The affinity of metal chelates for amino acids, such as histidine, is widely used in purifying proteins, most notably through six-histidine `tails. We have found that metal affinity interactions can also be applied to separation of single-stranded nucleic acids through interactions involving exposed purines. Here we describe a metal affinity precipitation method to resolve RNA from linear and plasmid DNA. A copper-charged copolymer of N-isopropyl acrylamide (NIPAM) and vinyl imidazole (VI) is used to purify plasmid from an alkaline lysate of E. coli. The NIPAM units confer reversible solubility on the copolymer while the imidazole chelates metal ions in a manner accessible to interaction with soluble ligands. Microbial induced carbonate precipitation (MICP) is a common occurrence of geochemistry influences in many fields, such as biological, geographical, and engineering systems. However, the processes that control interactions between carbonate biomineralization and biofilm properties are poorly understood. We develop a method for real time, in situ and nondestructive imaging with confocal scanning microscopy. This method provides a possible way to observe biomineralization process and the morphology of biomineralized deposits within biofilms. We use this method to show calcite biominerals produced by Pseudomonas aeruginosa biofilms which extremely change biofilm structures. The distribution of calcite precipitation produced in situ biomineralization is highly heterogeneous in biofilms and also to occur primarily on the bottom of biofilms. The drop size distribution in rain can be computed from the Doppler spectrum, provided that the updraft can be estimated, but difficulties are involved in the case of snow. Doppler spectra and their moments are computed for rain by using various power law relations of fall speed υ versus particle diameter D and an exponential fit to the actual fall speed data. In the former case, there is no sharp upper bound to the spectra and all the spectral moments increase with rainfall rate R without limit; in the latter case, there is a sharp upper bound of the spectra corresponding to the limiting terminal velocity of raindrops, and the spectral moments approach an asymptote. Inconel© 625 is a Nickel-base superalloy used in aerospace industry as turbine shrouds, spray bars, hydraulic tubing, armoring and thrust reversers systems [1]. Apparatus for producing nano-particles comprises a furnace defining a vapor region therein. A precipitation conduit having an inlet end and an outlet end is positioned with respect to the furnace so that the inlet end is open to the vapor region. A quench fluid port positioned within the precipitation conduit provides a quench fluid stream to the precipitation conduit to precipitate nano-particles within the precipitation conduit. A product collection apparatus connected to the outlet end of the precipitation conduit collects the nano-particles produced within the precipitation conduit.
After speaking with Axa regarding the DNA concentrations, he would like the DNA from the ethanol-fixed tissue to be more concentrated, and he wants them in ddH2O instead of Buffer AE (from the Qiagen DNeasy Kit). So, I preformed a standard ethanol precipitation. Added 0.1 volumes of 3M sodium acetate (pH = 5.2) [15uL], 2.5 volumes of 100% ethanol [412.5uL] and incubated @ -20C over the weekend.. Pelleted DNA by spinning 16,000g, 15mins @ 4C. Discarded supe, washed pellets with 1mL 70% ethanol and re-pelleted the DNA. Performed a second wash with 70% EtOH, pelleted DNA, discarded supe, resuspended DNA in 75uL of NanoPure H2O, and specd on NanoDrop1000.. Results:. The sensitivity of warm- and mixed-phase orographic precipitation to the aerosol background with simultaneous changes in the abundance of cloud condensation nuclei and ice nucleating particles is explored in an idealized, two-dimensional modeling study. The concept of precipitation susceptibility dlnP/dlnN, where P is the precipitation mixing ratio and N the cloud droplet number, is adapted for orographic clouds. Precipitation susceptibility is found to be low because perturbations to different precipitation formation pathways compensate each other. For mixed-phase conditions, this in particular means a redistribution between warm and cold pathways. Precipitation is usually classified as convective or stratiform. A mathematical model characterizing the operation of the benzene stripping columns in the In-Tank Precipitation (ITP) process has been developed. The model was verified using operations data of two stripping columns, and the model predicted reasonable, conservative values of the height of a transfer unit (HTU) and outlet solute concentrations.
