A resorbable bone replacement material made of calcium phosphate particles of different phases which are embedded in an inventive-specific cross-linked collagen matrix. The goal is to form a non-brittle, bone replacement moulded body having a positive fit, i.e. having a shape which is anatomic and/or corresponds to the defect, which perfectly fills the bone defect and can be resorbed thereby. Said goal is achieved by producing the bone replacement material made of a mixture of calcium phosphate particles which is embedded in an inventive cross-linked collagen matrix. In particular, the collagen cross-linking is achieved by a Laccase-induced peptide cross-linking and suitable bridge molecules. Essentially substituted dihydroxyarmotes and/or substrates of the lignolytic polyphenoloxidases, such as Laccases, are suitable as bridge molecules. Also, monocyclic ortho-dihydroxyaromates, monocyclic para-dihydroxyaromates, bicyclic monohydroxyaromates, polycyclic monohydroxyaromates, bicyclic dihydroxyaromates,
A biologic artificial bone includes an artificial fiber material formed from a synthetic polymer with mechanical properties similar to type I collagen. A biocompatible liquid substance is impregnated in the fiber material that hardens and stiffens the fiber material. A bone substitute is impregnated in the hardened and stiffened fiber material forming an artificial bone composite. Vascular channels are formed in the artificial bone composite to facilitate in-growth of vessels and bone forming cells. The construction and methods achieve an artificial composite structure that is similar to natural bone with comparable properties.
Critical size bone defects and non-union fractions are still challenging to treat. Cell-loaded bone substitutes have shown improved bone ingrowth and bone formation. However, a lack of methods for homogenously colonizing scaffolds limits the maximum volume of bone grafts. Additionally, therapy robustness is impaired by heterogeneous cell populations after graft generation. Our aim was to establish a technology for generating grafts with a size of 10.5 mm in diameter and 25 mm of height, and thus for grafts suited for treatment of critical size bone defects. Therefore, a novel tailor-made bioreactor system was developed, allowing standardized flow conditions in a porous poly(L-lactide-co-caprolactone) material. Scaffolds were seeded with primary human mesenchymal stem cells derived from four different donors. In contrast to static experimental conditions, homogenous cell distributions were accomplished under dynamic culture. Additionally, culture in the bioreactor system allowed the induction of ...
There is disclosed a bone substitute material for medical use which satisfies all the requirements of (1) no histotoxicity, (2) osteoconductivity, (3) bone replacement capability, and (4) mechanical strength necessary for a bone reconstruction operation. The bone substitute material for medical use is predominantly composed of carbonate apatite and produced through the formation of carbonate apatite by contacting a block of calcium compound with a phosphate-containing solution, wherein the calcium compound block contains substantially no powders, and at least one of said calcium compound block and said phosphate solution contains a carbonate group, without any sintering. The block of calcium compound is preferably one prepared using an artificially synthesized calcium compound, most preferably a foamed calcium compound.
EthOss® is a totally synthetic bone graft material for guided bone regeneration, with a built-in membrane. By creating a calcium-rich environment which is ideal for bone growth, it is completely reabsorbed by a patients new bone as it grows.
GBR is a technique used for the regeneration of lost alveolar bone by the help of a grafting material and a barrier membrane. The defect is filled with a bone graft material (allogeneic-, xenogeneic- or alloplastic bone substitute materials) that serves as a scaffold for ingrowth of bone forming cells (osteoconduction) and blood vessels, and prevents collapse of the overlying membrane and soft tissue. The membrane acts as a barrier agains ingrowth of fast proliferating connective tissue cells, hence, giving the bone forming cells time and space for the osseous regeneration of the defect. Moreover, the membrane stabilizes the bone graft material and prevents the migration of particles.
Disclosed herein is a bone graft applicator assembly including a specially designed applicator, attached to a reservoir of bone graft material, such that the graft material can be moved into the delivery portion while the tip of the delivery portion is in place. The instantly disclosed device allows for the collapse of the delivery tip so that the tip can be placed between vertebras or alternatively within any bony structure or implant where the placement of bone graft material would be desirable, and then expanded to allow the bone graft material to be placed between the vertebras or the like while maintaining the geometry of the space into which the bone graft material is being placed.
The objective of this study was to investigate the surface modification of a natural bone substitute, Bio-Oss, coated with a synthetic polymer poly- D,L-lactide (PLA), in order to improve cell growth. Bio-Oss is a natural bone substitute made of the mineralized portion of bovine bone. The material is used mainly to fill bone defects in periodontal and maxillofacial surgery and permit reossification. Poly-α-hydroxyacids such as polylactic acid are receiving an increasing attention due to their ability to retain a great quantity of water, good biocompatibility, low interfacial tension, and minimal mechanical and frictional irritation. All of these features are appealing from the perspective of bioenvironmental mimicking. The human osteosarcoma cell line SAOS-2 was added to the top of scaffolds uncoated or coated with PLA and incubated at 37◦C in 5%CO2 for 15 days. PLA-coated scaffolds improved cell growth. Polymer degradation behaviour, extraction and measurement of the extracellular matrix ...

A method for fabricating a substitute component for bone, including the processes of: provision of a chemical spray including at least three of calcium chloride, hydrogen phosphate, hydrogen carbonate and water to form a combined solution; reaction and precipitation of the combined solution onto a substrate; allowing the precipitated particles to form a porous structure on the substrate; applying substantially isostatic pressure to the porous structure to form a compressed structure; and (optional) providing one or more through-holes in the compressed structure to promote osteoinduction.
Because of their bioactive properties and chemical similarity to the inorganic component of bone, calcium phosphate (CaP) materials are widely used for bone regeneration. Six commercially available CaP bone substitutes (Bio-Oss, Actifuse, Bi-Ostetic, MBCP, Vitoss and chronOs) as well as two tricalcium phosph
Most bone augmentation procedures involve the use of bone grafts. An excellent choice for a bone graft is your own bone (Autogenous bone ). Bone harvested from the patients own body is the gold standard and often times has the most predictable results. This is described as the best type of graft because such bone is live with active cellular elements that enhance bone growth, whereas other types of grafts are devoid of any active cellular material. synthetic bone graft material. This most likely will come from the adjacent area where implant is being placed or from your chin or ramus (the back part of your lower jaw). If your dentist cannot get enough bone from these areas, he or she may need to get bone from your hip or shin bone (tibia) instead. If you dont like the idea of having bone removed from your body to be placed in your jaw, other excellent option which is commonly used is synthetic bone graft material. These bone graft substitutes do not require a second surgical site as does the ...

Large infected bone defects (IBD) are very complicated to treat due to their high variability; they often require multiple procedures. Bone autografts are the gold standard for treatment but have several drawbacks, such as a need for a second surgery site, limited grafting material, and donor site morbidity. The objective of this research was to develop a moldable synthetic bone grafting material capable of releasing both antimicrobial and osteogenic drugs over a clinically relevant time course for the treatment of IBDs. Current treatment methods for large IBDs require two separate procedures to treat the bone defect and the infection. This research sought to combine these two procedures into one implantable composite bone graft substitute for the treatment IBDs. To begin, the degradation and mechanical properties of the calcium sulfate (CS) based composite material were evaluated for different compositions. Next, the controlled drug release profiles from the composite was achieved by using a shell and

An injectable bone mineral substitute material composition comprises an inorganic bone cement powder and a biologically compatible oil. The oil is an intermixture with the cement powder at a concentration of less than 10 wt % of the total weight of the composition in order to improve the rheology of the same. In a method of intermixing a powder of an implant material and a biologically compatible oil to a composition the oil is mixed with the powder of implant material at an elevated temperature.
Alongside the standard donor bone transfer, bone substitutes have established themselves, improved in quality and are being increasingly used in the surgical treatment of dysmorphic defects or defects following inflammation, tumor surgery or trauma, particularly in the last three decades. With the discussion about infectious disease transmission through tissue from bone banks, the use of donor bone has receded into the background. For this reason, suitable bone substitutes are being sought constantly. Implants of many different physical and chemical substance groups have been tested for their suitability as bone substitutes, but an ideal material has yet to be found. In animal experiment models and their clinical application in humans, various bone substitutes have been evaluated for the dynamic of the resulting bone formation using nuclear medical and radiological procedures and have been examined histologically for their value as functional bone substitutes. Compared to autogenous bone grafts, ...
Bone substitutes with enhanced biological activity are required for the replacement, repair and regeneration of skeletal tissue. This project aims to design a bioreactor for stem cell expansion on novel hydroxyapatite bone substitute materials designed for accelerated osseointegration of implants. Mathematical modelling has been used alongside physical and biological experiments to define the bioreactor environment. Initial experiments have been carried out using the osteogenic cell line MG63 seeded on both porous particles1,2 and disks of HA/TCP (hydroxyapatite/tricalcium phosphate). A purpose built flow chamber has been developed to study the shear stress required to detach cells from the HA/TCP surfaces. The CFD package FLUENT has been used to model the fluid dynamics in a fluidised bed bioreactor to investigate the distributor design, aspect ratio and flowrates of the system. Experiments have been carried out to investigate the fluidisation behaviour of the HA/TCP particles. Particles (2-4 ...

Purpose: To evaluate bone graft substitutes used in spine fusion surgery and determine the feasibility of studying the use of adult stem cells. Hypothesis: Using a well designed, randomized clinical trial to compare bone graft substitutes used in spine fusion surgery will help determine the best alternative to autologous bone graft. Design: Retrospective data on two bone graft substitutes will be evaluated. A protocol for studying stem cells in spine fusion will be drafted and the feasibility of implementing the trial will be analyzed. Results: It is difficult to design a randomized clinical trial to investigate a new surgical technique. The lack of standardization among spine surgeons makes it difficult to control for confounding variables.

phdthesis{f7ddb7f0-7bc5-427f-bc3b-742fdbc376e4, abstract = {Two formulations of biphasic injectable bone substitutes have been developed for the treatment of bone defects and stabilisation of fragility fractures.,br/,,br, ,br/,,br, The first formulation consists of a calcium sulphate matrix with embedded hydroxyapatite (HA) particles to provide osteoconductivity. The mechanical properties of this formulation were investigated by focussing on the factors affecting the compressive strength of the bone substitute. It was found that the relation between liquid and powder in the formulation was the key factor. By adding less liquid the strength increased. However, a lower amount of liquid makes the paste difficult to inject. Other factors that affected the strength were the amount of HA particles, their size and morphology. Increased strength was obtained for a lower content of HA particles with smaller size and smaller specific surface area. It was possible to obtain a strength comparable to ...
In principle, bone has a good self-healing capacity. However, for defects larger than a certain size (critical size defect) spontaneous healing of bone injury is not possible. Such defects can occur in many diseases such as osteoarthritis, bone cysts and tumors, or as a result of surgical procedures, for instance osteolyses associated with loosened endoprostheses or osteotomies. The gold standard for the treatment of bone defects is an autologous bone transplantation; however, disadvantages of the method include follow-up operations that are necessary in addition to the bone harvesting from the iliac crest, which are connected with significant comorbidity.1 Availability of natural bone is restricted and often not sufficient to heal large bone defects, while maintenance of an extensive bone bank is expensive and complicated by long-term tissue preservation issues. To overcome these difficulties, a number of synthetic and partial synthetic bone substitute materials have been developed. In ...

More than a decade has passed since the first injectable bone substitutes were introduced for use in orthopaedic trauma, and over recent years the number of commercial products has increased dramatically. Despite the fact that these bone substitutes have been on the market for many years, knowledge amongst potential users on how and when they might be useful is still fairly limited. Most injectable bone substitutes belong to one of two major groups: by far the largest group contains products based on various calcium phosphate (CP) mixtures, whilst the smaller group consists of calcium sulphate (CS) compounds. Following mixing, the CP or CS paste can be injected into - for instance - a fracture space for augmentation as an alternative to bone graft, or around a screw for augmentation if the bone is weak. Within minutes an in situ process makes the substitute hard; the mechanical strength in compression resembles that of cancellous bone, whereas the strength in bending and shear is lower. Over ...

There is an increasing need for synthetic bone substitute materials that decrease the need for allografts and autografts. In this study, composites of β-TCP and a biodegradable poly(L-lactide-co-ε-caprolactone) were manufactured using extrusion to form biodegradable composites with high β-TCP contents for osteoconductivity. The hydrolytic degradation of the composites containing 0, 10, 20, 35 and 50% of β-TCP was studied in vitro for 52 weeks. During the study, it was observed that β-TCP did not have an effect on the degradation rate of the polymer matrix. However, the crystallinity of the materials increased throughout the test series and changes in Tgs were also observed as the comonomer ratio of the polymer matrix changed as the degradation proceeded. The results show that the materials have desirable degradation properties and, thus, possess great potential as bioabsorbable and osteoconductive bone filling materials ...

Calcium sulphate (or calcium sulfate) is a common laboratory and industrial chemical. In the form of gamma-anhydrite (the nearly anhydrous form), it is used as a desiccant. It is also used as a coagulant in products like tofu. In the natural state, unrefined calcium sulphate is a translucent, crystalline white rock. When sold as a colour-indicating variant under the name Drierite, it appears blue or pink due to impregnation with Cobalt(II) chloride, which functions as a moisture indicator. The hemihydrate (CaSO4.~0.5H2O) is better known as plaster of Paris, while the dihydrate (CaSO4.2H2O) occurs naturally as gypsum. The anhydrous form occurs naturally as beta-anhydrite. Depending on the method of calcination of calcium sulphate dihydrate, specific hemihydrates are sometimes distinguished: alpha-hemihydrate and beta-hemihydrate. They appear to differ only in crystal shape. Alpha-hemihydrate crystals are more prismatic than beta-hemihydrate crystals and, when mixed with water, form a much ...

The developments in the field of regenerative medicine have contributed highly to clinical applications in maxillofacial surgery. A few years ago, a method has been developed by UMC Utrecht to fix small defects with artificial bone. This artificial bone is made of beta-tricalcium phosphate, a substance that is found in our bones, but which can also attract bone-forming stem cells. Small defects, such as a cleft palate in schisis can be perfectly filled. Patients from all over the country come here for this treatment, says prof. Rosenberg. Unfortunately, this method does not work for larger defects. That is because the artificial bone does not contain blood vessels, which is not a problem for a smaller piece of bone. However, larger bones can get infected with serious consequences. Within the strategic program Regenerative Medicine & Stem Cells we are working on the development of the growth of a network of blood vessels in which bone can grow. A promising development, which is not yet ...

A dental implant is essentially an artificial tooth root which is attached to the jaw bone. Eventually, a replacement tooth or bridge will be firmly fixed to this root, restoring complete function to the tooth. The key to a successful and long-lasting implant is the quality and quantity of jawbone to which the implant will be attached. If bone has been lost due to injury or periodontal disease, a sinus augmentation can raise the sinus floor to allow for new bone formation.. In the most common sinus augmentation technique, a tiny incision is made near the upper premolar or molar region to expose the jawbone. A small opening is cut into the bone and the membrane lining the sinus on the other side of the opening is gently pushed upward. The underlying space is filled with bone graft material and the incision is closed. The bone which is used for this procedure may be from your own body or from a cadaver. Sometimes the dentist might use synthetic materials which can also stimulate bone formation. ...
Bone has a unique capacity to regenerate itself completely. The major limiting factor is maintenance of space for bone formation. Bone graft materials have been used to facilitate bone formation within a space by occupying the space and allowing subsequent bone growth. Osseous grafting has been shown to be clinically successful in the management of human periodontal defects. Autogenous bone graft is considered as the gold standard for bone regeneration due to its unique properties of osteogenesis, osteoconduction, and osteoinduction. For more predictable regeneration of bone, autografts can be combined with platelet concentrates. This clinical case letter reports a case of Siebert\s class I ridge defect which was treated with the staged guided bone regeneration (GBR) approach using autogenous block graft and platelet-rich fibrin (PRF).. ...

A bone cement having a dry component including a large constituent and a small constituent. The small constituent fills a substantial volume of the interstitial spaces between the particles of the large constituent. Therefore, only a second or minor interstitial space is left remaining between the individual particles of the small constituent and the particles of the small constituent and the particles of the large constituent. Therefore, a reduced amount of a polymerizable component need be added to the dry component to form a bone cement. Such a bone cement formulation decreases the exothermic temperature of the bone cement and decreases the possibility of tissue necrosis in the implantation area.
This article reviews nanocomposites focusing on their impact and recent trends in the field of bone grafting. Although autogenous- and allogeneic-bone grafts have been used for a long time in bone therapies, there is still a donor shortage and infection risk. As an alternative, synthetic biomaterials have been developed and clinically used as bone grafts, but most of them differ substantially from natural bone either compositionally or structurally. It remains a great challenge to design an ideal bone graft that emulates natures own structure. Owing to the composition and structural similarity to natural bone, most of the current investigations involve the use of nanocomposites, particularly hydroxyapatite/collagen system, as promising bone grafts, but it is surprising that none of the reports review the rationale and design strategy of such nanocomposites in detail for the benefit of researchers. Accordingly, this article addresses the state-of-the-art of those nanocomposites and provides ...

Adipose tissue is an attractive source of mesenchymal stem cells (MSCs) because of its abundance and accessibility. We have previously defined a population of native MSCs termed perivascular stem cells (PSCs), purified from diverse human tissues, including adipose tissue. Human PSCs (hPSCs) are a bipartite cell population composed of pericytes (CD146+CD34-CD45-) and adventitial cells (CD146-CD34+CD45-), isolated by fluorescence-activated cell sorting and with properties identical to those of culture identified MSCs. Our previous studies showed that hPSCs exhibit improved bone formation compared with a sample-matched unpurified population (termed stromal vascular fraction); however, it is not known whether hPSCs would be efficacious in a spinal fusion model. To investigate, we evaluated the osteogenic potential of freshly sorted hPSCs without culture expansion and differentiation in a rat model of posterolateral lumbar spinal fusion. We compared increasing dosages of implanted hPSCs to assess for ...

0036]Any suitable osteogenic material or composition is contemplated for placement within the chamber 40. Illustrative osteogenic material include, for example, autograft, allograft, xenograft, demineralized bone, synthetic and natural bone graft substitutes, such as bio-ceramics and polymers, and osteo-inductive factors. Where bony material is placed within the chamber 40, the material can be pre-packed before the device is implanted, or can be inserted through the apertures after the device is in position. A separate carrier to hold the materials within the chambers of the device can also be used. These carriers can include collagen-based carriers, bio-ceramic materials, hydroxyapatite and calcium phosphate compositions. The carrier material can be provided in the form of a sponge, a block, folded sheet, putty, paste, graft material or other suitable form. Moreover, the osteogenic compositions contained within the vertebral replacement device can comprise an effective amount of a bone ...

CA flow screeds must be abraded, vacuumed and, as with all calcium sulphate substrates, primed with ASO-Unigrund. Heated screeds must be commissioned to recognised technical regulations before installing finishes. The readiness to receive floor finishes is determined by a CM device (carbide hygrometer). (CA screeds without underfloor heating: 0.5 CM%, CA screeds with underfloor heating: 0.3 CM%).. NOTE: Ensure that there is no rising damp coming from the substrate. This type of installation does not comply with the generally recognised rules of technology and must be contractually agreed separately.. ...

Bone grafting, normally a minor surgical procedure done in the dental office, is used to build up new bone in the area of your jaw that used to hold teeth. A small incision is made in your gum to expose the bone beneath it, and then grafting material is added. Most often, the grafting material is processed bone that serves as a scaffold, around which your body will actually deposit new bone cells. The grafting material will eventually be absorbed by your body and replaced by your own new bone.. The grafting material needed can come from a variety of sources. Sometimes it comes from your own body. Very often, however, it is bone from an animal or human donor that is processed by a laboratory to make it sterile and safe. Grafting material can even be synthetic. It comes in a variety of forms: powder, granules, putty or even a gel that can be injected through a syringe.. ...
