserve fresh crops throughout the year for, for example, biogas production. Various preservation methods, namely ensiling with and without acid addition for whole crop maize, fibre hemp and faba bean were investigated. For the drier fibre hemp, alkaline urea treatment was studied as well. These treatments were also explored as mild pretreatment methods to improve the disassembly and hydrolysis of these lignocellulosic substrates. The investigated storage treatments increased the availability of the substrates for biogas production from hemp and in most cases from whole maize but not from faba bean. Ensiling of hemp, without or with addition of formic acid, increased methane production by more than 50% compared to fresh hemp. Ensiling resulted in substantially increased
The effects of cold storage using Custodiol® (Histidine-Tryptophan-Ketoglutarate, HTK) or isotonic saline solution on mitochondrial function in hearts (left and rights ventricles) and various blood vessels of pigs were investigated. Hearts, saphenous veins, internal-mammary-arteries and aortas of male landrace pigs were harvested and exposed to cold ischemia in either saline or Custodiol-HTK solution. Mitochondrial function was measured in situ in permeabilized fibers by high-resolution respirometry. Mitochondrial respiratory capacities (maximal respiration rates) were similar in the right and left ventricle in controls and after 14 h of cold storage were significantly better preserved in Custodiol-HTK than in saline solution. Mitochondrial respiration rates in various blood vessels including aorta, arteries and veins were less than 5% of myocardium rates. In contrast to the pig heart, in some blood vessels, like veins, mitochondrial function remained stable even after 24 h of cold ischemia. HTK

N2 - Ischemia/reperfusion injury (IRI) is the most common cause of early mortality following lung transplantation (LTx). We hypothesized that nitrite, an endogenous source of nitric oxide (NO), may protect lung grafts from IRI. Rat lung grafts were stored in preservation solution at 4°C for 6 hours. Both grafts and recipients were treated with nitrite. Nitrite treatment was associated with significantly higher levels of tissue oxygenation, lower levels of cytokines and neutrophil/macrophage infiltration, lower myeloperoxidase activity, reduced oxidative injury and increased ...

To assess reduction in heavy menstrual bleeding and dysmenorrhea following MRI guided Focused Ultrasound Surgery (MRgFUS) of focal and diffuse adenomyosis up to 12 months post-treatment a retrospective cohort study was done at a tertiary care academic medical center for obstetrics, gynecology and infertility. MRgFUS for adenomyosis uterus was done for thirty-seven patients presenting with symptoms of heavy menstrual bleeding and dysmenorrhea with MRI-suspected adenomyosis. The main outcome measure, was reduction in heavy menstrual bleeding, dysmenorrhea and Symptom Severity Scoring (SSS) over a 3, 6 and 12 month period. Secondary outcome was evidence of fertility preservation post procedure. D'Agostino & Pearson omnibus normality test, one-way Ananova, Pearson's correlation coefficient analysis was performed on the data. Statistical significances, p-value and r-value were determined. Out of 37 patients who were treated by MRgFUS, 26 had sufficient follow-up to be included in the analysis. SSS calculated
Author: Schlesser Vincent, Hecq Jean-Daniel, Vanbeckbergen Danielle, Jamart Jacques, Galanti Laurence M, Year: 2002, Abstract: Cefepime is often used in antibiotic infusions for hospitalized patients. Using intravenous solutions of cefepime prepared in advance can be efficient, but little information is available about the stability of that antibiotic after freezing followed by microwave thawing. The purpose of this study was to investigate how freezing, long-term storage, and microwave thawing can affect the stability of cefepime in 5% dextrose injection.

Sugars are classified into three groups: monosaccharides, disaccharides, and trisaccharides. The simplest molecules of sugars are the monosaccharides, which include galactose, fructose and glucose, the only monosaccharides absorbed by humans. Disaccharides (including lactose, maltose and sucrose) and trisaccharides (including raffinose, found in cottonseed and sugar beets), are derived from the union of monosaccharides. All of these sugars provide approximately 4 cal/g.. Household 'sugar', or 'table sugar', is extracted mainly from sugar cane or beet. This sugar is a disaccharide composed of 50% glucose and 50% fructose linked by a-1,4 glycosidic bonds (Pancoast and Junk, 1980). Glucose, also known as dextrose or corn syrup, is produced from corn starch. Fructose is the sweetest of the simple sugars and is found as the monosaccharide, along with glucose and sucrose, in fruits and vegetables (Park and Yetley, 1993). It is generally present in honey and fruits and vegetables in similar amounts to ...
