The introduction of biological control agents to new environments requires host specificity tests to estimate potential non-target impacts of a prospective agent. Currently, the approach is conservative, and is based on physiological host ranges determined under captive rearing conditions, without consideration for ecological factors that may influence realized host range. We use historical data and current field data from introduced parasitoids that attack an endemic Lepidoptera species in Hawaii to validate a probabilistic risk assessment (PRA) procedure for non-target impacts. We use data on known host range and habitat use in the place of origin of the parasitoids to determine whether contemporary levels of non-target parasitism could have been predicted using PRA. Our results show that reasonable predictions of potential non-target impacts may be made if comprehensive data are available from places of origin of biological control agents, but scant data produce poor predictions. The work developed in this thesis aimed to increase understanding about the variability and stability in eleven biotypes of Pochonia chlamydosporia, a facultative parasitic fungus with potential as a biological control agent against root-knot (Meloidogyne spp.), false root-knot (Naccobus spp.) and cyst nematodes (Heterodera and Globodera, spp.). Differences in performance were assessed by measuring saprophytic and parasitic growth using in vitro bioassays. The objective of this research was to identify bacterial spot-causing pathogen and bacterial antagonists for management of the disease using biocontrol agents as environmentally friendly alternatives. Isolates of Bacillus spp. were obtained from the soil samples collected at different localities in Serbia. Antibacterial activity of natural antagonists toward the pathogen isolated from infected pepper leaves was examined using a modified well-diffusion assay and standard germination test. Our results confirmed the presence of Xanthomonas euvesicatoria as the causal agent of bacterial spot of pepper. Screening of 32 Bacillus spp. isolates for antibacterial activity showed that 8 isolates inhibit growth of examined X. euvesicatoria isolates. Four isolates identified as Bacillus subtilis exhibited the highest antibacterial activity by in vitro test (from 5 to 14 mm inhibition zone of bacterial growth). In 2008, a field survey was conducted in northern Argentina to collect natural enemies of Cestrum species (Solanaceae) for use as biological control agents in South Africa. The rust fungus Uromyces cestri Bertero ex Mont. (Pucciniales: Pucciniaceae) was found on Cestrum parqui L'Hér. and imported into quarantine facilities in South Africa. No damaging pathogens were found on Cestrum laevigatum Schltdl. Preliminary host-range studies showed that U. cestri was able to infect and cause disease on C. parqui and on Cestrum elegans (Brongn. ex Neumann) Schltdl. in South Africa, and could have potential for the biological control of these species. Author summary Fungal pathogens pose an emerging threat in crop production and thus human health. Trichoderma atroviride is considered a potential biocontrol agent against a broad spectrum of phytopathogens. Cell wall chitin was identified as promising target to combat fungal diseases. Here we uncovered the regulation of chitin and chitosan synthesis and their contribution to dynamic cell wall remodeling as protective components in self-defense reactions during the mycoparasitic attack of Trichoderma. The systematic evaluation of the newly identified chitin-modifying enzymes confirmed their concerted interplay and their essential contribution to a successful mycoparasitic invasion. These findings provide further valuable, more specific information on targeting critical factors in the fungal cell wall adaptation process for therapeutic purposes as well as improved biocontrol applications.
