Our study identifies an in vivo mechanism that regulates Yap activity within the zebrafish caudal fin blastema based on cell density differences along the regenerating tissue. The mesenchymal cells seem to respond to a mechanotransduction process that involves changes in cell morphology, junction assembly and cytoskeleton remodeling, which together lead to a graded control of tissue growth via Yap, the Hippo pathway effector.. This is a very hard question to answer because it involves a number of variables. The simplest answer is, samples preparation typically takes ~1 weeks for standard samples with all reagents on hand (QC, prep, QC). For newer or rarely used methods, the number can extend to a month if we need to gather reagents. Once the sample is ready, it goes in to the sequencing queue. This queue is often the longest part of the process. Once the full flowcell is ready and put on a sequencer, it takes ~1.3h per base to sequence (this used to be 1h but upgrades to the machines have slowed them down some). A 40nt read takes just over 2 days. Turn around (for PE samples) functionally takes about a 1/2 day. So a 100+100PE sample will take about 11 days once it is on the sequencer - if nothing goes wrong. Abstract: Electromagnetic fields (EMFs) inhibit the formation and differentiation of neural stem cells during embryonic development. In this study, the effects of prenatal exposure to EMF on the number of granule cells in the dentate gyrus of 4-week-old rats were investigated. This experiment used a control (Cont) group and an EMF exposed (EMF) group (three pregnant rats each group). The EMF group consisted of six offspring (n=6) of pregnant rats that were exposed to an EMF of up to 900 megahertz (MHz) for 60 min/day between the first and last days of gestation. I took mum to the haematologist yesterday for her checkup, and got her blood tested. The blood test results were not too bad, although the platelets were still down a bit. The red and white cell numbers are down a bit - but the platelets are in the 70s whereas 150 is in the normal range. This is an improvement on last tuesday when they were 6, but still a bit low. Mums kidneys are still not 100% either, but this is a known issue, and mum just needs to keep up her fluids at this stage. The doctor decided to not do any more lumbar puncture chemo for the moment, and has put mum on weekly blood tests just in case. She is thinking that the results are caused by the chemo. Sparse and dense cultures of chick embryo cells were affected differently by pH. The rates of cell multiplication and of thymidine-3H incorporation into DNA of dense cultures were increased as the pH was increased from 6.6 to 7.6. At pH higher than 7.6 the rate of multiplication decreased slightly in the dense cultures, but the rate of thymidine-3H incorporation continued to increase. The discrepancy was due in part to cell death and detachment at very high pH, and in part to a more rapid uptake of thymidine-3H at very high pH. Sparse cultures were much less sensitive to pH reduction and, when a suitably conditioned medium was used to minimize cell damage, very sparse cultures grew almost as well at pH 6.7 as at higher pH.
