TY - JOUR. T1 - Red blood cell storage lesion. AU - Kor, Daryl J. AU - Van Buskirk, Camille M.. AU - Gajic, Ognjen. PY - 2009. Y1 - 2009. N2 - The past two decades have witnessed increased scrutiny regarding efficacy and risk of the once unquestioned therapy of red blood cell RBC transfusion. Simultaneously, a variety of changes have been identified within the RBC and storage media during RBC preservation that are correlated with reduced tissue oxygenation and transfusion-associated adverse effects. These alterations are collectively termed the storage lesion and include extensive biochemical, biomechanical, and immunologic changes involving cells of diverse origin. Time-dependent falls is 2,3-diphosphoglycerate, intracellular RBC adenosine triphosphate, and nitric oxide have been shown to impact RBC deformability and delivery of oxygen to the end-organ. The accumulation of biologic response modifiers such as soluble CD40 ligand sCD4oL, lyso-phosphatidylcholine lyso-PC, and Regulated on ...

N2 - INTRODUCTION: Cardiac surgery using cardiopulmonary bypass in newborns, infants and small children often requires intraoperative red blood cell transfusions to prime the circuit and oxygenator and to replace blood lost during surgery. The purpose of this study was to investigate the influence of red blood cell storage time prior to transfusion on postoperative morbidity in pediatric cardiac operations. METHODS: One hundred ninety-two consecutive children aged five years or less who underwent cardiac operations using cardiopulmonary bypass and who received red blood cells for priming the cardiopulmonary bypass circuit comprised ...
A large multicenter randomized controlled trial of approximately 1700 critically ill patients status post complex cardiac surgery with sternotomy, called the Red Cell Storage Duration Study (RECESS), is currently ongoing. In the RECESS trial, study groups are randomized to either RBCs of less than or equal to 10 days storage time or to greater than or equal to 21 days. The primary outcome of RECESS is a change in multiple organ dysfunction score. Secondary outcomes in RECESS include all cause 28-day mortality, mechanical ventilation free days, and other clinical outcomes. The RECESS study presents a unique opportunity to investigate mechanisms associated with RBC storage duration in the context of clinical outcomes for well-characterized surgical study groups. The ancillary study described here is called the Mechanism and Repository Study (MARS) for RECESS.. The MARS study will analyze the most commonly reported and hypothesized mechanisms considered to be associated with the RBC storage lesion ...
Background and objectives: Platelet storage lesions (PSL) are the sum of deleterious changes that platelets undergo during storage, affecting platelet concentrate viability for transfusion. In veterinary medicine, transfusion medicine has gained popularity and recent advances have been made. There is a continuous effort to prolong the shelf life and improve the quality of platelet concentrates. Removal of leukocytes has been proven to reduce febrile post-transfusion reactions and decrease platelet premature activation and apoptosis in humans. The present study objectives were to determine if leukoreduced canine platelet concentrates have fewer and delayed onset of in vitro storage lesions compared with non-leukoreduced concentrates and to determine which variable best predicts in vitro platelet viability. Methods: Platelet concentrates from twelve dogs were stored between 22°C - 24°C under continuous agitation for 7 days. Each sample was equally divided into leukoreduced (LR), leukoreduced ...

This study is an ancillary (add-on) study to the clinical trial entitled Red Cell Storage Duration Study (RECESS), which has clinicaltrials.gov identifier NCT00991341. The RECESS study randomizes cardiac surgery patients to receive either red blood cell units stored for no more than 10 days, or red blood cell units stored for at least 21 days, if they need any red blood cell transfusions during their hospital stay for the surgery.. This ancillary study is entitled Impact of Blood Storage Duration on Physiologic Measures: RECESS Ancillary Physiologic Study (RECAP). The hypothesis of the RECAP study is that there will be differences between the two randomized treatment groups in how much the following measurements change, from shortly before a red blood cell transfusion to shortly after a red blood cell transfusion, and from before surgery to 24 hours after surgery:. ...

Microvesicles (MVs) as one of storage lesions may be derived from ...
BACKGROUND: Clinical trials are investigating the potential benefit resulting from a reduced maximum storage interval for red blood cells (RBCs). The key drivers that determine RBC age at the time of issue vary among individual hospitals. Although progressive reduction in the maximum storage period of RBCs would be expected to result in smaller hospital inventories and reduced blood availability, the magnitude of the effect is unknown. STUDY DESIGN AND METHODS: Data on current hospital blood inventories were collected from 11 hospitals and three blood centers in five nations. A general predictive model for the age of RBCs at the time of issue was developed based on considerations of demand for RBCs in the hospital. RESULTS: Age of RBCs at issue is sensitive to the following factors: ABO group, storage age at the time of receipt by the hospital, the restock interval, inventory reserve, mean demand, and variation in demand. CONCLUSIONS: A simple model, based on hospital demand, may serve as the basis for
The shelf-life of stored red blood cells (RBCs) is typically 42 and 35 days for human and canine RBCs, respectively. Given that blood is a precious and limited resource, both human and veterinary blood banks typically dispense the oldest RBC units first to reduce wastage. However, accumulating evidence suggests that transfusion of RBCs stored ≥ 14 days is associated with increased rates of morbidity and mortality in hospitalized human patients. In our study of healthy dogs receiving autologous RBC transfusions stored for 7 (fresh) and 28 (old) days, administration of old, but not fresh, RBC units was associated with a pro-inflammatory cytokine response, exemplified by monocyte chemoattractant protein (MCP)-1, and accompanied by increased neutrophil counts and decreased platelet counts.. We propose to evaluate markers of inflammation, hemolysis, and endothelial activation in a randomized clinical trial in which client-owned dogs with primary immune-mediated hemolytic anemia (IMHA) and ...

After 6 weeks of refrigerated storage, transfusion of autologous red cells to healthy human volunteers increased extravascular hemolysis, saturated serum transferrin, and produced circulating nontransferrin-bound iron. These outcomes, associated with increased risks of harm, provide evidence that the maximal allowable red cell storage duration should be reduced to the minimum sustainable by the blood supply, with 35 days as an attainable goal.. ...

As they sit in the fridge marinading in their own filthy excretions, human erythrocytes mutate into something grotesque, no longer resembling human blood and wholly incapable of carrying oxygen. It is unbelievable that anybody should want to infuse such material into their patients, and concerns have been raised about the safety of using particularly old cells as their near their expiry. In Australia, packed red blood cells can only be stored for a maximum of 42 days (which is slightly conservative - internationally, the maximum PRBC shelf life is 49 days). Surprisingly, international clinical trial data supports the safety of existing practice. CICM SAQs mention storage lesions often, but only Question 29.1 from the first paper of 2010 has actually asked anything directly about them.

Morphometric analysis of avian spermatozoa from sperm samples preserved in formalin is a frequently adopted procedure in basic science (e.g. evolutionary ecology) and applied disciplines (e.g. animal breeding). Many research questions such as individual-based longitudinal studies of sperm traits require comparisons of formalin-stored sperm samples collected across multiple sampling events, which may be separated by years. Such analyses presuppose that prolonged storage in formalin does not affect sperm morphology, an assumption often implicitly made in the analysis of avian sperm morphology. This assumption, however, has never been tested, although for many study designs a potential effect of sperm storage duration may well confound the focal analysis. Based on pairwise comparisons of 22 experimental ejaculates from three passerine bird species, we found no evidence that differential storage duration of more than 1 year had affected the total length of spermatozoa stored in a 5 % formaldehyde ...

Well, only four more weeks to go!!!!! Ive been counting in weeks rather than months of late as it sounded like less time (8 weeks as opposed to 2 months etc) but we have reached the point where 1 month sounds shorter than 4 weeks which to me means that we are really close now. Im so excited.. Almost too excited that Im worried that Baby Pregnantcitygirl will arrive and then all this excitement will slowly trickle away to leave a crying baby that will be a lot of sleepless nights and hard work. I say that very lightheartedly of course because I know that I will love this baby more than life itself, hell, our 12 year old cat has weed on 3 duvets in the last 2 weeks (stress-dont ask) and we have had no dryer (broken and replaced today - hooray, we have dry towels after 30 minutes) and yet I have forgiven her over and over again without even trying! Perhaps Im softer than I thought.. These last 2 weeks have brought NCT classes (2 full days, 1 evening,1 breastfeeding). I really didnt want to go ...
The results of the baseline grain quality evaluations at the start of the investigation (day 0) are presented in Table 2. The effect of storage duration and botanical treatments on the percent grain moisture content (%MC) are presented in Table 3 and figures 1a & b, respectively. There were significant (P=0.05) storage duration, treatment and storage duration by treatment interaction effects on the percent grain moisture content. The treatment effects were significant for 60, 90 and 150 days of grain storage. Storage duration influenced the %MC of both untreated and treated grains. The grain MC increased significantly between 0 and 30 days for all treatments including the untreated control. Thereafter, there was a significant reduction (P=0.05) in %MC for all the treatments for the next 3 months (up to 120 days) of grain storage. Grains treated with the synthetic insecticide had consistently lower (P=0.05) %MC than those treated with botanicals or the untreated control during the entire five ...
Proven and potential clinical benefits of washing red blood cells before transfusion: current perspectives Amy E Schmidt, Majed A Refaai, Scott A Kirkley, Neil Blumberg Department of Pathology and Laboratory Medicine, University of Rochester Medical Center, Rochester, NY, USA Abstract: Red blood cells (RBCs) are washed for a variety of reasons such as to remove excess potassium, cytokines, and other allergen proteins from the supernatant and/or to mitigate the effects of the storage lesion. The storage lesion is a product of RBC aging and include leakage of potassium and chloride from the RBCs, depletion of 2,3-diphosphoglycerate and adenosine triphosphate, loss of phospholipids and cholesterol, exposure of phosphatidylserine, elaboration of lipid mediators, loss of glutathione, autoxidation of hemoglobin to methemoglobin contributing to decreased blood flow viscosity and adherence to endothelial cells, increased microparticle formation, and disruption of NO-mediated vasodilation. I am in the process of setting up a cell cycle analysis on K562 cells transfected with my gene and GFP (under a separate promoter) using PI. Im using the information found in the references below in order to do this experiment without losing the GFP during the permeabilization process. I will do the experiment over a time course of 5 days (removing a sample of cells each day) and would like to do the analysis at the end of the 5th day. My question is the proper way to store the cells. I am going to use 1% PFA to fix the cells followed by 70% EtOH permeabilization. Ive read that you can store the cells but they do not provide many details ...

Following submissions by Spokes and several individuals, the Scottish government has accepted that its proposed bronze/ silver/ gold standards for sustainable buildings may be too weak on bike storage and need reconsidered. Thank you if you made comments to the consultation!. Last year the Scottish consulted on proposed award levels for sustainable buildings, including houses and flats.. Spokes was appalled to discover that it would be possible for a developer to achieve a gold award for a sustainable dwelling by providing a space which could accomodate either one bicycle or one pram!! If the developer was really stingy, they could still achieve a bronze or silver sustainable-building award whilst providing zero cycle storage.. Such standards are way below the standards already applied to all new buildings by councils such as Edinburgh. You would think that to achieve a sustainability award a developer should have to exceed the basic standards already set by councils, rather than undercutting ...
About 5,000 liver transplants are performed annually in the United States, and a key factor ensuring success is maintaining a healthy donor organ that can function effectively in the recipient. However, transplanted livers typically incur substantial damage when blood flow is restored to the organ after the transplant.
Cell preparation. Sensory neurons were used within 24 hr of dissociation from adult Sprague Dawley rats and were stored at room temperature in L15 media plus nerve growth factor (50 ng/ml). Only cells with Ca-channel current amplitudes of ,1 nA were used for analysis. We observed no differences in Ca-channel current, opioid responses, or mRNA expression between freshly dissociated cells and those studied 1 d later. For example, in a random selection of tooth-pulp afferents studied immediately after dissociation versus those studied 24 hr later, MOR mRNA was detectable in 51% of fresh (n = 33) and 57% of 24 hr (n = 120) cells; those that expressed MOR mRNA had mean concentrations of 13.8 ± 3.2 and 17.3 ± 2.6 pm in fresh and 24 hr cells, respectively.. All methods for surgeries, dissections, dissociations, and cell storage are as described previously (Eckert et al., 1997), except for small adjustments of incubation times. The two surgeries used were shallow dental drilling to deposit crystals of ...

Value of Pumped Storage in a Thermal System. Construction of pumped storage hydroelectric capacity has not progressed as rapidly as desired particularly in all thermal systems. The wide interest in real but intangible benefits may have diverted attention from the value that will justify the construction of pumped storage capacity namely, low load factor peaking capacity. The wide acceptance of heavy energy penalties applied to pumped storage has also worked against this factor. Careful evaluations of this penalty considering lifetime load factors of thermal equipment with and without pumped storage may show the penalty to be small and, in some cases, a credit.
Ask IT professionals what their number one priority is in the next 12 months and in coming years and theyre likely to tell you increased usage of virtualization. No big surprise really since, for some time now, virtualization has been one of the most effective tools for driving efficiencies and effectiveness (and of course, saving money) across IT.. Driving those efficiencies, it turns out, will come in large part through convergence and consolidation virtualization brings about, and unified storage (or multiprotocol or federated storage) may be the key to making it all a reality. Why? Because while separate specialist storage platforms have gone a long way to increasing IT effectiveness, theyve also taken their toll on resource efficiency - something unified storage can finally put a serious dent in.. Most IT pros know this already, of course. Which is why a recent survey revealed that 70% of organizations are [Read more…]. ...
