Hexachlorophene - Wikipedia
Hexachlorophene, also known as Nabac, is an organochlorine compound that was once widely used as a disinfectant. The compound occurs as a white odorless solid, although commercial samples can be off-white and possess a slightly phenolic odor. It is insoluble in water but dissolves in acetone, ethanol, diethyl ether, and chloroform. In medicine, hexachlorophene is a useful as a topical anti-infective, anti-bacterial agent, often used in soaps and toothpaste. It is also used in agriculture as a soil fungicide, plant bactericide, and acaricide. In 1972, the U.S. Food and Drug Administration (FDA) halted the production and distribution of products containing more than 1% of hexachlorophene. After that point, most products that contain hexachlorophene were available only by prescription from a doctor. (Hexachlorophene was used in shampoos, soaps and other personal care products until about a decade ago.) Rats were fed hexachlorophene orally for 3 weeks, then given Piracetam or one of 5 other drugs by injection for 6 days. Hexachlorophene seriously disrupted the rats ability to navigate a horizontal ladder without frequently falling off the rungs. Piracetam reduced the fall rate 75% compared to saline-injected controls on the first day of treatment. None of the other drugs came close to that improvement. (24). Urease is a nickel-dependent amidohydrolase that catalyses the decomposition of urea into carbamate and ammonia, a reaction that constitutes an important source of nitrogen for bacteria, fungi and plants. It is recognized as a potential antimicrobial target with an impact on medicine, agriculture, and the environment. The list of possible urease inhibitors is continuously increasing, with a special interest in those that interact with and block the flexible active site flap. We show that disulfiram inhibits urease in Citrullus vulgaris (CVU), following a non-competitive mechanism, and may be one of this kind of inhibitors. Disulfiram is a well-known thiol reagent that has been approved by the FDA for treatment of chronic alcoholism. We also found that other thiol reactive compounds (l-captopril and Bithionol) and quercetin inhibits CVU. These inhibitors protect the enzyme against its full inactivation by the thiol-specific reagent Aldrithiol (2,2′-dipyridyl disulphide, DPS), suggesting that the three

Abstract
Between 1974 and 1985, 10 patients were studied who presented with either gastrointestinal complaints or unexplained peripheral blood eosinophilia, and had eggs typical for N. salmincola recovered from their stools. Clinical findings in 8 of the 10 included increased frequency of bowel movements or diarrhea (6), peripheral blood eosinophilia (6), abdominal discomfort (5), nausea and vomiting (3), weight loss (2), and fatigue (2). Two were asymptomatic. Eight recalled eating fish prior to the onset of symptoms. Anthelminthic treatment consisting of three 2-g doses of niclosamide (2 patients) or two 50 mg/kg doses of bithionol (1 patient) proved effective. In the remaining individuals symptoms resolved slowly over several months.

The idea behind this study was to see if the normal copy can be enhanced to make up for the non-functional copy, said Ali Djalilian, UIC professor of ophthalmology and corresponding author of the paper. Researchers used a mouse model of the human disease aniridia, an eye disorder that affects the iris and causes substantial visual impairment and can also be associated with systemic abnormalities. In aniridia, one copy of the gene PAX6 is normal and the other copy is non-functional. Here we identify hundreds of new drug-disease associations for over 900 FDA-approved drugs by quantifying the network proximity of disease genes and drug targets in the human (protein-protein) interactome. We select four network-predicted associations to test their causal relationship using large healthcare databases with over 220 million patients and state-of-the-art pharmacoepidemiologic analyses. Using propensity score matching, two of four network-based predictions are validated in patient-level data: carbamazepine is associated with an increased risk of coronary artery disease (CAD) [hazard ratio (HR) 1.56, 95% confidence interval (CI) 1.12-2.18], and hydroxychloroquine is associated with a decreased risk of CAD (HR 0.76, 95% CI 0.59-0.97). Parasite of the Day: Anoplocephala manubriata
The tapeworms described in this study were retrieved from a young male elephant that died at the Udawalawe Elephant Transit Home in Sri Lanka. Anoplocephala manubriata has very muscular suckers on its scolex which allows it to keep a firm grip on the host intestinal wall. But this is not so great for the elephant - the suction from the tapeworms suckers essentially end up leaving hickeys on the elephants intestinal mucosa, which is not a particularly healthy place for an elephant to get love bites, especially if they have been left there by a bunch of tapeworms. Indeed, the elephant that was necropsied in this study was found to have multiple lesions and ulcers on the gut lining as a result of these parasitic love bites. This tapeworm seems to be far more common among younger elephants than adults, possibly because older elephants have more developed immune systems, and have build up some kind of resistant towards these parasites ...
