The isolates formed two AFLP clusters and belonged to the genus Vibrio, more precisely to the Splendidus clade. In this study, phylogenetic analyses based on sequences of the housekeeping genes rpoA, rpoD, pyrH, atpA and recA supported their inclusion in that clade forming two well differentiated groups with respect to the rest of the species within the clade, and confirmed they formed two groups, separated from the rest of the species of the clade. DNA-DNA hybridizations demonstrated that the isolates constitute two novel Vibrio species, which can be phenotypically differentiated from their closest relatives. Among the potential biotic factors affecting population dynamics, parasitism has received relatively little attention. The purpose of this study was to inventory marine bivalve parasites and to investigate relations between different parasite communities. Five intertidal stations along the French Atlantic shore were studied between Brittany and Arcachon Bay. Two bivalves (the edible cockle, Cerastoderma edule, and the Manila clam, Ruditapes philippinarum) were sampled and examined. The parasites included a bacterium (Vibrio tapetis), a protozoan (Perkinsus sp.), and digenetic trematode metacercariae (10 species). Perkinsus sp. prevalence and density in clams were higher than those measured in cockles (30 250 vs. 36 cells g−1 of gill wet weight, respectively) while digeneans were more common in cockles (46 metacercariae per cockle host vs. 1 per clam host). Chemically skinned anterior byssus retractor muscle (ABRM) preparations were prepared by treatment with the nonionic detergents saponin and Triton X-100. Both maximum peak tension and rate of contraction were found to be greater in saponin-treated ABRM than in ABRM treated with Triton X-100. Active tension was initiated at a concentration of free Ca2+ above 0.1 microM, and maximum tension development was found at a [Ca2+] = approximately 32 microM. During exposure of the muscle preparation to optimal Ca2+ concentration, a high and almost constant tension level was sustained. The force recovery was high after a quick release during this period indicating the presence of an active state rather than a catch state. State 3 respiration, maximally stimulated by 5 micromol l(−)(1) sulfide with a P/O ratio of 0.89 and a respiratory control ratio (RCR) of 1.40, remained refractory to sulfide at higher concentrations except in the presence of salicylhydroxamic acid (SHAM), an inhibitor of alternative oxidases. We have studied differential gene expression related to differences in growth rate in adult individuals of the commercial marine clam Ruditapes decussatus. Gene expression in the gills and the digestive gland was analyzed in 5 fast-growing and five slow-growing animals by means of an oligonucleotide microarray containing 14,003 probes. A total of 356 differentially expressed genes (DEG) were found. Our research involves the development of one such model. Gonadal tumors have been described in the softshell clam (Mya arenaria) in Maine and the hardshell clam (Mercenaria spp.) from Florida. Prevalence of these tumors is as high as 40% in some populations in eastern Maine and 60% in some areas along the Indian River in Florida. ABSTRACT: The blackening of tissues or mucus of benthic animals from sulphidic environments is a remarkable phenomenon whose ecological interpretation is disputed. In the Baltic clam Macoma balthica the mantle edge turned black after sulphide exposure owing to numerous precipitates in the extracellular matrix underneath the epidermal cells. In the apical parts of these cells, similar precipitates were found, albeit in lower abundance. Elemental analyses showed that copper (214.7 µg g-1 ww [wet weight]) and sulphur (1328.6 µg g-1 ww) were the main components, with iron (311.2 µg g-1 ww) and zinc (112.7 µg g-1 ww) in lower concentration. Apparently, these precipitates become phagocytosed by amoebocytes and concentrated in haemocytic granules. This is interpreted as a pathway of removal from the mantle edge. Previously, we reported that recA was probably lost in the early stage of RGE in Calyptogena clam symbionts [8]. The present study showed that some of the extant clam symbionts still have intact recA (Figure 2). We hypothesized that in the early phase of RGE of the clam symbionts before the loss of recA, large-sized deletions occurred due to RecA-dependent recombination [8]. This type of deletion requires repeated sequences larger than 200 bp, which have been depleted from the genomes of Rma and Vok [8, 19]. It is still not clear whether the genomes of the Calyptogena clam symbionts containing recA have large-sized (, 200 bp) repeated sequences. The presence of intact or of nearly intact recA and of mutY in clade II symbionts except for Rma suggests that the genomes of clade II symbionts are larger than those of clade I symbionts and that their RGE is in an earlier stage than in clade I symbionts. Perturbation of the reproductive cycle as well as vitellin-like protein synthesis have already been reported in Mya arenaria sampled in contaminated areas of the St. Lawrence maritime estuary (Québec, Canada). To assess the potential role of endocrine disruptors in modulating the reproductive cycle in clams, the role of sex steroids has to be clarified. We determined the levels of estradiol-17beta and testosterone during the reproductive cycle in female and male M. arenaria. Both steroids were measured by ELISA in the gonads and no differences in steroid profiles appeared between sexes. Estrogen levels varied between 150 and 400 pg g(-1) wet weight over gametogenesis and were near 10 times higher than testosterone levels. Results showed transient increases of both steroids at the onset of vitellogenesis in females and during the spawning stage in both sexes. Until recently, read lengths on the Solexa/Illumina system were too short to reliably assemble transcriptomes without a reference sequence, especially for non-model organisms. However, with read lengths up to 100 nucleotides available in the current version, an assembly without reference genome should be possible. For this study we created an EST data set for the common pond snail Radix balthica by Illumina sequencing of a normalized transcriptome. Performance of three different short read assemblers was compared with respect to: the number of contigs, their length, depth of coverage, their quality in various BLAST searches and the alignment to mitochondrial genes. A single sequencing run of a normalized RNA pool resulted in 16,923,850 paired end reads with median read length of 61 bases. The generation of EST information is an essential step in the genomic characterisation of species. In the context of the European Network Marine Genomics, a common goal was to significantly increase the amount of ESTs in commercial marine mollusk species and more specifically in the less studied but ecologically and commercially important groups, such as mussel and clam genera. Normalized cDNA libraries were constructed for four different relevant bivalves species (Crassostrea gigas, Mytilus edulis, Ruditapes decussatus and Bathymodiolus azoricus), using numerous tissues and physiological conditions.
