The isolates formed two AFLP clusters and belonged to the genus Vibrio, more precisely to the Splendidus clade. In this study, phylogenetic analyses based on sequences of the housekeeping genes rpoA, rpoD, pyrH, atpA and recA supported their inclusion in that clade forming two well differentiated groups with respect to the rest of the species within the clade, and confirmed they formed two groups, separated from the rest of the species of the clade. Among the potential biotic factors affecting population dynamics, parasitism has received relatively little attention. The purpose of this study was to inventory marine bivalve parasites and to investigate relations between different parasite communities. Five intertidal stations along the French Atlantic shore were studied between Brittany and Arcachon Bay. Two bivalves (the edible cockle, Cerastoderma edule, and the Manila clam, Ruditapes philippinarum) were sampled and examined. The parasites included a bacterium (Vibrio tapetis), a protozoan (Perkinsus sp.), and digenetic trematode metacercariae (10 species). Perkinsus sp. prevalence and density in clams were higher than those measured in cockles (30 250 vs. 36 cells g−1 of gill wet weight, respectively) while digeneans were more common in cockles (46 metacercariae per cockle host vs. 1 per clam host). Chemically skinned anterior byssus retractor muscle (ABRM) preparations were prepared by treatment with the nonionic detergents saponin and Triton X-100. Both maximum peak tension and rate of contraction were found to be greater in saponin-treated ABRM than in ABRM treated with Triton X-100. Active tension was initiated at a concentration of free Ca2+ above 0.1 microM, and maximum tension development was found at a [Ca2+] = approximately 32 microM. During exposure of the muscle preparation to optimal Ca2+ concentration, a high and almost constant tension level was sustained. The force recovery was high after a quick release during this period indicating the presence of an "active" state rather than a "catch" state. The ribbed mussel Geukensia demissa inhabits intertidal Spartina grass marshes characterized by sulfide-rich sediments. Sulfide poisons aerobic respiration, and G. demissa may cope in this seemingly inhospitable environment by oxidizing sulfide in gill mitochondria. Well-coupled mitochondria isolated from G. demissa gills were used to investigate sulfide oxidation and ATP synthesis. State 3 respiration, maximally stimulated by 5 micromol l(−)(1) sulfide with a P/O ratio of 0.89 and a respiratory control ratio (RCR) of 1.40, remained refractory to sulfide at higher concentrations except in the presence of salicylhydroxamic acid (SHAM), an inhibitor of alternative oxidases. Sulfide-stimulated ATP production was 3-5 times greater than that stimulated by malate and succinate, respectively, giving an ATP/sulfide ratio of 0.63. Growth rate is one of the most important traits from the point of view of individual fitness and commercial production in mollusks, but its molecular and physiological basis is poorly known. We have studied differential gene expression related to differences in growth rate in adult individuals of the commercial marine clam Ruditapes decussatus. Gene expression in the gills and the digestive gland was analyzed in 5 fast-growing and five slow-growing animals by means of an oligonucleotide microarray containing 14,003 probes. A total of 356 differentially expressed genes (DEG) were found. While both genetic and environmental factors have been implicated in their etiology, the extent of the contribution of environmental factors to human diseases remains controversial. To better address the role of environmental exposures in cancer etiology, there has been an increasing focus on the development of nontraditional, environmentally relevant models. Our research involves the development of one such model. Gonadal tumors have been described in the softshell clam (Mya arenaria) in Maine and the hardshell clam (Mercenaria spp.) from Florida. Prevalence of these tumors is as high as 40% in some populations in eastern Maine and 60% in some areas along the Indian River in Florida. The average tumor prevalence in Maine and Florida is approximately 20 and 11%, respectively. ABSTRACT: The blackening of tissues or mucus of benthic animals from sulphidic environments is a remarkable phenomenon whose ecological interpretation is disputed. In the Baltic clam Macoma balthica the mantle edge turned black after sulphide exposure owing to numerous precipitates in the extracellular matrix underneath the epidermal cells. In the apical parts of these cells, similar precipitates were found, albeit in lower abundance. Elemental analyses showed that copper (214.7 µg g-1 ww [wet weight]) and sulphur (1328.6 µg g-1 ww) were the main components, with iron (311.2 µg g-1 ww) and zinc (112.7 µg g-1 ww) in lower concentration. Perturbation of the reproductive cycle as well as vitellin-like protein synthesis have already been reported in Mya arenaria sampled in contaminated areas of the St. Lawrence maritime estuary (Québec, Canada). To assess the potential role of endocrine disruptors in modulating the reproductive cycle in clams, the role of sex steroids has to be clarified. We determined the levels of estradiol-17beta and testosterone during the reproductive cycle in female and male M. arenaria. Both steroids were measured by ELISA in the gonads and no differences in steroid profiles appeared between sexes. Estrogen levels varied between 150 and 400 pg g(-1) wet weight over gametogenesis and were near 10 times higher than testosterone levels. Results showed transient increases of both steroids at the onset of vitellogenesis in females and during the spawning stage in both sexes. The shipworm Lyrodus pedicellatus is a wood-boring bivalve with an unusual vermiform body. Although its larvae are brooded, they retain the general appearance of a typical bivalve veliger-type larva. Here, we describe myogenesis of L. pedicellatus revealed by filamentous actin labelling and discuss the data in a comparative framework in order to test for homologous structures that might be part of the bivalve (larval) muscular ground pattern. Five major muscle systems were identified: a velum retractor, foot retractor, larval retractor, a distinct mantle musculature and an adductor system. For a short period of larval life, an additional ventral larval retractor is present. Early in development, a velum muscle ring and an oral velum musculature emerge. In late stages the lateral and dorsal mantle musculature, paired finger-shaped muscles, an accessory adductor and a pedal plexus are formed.
