A natural bromophenol found in seaweeds, 3-bromo-4,5-dihydroxybenzaldehyde (BDB), has been shown to possess antioxidant effects. This study aimed to investigate the mechanism by which BDB protects skin cells subjected to oxidative stress. The effect of BDB on the protein and mRNA levels of glutathione-related enzymes and the cell survival of human keratinocytes (HaCaT cells) was investigated. BDB treatment increased the protein and mRNA levels of glutathione synthesizing enzymes and enhanced the production of reduced glutathione in HaCaT cells. Furthermore, BDB activated NF-E2-related factor 2 (Nrf2) and promoted its localization into the nucleus by phosphorylating its up-stream signaling proteins, extracellular signal-regulated kinase and protein kinase B. Thus, BDB increased the production of reduced glutathione and established cellular protection against oxidative stress via an Nrf2-mediated pathway.

ABSTRACT: Interactions of vanillin with soy, casein, and whey proteins were studied in aqueous model systems using a thermodynamic approach. Vanillin-protein binding was examined at 12 and 4 °C using 3 proteins and 6 vanillin concentrations. The results were analyzed using a Klotz plot. Number of binding sites (n) and dissociation constants (Kd) were derived from the plots. The thermodynamic parameters (ΔG°, ΔH°, and ΔS°) for the bindings were calculated. Under identical experimental conditions, whey protein was found to have higher affinity to vanillin than the other 2 proteins. Bindings of vanillin with casein and whey protein were enthalpy driven, while the interaction of vanillin with soy protein was highly entropy driven. The results inferred that conformational changes of soy protein might be important in binding of vanillin. ...

In this study, a series of data-driven experiments were performed in order to better understand limitations associated with biosynthesis of vanillate, which is the immediate precursor to vanillin. Time-course experiments monitoring production of heterologous metabolites in the E. coli de novo vanillin pathway revealed a bottleneck in conversion of protocatechuate to vanillate. Perturbations in central metabolism intended to increase flux into the heterologous pathway increased average vanillate titers from 132 to 205 mg/L, but protocatechuate remained the dominant heterologous product on a molar basis.
