Ornithine cyclodeaminase (OCD) is an NAD+-dependent deaminase that is found in bacterial species such as Pseudomonas putida. Research and innovation in recent years has led to a paradigm shift in the bioengineering community regarding biofilms. Rather than focusing on their negative impact in disease and materials fouling, we, and other groups, are re-conceptualizing biofilms as an engineering platform. Our group has developed a novel strategy for producing rationally designed biocatalytic surfaces based on Biofilm Integrated Nanofiber Display (BIND). BIND uses the E. coli curli system to create extracellular functional nanofiber networks. In this work, we used protein capture tags displayed on curli fibers to transform biofilms into biocatalytic surfaces. We wanted to address the issues that currently hinder the applicability of surface displayed enzyme catalysts - although genetically engineered and scalable, the enzymes displayed through genetic fusion to cell surface proteins are often improperly folded and have reduced activity. The immobilization of enzymes and other proteins into ordered thin materials has attracted considerable attention over the past few years. This research has demonstrated that biomolecules immobilized in different [Langmuir-Blodgett (LB)/Langmuir-Schaefer (LS)] matrixes retain their functional characteristics to a large extent. These new materials are of interest for applications as biosensors and biocatalysts. We review the growing field of oxidases immobilized onto ordered Langmiur-Blodgett and Langmuir-Schaefer films. Strategies for the preparation of solid supports and the essential properties of the resulting materials with respect to the envisaged applications are presented. Basic effects of the nature of the adsorption and various aspects of the application of these materials as biosensors, biocatalysts are discussed. Outlook of potential applications and further challenges are also provided.

We have identified a novel component of the mitochondrial ISC assembly system, Iba57p (previously termed Caf17p), in a genome-wide screen for S. cerevisiae mutants that carry a coupled lysine and glutamate auxotrophy, which is indicative of defects of aconitase and homoaconitase maturation. Iba57p was demonstrated to be crucial for de novo Fe/S cluster incorporation into these mitochondrial aconitase-type Fe/S proteins. In addition, Iba57p is required for the in vivo enzymatic functions of the mitochondrial radical-SAM Fe/S proteins biotin synthase and lipoic acid synthase. Iba57p interacts with Isa1p and Isa2p, a finding consistent with the virtually identical phenotypes of isa1Δ, isa2Δ and iba57Δ cells (29, 31, 48). No defects in the maturation of other Fe/S proteins were detected in cells depleted for Iba57p or Isa1p/Isa2p (Mühlenhoff et al., unpublished). The applicability and economic feasibility of biocatalysis for chiral amine synthesis is reviewed and the findings were compared to established chemical processes using relevant process parameters (TON, TOF and STY). This review clearly showcases the potential of biocatalysis for the synthesis of chiral amines and provides a valuable guide for synthetic chemists who want to benefit from these new opportunities. Next, biocatalysis is applied for the synthesis of an amino alcohol with two stereocentres: A novel route for the synthesis of all four stereoisomers of 4-amino-1-phenylpentane-2-ol is presented. Enzymes were applied to install both stereocentres successively, which allowed the selective synthesis with high yields and optical purities. The 4-α-helix bundle is a macromolecular structure, where four amphiphilic α-helical peptide strands form a hydrophobic core.
