The affinity of metal chelates for amino acids, such as histidine, is widely used in purifying proteins, most notably through six-histidine `tails. We have found that metal affinity interactions can also be applied to separation of single-stranded nucleic acids through interactions involving exposed purines. Here we describe a metal affinity precipitation method to resolve RNA from linear and plasmid DNA. A copper-charged copolymer of N-isopropyl acrylamide (NIPAM) and vinyl imidazole (VI) is used to purify plasmid from an alkaline lysate of E. coli. The NIPAM units confer reversible solubility on the copolymer while the imidazole chelates metal ions in a manner accessible to interaction with soluble ligands. RNA was separated from the plasmid by precipitation along with the polymer in the presence of 800 mM NaCl. The drop size distribution in rain can be computed from the Doppler spectrum, provided that the updraft can be estimated, but difficulties are involved in the case of snow. Doppler spectra and their moments are computed for rain by using various power law relations of fall speed υ versus particle diameter D and an exponential fit to the actual fall speed data. In the former case, there is no sharp upper bound to the spectra and all the spectral moments increase with rainfall rate R without limit; in the latter case, there is a sharp upper bound of the spectra corresponding to the limiting terminal velocity of raindrops, and the spectral moments approach an asymptote. After speaking with Axa regarding the DNA concentrations, he would like the DNA from the ethanol-fixed tissue to be more concentrated, and he wants them in ddH2O instead of Buffer AE (from the Qiagen DNeasy Kit). So, I preformed a standard ethanol precipitation. Added 0.1 volumes of 3M sodium acetate (pH = 5.2) [15uL], 2.5 volumes of 100% ethanol [412.5uL] and incubated @ -20C over the weekend.. Pelleted DNA by spinning 16,000g, 15mins @ 4C. Discarded supe, washed pellets with 1mL 70% ethanol and re-pelleted the DNA. A method for preparing an aqueous solution enriched in EG III from an aqueous mixture containing cellulase proteins, xylanase and EG III is disclosed. The method involves adding an amount of a low molecular weight alcohol selected from the group consisting of ethanol, methanol, propanol and mixtures thereof to the aqueous mixture containing cellulase proteins, xylanase and EG III and an organic salt under conditions wherein substantially all of the cellulase proteins other than EG III and xylanase are precipitated out of the aqueous mixture. The method then involves removing the precipitate from the aqueous mixture so as to recover an aqueous supernate enriched in EG III. Next, the method involves adding an amount of an inorganic salt to the supernate produced in step b) so as to form a second precipitate and a second supernate and then finally collecting the second supernate from the second precipitate to obtain a supernate enriched in EG III.

|p|Water desalination by membrane distillation is restricted by calcium carbonate precipitation on the membrane surface, what resulted in a progressive decline of the permeate flux. The module rinsing by HCl solution removed this deposit, however, such operation could cause the membrane wettability, associated with lowering of the permeate flux. This phenomenon was inhibited when the membrane modules with prefiltration element, assembled directly at the inlet to module, were used. The salt deposits were formed mainly on the surface of the filtration element (nets area constituted over 10% of the membranes area), what protected the membranes assembled inside the module. The filter efficiency decreased when the deposit layer covered the entire surface of pre-filter element, and after a few hours (period of 10-20 h) the formation of crystallite on the membrane surface was also observed. Therefore, the formed deposit was systematically removed (at every 5 h) from this pre-filter by 3 wt.%

The effect of insulin administration on immune complex (IC) formation in diabetic patients was analysed in vivo and in vitro. Firstly, serial studies of IC status were performed over a mean period of 18 months in 44 diabetic patients, 37 of whom were receiving standard insulin therapy. Thirty patients changed to monocomponent (MC) insulin while seven commenced MC insulin after tablet failure. The other seven patients remained on standard insulin throughout the study. Secondly, nine patients had serial measurements of IC over a 6-8 h period following a routine morning dose of MC insulin; eight control subjects were similarly studied. The insulin content of IC in insulin treated patients was assessed in vitro by examining, (a) the selective precipitation of antibody bound insulin by 3% polyethylene glycol (PEG) and (b) the insulin specificity of antisera raised against PEG precipitates of IC positive sera. Precipitation of insulin is a problem with mechanical insulin infusion devices. Therefore conditions affecting insulin precipitation have been studied in a recirculating system using a peristaltic pump. Acetate buffer favoured precipitation and a pH of 2, 3 or 8 resulted in no less (and often more) precipitation than a pH of 7 using either acetate of phosphate buffer. However, no precipitation occurred in a phosphate buffered neutral preparation after ten days of pumping through tubing pretreated with EDTA. In comparison at pH 7, using acetate buffer with untreated tubing, more than 95% of insulin was precipitated in ten days. A heavy metal-insulin association appears to be a major factor in the precipitation. However, some formation of insulin fibrils is probable as the precipitate is autocatalytic and partially acid insoluble. Phosphate buffered highly purified porcine insulin is suitable for relatively prolonged infusion if metal ion contamination of the delivery system is minimised.

Rat C-reactive protein (CRP) is a serum glycoprotein belonging to the pentraxin family of proteins. In this study we have shown the specific binding of 125I-CRP to rat peritoneal macrophages at 4 degrees C. This binding was dependent upon incubation time, CRP and cell concentrations, and was not inhibited by either phosphorylcholine or human IgG. At 37 degrees C, the surface-bound 125I-CRP was internalized and degraded. The degradation of 125I-CRP was measured by the formation of 125I-labelled trichloroacetic-acid-soluble CRP peptides by either precipitation assays or by h.p.l.c. of the incubation medium using a gel-filtration column. Since chloroquine and leupeptin inhibited CRP degradation, it was concluded that degradation of CRP occurred in the lysosomal compartment of the macrophage. The average particle size was determined from the X-ray line broadening. Samples were characterized by XRD, FTIR and UV. The composition was verified by EDAX spectrum. The hexagonal structure of the sample was identified. The size of the particles increased as the annealing temperature was increased. The crystallite size varied from 5 nm to 34 nm as the calcination temperature increased. At around $700^{\circ}$C, ZnS is converted into ZnO phase due to oxidation. The emission peak of the sample is observed at 300 nm resulting in blue emission. The solid state theory based on the delocalized electron and hole within the confined volume can explain the blue-shifted optical absorption spectra.
